The coronavirus disease 2019(COVID-19)has spread *** mixed liver impairment has been reported in COVID-19 patients,the association of liver injury caused by specific subtype especially chronic hepatitis B(CHB)with COVID-19 has not been *** this multi-center,retrospective,and observational cohort study,109 CHB and 327 non-CHB patients with COVID-19 were propensity score matched at an approximate ratio of 3:1 on the basis of age,sex,and *** characteristics,laboratory examinations,disease severity,and clinical outcomes were ***,univariable and multivariable logistic and Cox regression models were used to explore the risk factors for disease severity and mortality,respectively.A higher proportion of CHB patients(30 of 109(27.52%))developed into severe status than non-CHB patients(17 of 327(5.20%)).In addition to previously reported liver impairment markers,such as alanine aminotransferase,aspartate aminotransferase,alkaline phosphatase,and total bilirubin,we identified several novel risk factors including elevated lactate dehydrogenase(≥245 U/L,hazard ratio(HR)=8.639,95%confidence interval(CI)=2.528–29.523;P<0.001)and coagulation-related biomarker D-dimer(≥0.5μg/mL,HR=4.321,95%CI=1.443–12.939;P=0.009)and decreased albumin(<35 g/L,HR=0.131,95%CI=0.048–0.361;P<0.001)and albumin/globulin ratio(<1.5,HR=0.123,95%CI=0.017–0.918;P=0.041).In conclusion,COVID-19 patients with CHB were more likely to develop into severe illness and *** risk factors that we identified may be helpful for early clinical surveillance of critical progression.

Dear Editor, mTORCI, as a center regulatory hub of metabolism, senses the cellular energy status, nutrition and extracellular stimuli and regulates cell growth, differentiation and functions of immune cells (Powell et al., 2012). Lysosomal localization of key signal comp orients is critical for mTORCI activati on: mTORCI activation requires co-localization of activated Rheb and mTORCI to the lysosome membrane (Buerger et al., 2006). Signals in eluding growth factors, cellular stresses and energy levels act on the disruption the formation of tuberous sclerosis complex (TSC) complex, comprised of TSC1, TSC2 and TBC1D7, which leads to the translocation and activation of Rheb on the lysosome membrane (Dibble et al., 2012). In response to nutrient levels, specifically the availability of amino acids and glucose (Efeyan et al., 2013), mTORCI is recruited to the lysosomal surface by Rag GTPases that are heterodimers of RagA or RagC bound to RagB or RagD. Multiple protein complexes have been implicated in regulation of mTORCI upon nutrient sensing including Ragulator, GATOR1, GATOR2, KICSTOR and vacuolar ATPases (Wolfson et al., 2017). Vacuolar ATPases are large multiple-protein complexes that acidify the lysosome and may mediate additional functions independent of their proton pump activity (Nishi and Forgac, 2002).

Summary:The COVID-19 pandemic caused by SARS-CoV2 is characterized by a remarkable variation in clinical severity ranging from a mild illness to a fatal multi-organ *** the dysregulated human immune responses in the fatal subjects is critical for management of COVID-19 patients and the *** this study,we examined the immune cell compositions in the lung tissues and hilar lymph nodes using immunohistochemistry on 6 deceased COVID-19 patients and 4 focal organizing pneumonia(FOP)patients who underwent lung surgery and served as *** found a dominant presence of macrophages and a general deficiency of T cells and B cells in the lung tissues from deceased COVID-19 *** contrast to the FOP patients,Tfh cells and germinal center formation were largely absent in the draining hilar lymph nodes in the deceased COVID-19 *** was correlated with reduced IgM and IgG levels compared to convalescent COVID-19 *** summary,our data highlight a defect of germinal center structure in deceased COVID-19 patients leading to an impaired humoral *** the mechanisms of this deficiency will be one of the key points for the management of this epidemic.

　　The genus Gentiana (Gentianaceae) contains about 400 species, which distribute almost all over the world. Some species have been investigated on phytochemistry and they could be characterized by the presence of some secoiridoid glucosides (bitter principles), flavone-C-glucosides and xanthone aglycones and glucosides,[1] but nothing is known, up to now, about the chemical constitutents of Gentiana piasezkii Maxim. Members of the genus Gentiana have been used as traditional Chinese herbal medicines to treat hepatitis.[2] In order to find active compounds as lead molecule for medicine, we have studied on constituents of Gentiana piasezkii Maxim and found two new natural occurring,6'-O-(4-hydroxy-veratroyloxy)-arbutin (1) and luteolin 7-(3-hydroxy-4-methoxyl-cinnamoyl)-8-C-glucoside (2) from n-BuOH soluble fraction of an ethanolic extract of the whole plant. Their structures were elucidated by spectroscopic methods including 2D NMR techniques (1H-1H COSY, HMQC and HMBC). So we describe herein the structures of two new phenolic glucosides.……

Dissipation-free approach for realizing non-Hermitian dynamics in a superconducting circuit

Non-Hermitian dynamics exhibits a wealth of surprising and potentially useful phenomena. However, it is typically realized by coupling a system with thermal reservoirs, which makes the system suffer from thermal fluctuations associated with the dissipation. Here, we propose a dissipation-free approach to realize non-Hermitian dynamics using a superconducting circuit composed of two resonators and a superconducting qutrit. The non-Hermiticity arises in the dynamical matrix for the evolution of the two resonators in the Heisenberg picture, via a photon-number non-conserved dynamics. The energy spectrum of the non-Hermitian dynamical matrix can be retrieved by measuring the state of the two resonators, and a phase transition of the energy spectrum can be observed by varying the control parameters. In the realization of the non-Hermitian dynamics, the dissipation of the system and the post-selection are not required. Thus, the approach is implemented in a deterministic way, and the reservoir-induced noise is suppressed. Numerical simulations indicate that the energy spectrum of the non-Hermitian dynamical matrix obtained via the measurement of the resonators is in accordance with the theoretical prediction. Moreover, we demonstrate the parity discrimination for the state of two four-level qudits as an application, exhibiting the potential of the non-Hermitian dynamics in the field of quantum measurement. This work opens a new avenue for the realization of non-Hermitian dynamics and may have a significant implication in exploring the non-Hermitian phenomena.

The immunotherapeutic effect of dendritic cells vaccine modified with interleukin-18 gene and tumor cell lysate on mice with pancreatic carcinoma

AIM：To estimate the effect of a therapeutic vaccine against pancreatic carcinoma based on dendritic cell(DC)vaccine modified with tumor lysate and Interleukin-18 ***:The BALB/C mice model of pancreatic arcinoma was induced with DMBA,DCvaccine was construted through pulsed with tumor lysate and transfected by the recombinant adenoviral vector encoding *** immnotherapeutic effects of DC vaccine on mice with pancreatic carcinoma were assessed (divided intoD-IL18-Lysate group,DC-Lysate group,DC-IL18 group,DC group,PBSgroup).RESULTS:After vaccination of the DC vaccine,the concentration of IL-18and IFN-γwere 2161±439ng·L^-1and435±72ng·L^-1inDC-IL18-Lysate group and there was significant difference compared with other groups(P<0.01)After vaccination of the DCcvaccine,the transplanted tumors were observed on 30days inDC-Lysate *** ***3days in control group,but mice remained tumor-free for at least50days in DC-IL18-Lysate group and there was significant difference between DC-IL18-Lysate group and other groups(P<0.01).The median survival exceeds62days in DC-IL18-Lysate *** the medsian surival was 48.6days in DC-Lysate group,33days in DC-IL-18group,17days in *** survival period was obviously prolonged in DC-IL18-Lysate group than in other groups(P<0.05,P<0.01).The weight of pancreatic tumor was0.22±0.083ginDC-IL18-Lysate group,1.45±0.74ginDC-Lysate group,1.89±1.34gin DC-IL18group,3.0±1.6ginDCgroup,2.9±2.0ginPBSgroup and the weight of tumor obviously reduced in DC-IL-18-Lysate group than in other groups(P<0.05,P<0.01).CONCLUSION:DC vaccine modified with tumo lysate and Interleukin-18gene can induce a specific and effective immune response against pancreatic cancinoma,cell.
