Small molecule inhibitors have dominated the pharmaceutical landscape for a long time as the primary therapeutic paradigm targeting pathogenic ***,their efficacy heavily relies on the amino acid composition and spatial constitution of proteins,rendering them susceptible to drug resistance and failing to target undruggable *** recent years,the advent of targeted protein degradation(TPD)technology has captured substantial attention from both industry and *** an event-driven mode,TPD offers a novel approach to eliminate pathogenic proteins by promoting their degrada-tion,thus circumventing the limitations associated with traditional small molecule ***-bic tag tethering degrader(HyTTD)technology represents one such TPD approach that is currently in the burgeoning *** employ endogenous protein degradation systems to induce the degrada-tion of target proteins through the proteasome pathway,which displays significant potential for medical *** this review,we provide a comprehensive overview of the development history and the reported mechanism of action of ***,we delve into the physiological roles,structure-activity re-lationships,and medical implications of HyTTDs targeting various disease-associated ***,we propose insights into the challenges that necessitate resolution for the successful development of HyTTDs,with the ultimate goal of initiating a new age of clinical treatment leveraging the immense po-tential of HyTTDs.

The corrosion rates of additive-manufactured Mg alloys are higher than their as-cast counterparts,possibly due to increased kinetics for the hydrogen evolution reaction on secondary phases,which may include oxide *** Kelvin Probe Force Microscopy demonstrated that MgO inclusions could act as cathodes for Mg corrosion,but their low conductivity likely precludes ***,the density of state calculations through density functional theory using hybrid HSE06 functional revealed overlapping electronic states at the Mg/MgO interface,which facilitates electron transfers and participates in redox *** determination of the hydrogen absorption energy at the Mg/MgO interface reveals it to be an excellent catalytic site,with HER being found to be a factor of 23x more efficient at the interface than on metallic *** results not only support the plausibility of the Mg/MgO interface being an effective cathode to the adjacent anodic Mg matrix during corrosion but also contribute to the understanding of the enhanced cathodic activities observed during the anodic dissolution of magnesium.

The development of efficient and robust anode materials for stable alkaline seawater electrolysis is severely limited by chlorine evolution reaction and chloride ***,the sulfur-doped cobalt-nickel bimetallic phosphides(CoNiPS)are specifically designed as a pre-catalyst for navigating a surface reconstruction to fabricate the anions(PO^(3-)_(4) and SO^(2-)_(4))-decorated Co(Ni)OOH catalyst(R-CoNiPS)with exceptional durability and high activity for stable alkaline seawater oxidation(ASO).Various experiment techniques together with theoretical simulations both demonstrate that the in situ-generated PO^(3-)_(4) and SO^(2-)_(4) anions on catalyst surface can improve the oxygen evolution reaction(OER)activity,regulating and stabilizing the catalytic active species Co(Ni)OOH,as well as make a critical role in inhibiting the adsorp-tion of chloride ions and extending the service life of ***,this R-CoNiPS electrode exhi-bits superb OER activity toward AsO and stands out among the non-precious ASO electrocatalysts reported recently,requiring low overpotentials of 420 and 440 mV to attain large current densities of 500 and 1000 mA cm^(-2) in an alkaline natural seawater electrolyte,***,the catalyst displays a negligible chloride corrosion at room temperature during ASO operation(>200 h)at 500 mA cm^(-2).This work opens up a new viewpoint for designing high-activity and durable electrocata-lystsforseawaterelectrolysis.

The research purpose of this paper is to explore the influence of the baffle plate on the airflow in the rear cavity of the centrifugal impeller and optimize the performance of the secondary air system’s air bleed *** this paper,a comprehensive experimental study was carried out on the flow characteristics in the impeller rear cavity with baffle *** windage torque,flow structure and pressure drop between inlet and outlet were measured *** experiment was carried out with the condition that the range of rotational Reynolds number was from 8.33×10^(5)to 22.2×10^(5)and the range of mass flow rate coefficient was from 0.92×10^(4)to 2.92×10^(4).The results show that the static cavity and the narrow stator-rotor cavity formed by the baffle plate effectively suppress the overall swirl coefficient in the ***,the static pressure and total pressure drop in the rotor-stator cavity were *** influence of the baffle plate on the windage torque of the rotary disk is related to the turbulence *** large turbulence parameters,the windage torque would be reduced with baffle plate,while under small turbulence parameters,the baffle plate would increase with baffle *** general,the baffle plate can improve the flow capacity and optimize the bleed air performance with proper structure and operation conditions.

Oral squamous cell carcinoma(OSCC)is known as one of the most malignant tumors with high recurrence and fatality *** poor tumor-targeting ability of traditional chemotherapeutic drugs has been a grand challenge for anti-OSCC *** that,a large quantity of tumor associated macrophages in OSCC tissues further diminish the anti-tumor effects of these ***,we produced a therapeutic nano drug delivery system(FA-PEG-PLA-JQ1)through encapsulating JQ1[a small-molecule inhibitor of bromodomain containing protein 4(BRD4)]into the folic acid(FA)-modified nanoparticle(PEG-PLA),which could prolong the half-life of JQ1 and target the tumor *** then,JQ1 released from this nanoparticle could prevent OSCC growth inducing tumor cell apoptosis,inhibiting tumor angiogenesis and the polarization of M2 type *** conclusion,our date demonstrated the therapeutic benefits of FA-PEG-PLA-JQ1 against OSCC in vivo or in vitro,which could be a novel treatment strategy for OSCC in coming days.

Many empirical and analytical methods have been proposed to predict fracturing pressure in cohesive *** of them take into account three to four specific influencing factors and rely on the assumption of a failure *** this study,a novel data-mining approach based on the XGBoost algorithm is investigated for predicting fracture initiation in cohesive *** has the advantage of handling multiple influencing factors simultaneously,without pre-determining a failure mode.A dataset of 416 samples consisting of 14 distinct features was herein collected from past studies,and used for developing a regressor and a classifier model for fracturing pressure prediction and failure mode classification *** results show that the intrinsic characteristics of the soil govern the failure mode while the fracturing pressure is more sensitive to the stress *** XGBoost-based model was also tested against conventional approaches,as well as a similar machine learning algorithm namely random forest ***,several large-scale triaxial fracturing tests and an in-situ case study were carried out to further verify the generalization ability and applicability of the proposed data mining approach,and the results indicate a superior performance of the XGBoost model.

Identifying malicious traffic under concept drift based on intraclass consistency enhanced variational autoencoder

Accurate identification of malicious traffic is crucial for implementing effective defense countermeasures and has led to extensive research efforts. However, the continuously evolving techniques employed by adversaries have introduced the issues of concept drift, which significantly affects the performance of existing methods. To tackle this challenge, some researchers have focused on improving the separability of malicious traffic representation and designing drift detectors to reduce the number of false ***, these methods often overlook the importance of enhancing the generalization and intraclass consistency in the representation. Additionally, the detectors are not sufficiently sensitive to the variations among different malicious traffic classes, which results in poor performance and limited robustness. In this paper, we propose intraclass consistency enhanced variational autoencoder with Class-Perception detector(ICE-CP) to identify malicious traffic under concept drift. It comprises two key modules during training:intraclass consistency enhanced(ICE) representation learning and Class-Perception(CP) detector construction. In the first module, we employ a variational autoencoder(VAE) in conjunction with Kullback-Leibler(KL)-divergence and cross-entropy loss to model the distribution of each input malicious traffic flow. This approach simultaneously enhances the generalization, interclass consistency, and intraclass differences in the learned representation. Consequently, we obtain a compact representation and a trained classifier for nondrifting malicious traffic. In the second module, we design the CP detector, which generates a centroid and threshold for each malicious traffic class separately based on the learned representation, depicting the boundaries between drifting and non-drifting malicious traffic. Stretchable electronics have found widespread applications in various fields such as wearable electronics,soft robots,and *** an important promising alternative of traditional rigid conductors,liquid metals have demonstrated immense potential to provide high conductivity and stretchability for the stretchable electronic ***,limited by their fluidity and high surface tension,challenges remain in achieving liquid metal patterns with low-cost,high-precision,large-scale,and complex ***,a fabrication technique was proposed based on laser-induced graphene(LIG)stamps to enable liquid metal self-selectively adhere to *** metal patterns could thus be achieved in different designed geometries and could be transferred onto stretchable *** liquid metal patterns exhibit exceptional electrical conductivity(3.24×10^(6)S/m even under 1000%strain),high stretchability(1000%strain,maximum of 2500%),small resistance changes under significant deformations(with a quality factor of 62.5 under 1000%strain),and high resolution(down to 50μm).Utilizing the patterned liquid metals,a stretchable integrated multifunctional optoelectronic system was demonstrated,encompassing a stretchable display matrix,a pressure sensor array,a wireless powering coil,and cardiovascular sensors,which further highlight the remarkable application potential of liquid metals in optoelectronic user-interaction and advanced physiological monitoring.
