cDNA cloning,sequence analysis,and recombinant expression of akitonin beta,a C-type lectin-like protein from Agkistrodon acutus

AIM: To clone the cDNA of a new member of snake venom C-type lectin-like proteins, to study its structure- function relationships and to achieve its recombinant production. METHODS: PCR primers were designed based on the homology and cDNA was amplified by RT-PCR using total RNA from snake venom gland as the template. The PCR products were cloned into the plasmid pGEM-T and sequenced. The deduced protein sequence was analyzed with some bioinformatic programs. A recombinant expression plasmid was constructed using pBAD- TOPO as vector and transformed into *** TOP10 competent cells. RESULTS: A novel cDNA sequence encod- ing akitonin β was found and accepted by GenBank (accession number AF3.7100). Akitonin β consists of a typical carbohydrate recognition domain (CRD) of C-type lectins, and it is homologous with other snake venom C-type lectin-like proteins. It was predicted to be a platelet antagonist. Upon induction with arabinose rAkitonin β express- ing in E coli was achieved at a high level (superior to 150 mg/L). The recombinant fusion protein exhibited inhibitory activities on rat platelet aggregation in vitro. CONCLUSION: A new member of snake venom C-type lectin-like proteins was discovered and characterized, and an efficient recombinant expression system was estab- lished for its production.

Efficient electrocatalysts are vital to large-current hydrogen production in commercial water splitting for green energy ***,a novel heterophase engineering strategy is described to produce polymorphic CoSe_(2)*** composition of the electrocatalysts consisting of both cubic CoSe_(2)(c-CoSe_(2))and orthorhombic CoSe_(2)(o-CoSe_(2))phases can be controlled *** results demonstrate that junction-induced spin-state modulation of Co atoms enhances the adsorption of intermediates and accelerates charge transfer resulting in superior large-current hydrogen evolution reaction(HER)***,the CoSe_(2)based heterophase catalyst with the optimal c-CoSe_(2)content requires an overpotential of merely 240 mV to achieve 1,000 mA·cm^(-2)as well as a Tafel slope of 50.4 mV·dec^(-1).Furthermore,the electrocatalyst can maintain a large current density of 1,500 mA·cm^(-2)for over 3.0 h without *** results reveal the advantages and potential of heterophase junction engineering pertaining to design and fabrication of low-cost transition metal catalysts for large-current water splitting.

Development and performance evaluation of high temperature resistant strong adsorption rigid blocking agent

As drilling wells continue to move into deep ultra-deep layers, the requirements for temperature resistance of drilling fluid treatments are getting higher and higher. Among them, blocking agent, as one of the key treatment agents, has also become a hot spot of research. In this study, a high temperature resistant strong adsorption rigid blocking agent(QW-1) was prepared using KH570 modified silica,acrylamide(AM) and allyltrimethylammonium chloride(TMAAC). QW-1 has good thermal stability,average particle size of 1.46 μm, water contact angle of 10.5., has a strong hydrophilicity, can be well dispersed in water. The experimental results showed that when 2 wt% QW-1 was added to recipe A(4 wt% bentonite slurry+0.5 wt% DSP-1(filtration loss depressant)), the API filtration loss decreased from 7.8to 6.4 m L. After aging at 240.C, the API loss of filtration was reduced from 21 to 14 m L, which has certain performance of high temperature loss of filtration. At the same time, it is effective in sealing 80-100mesh and 100-120 mesh sand beds as well as 3.and 5 μm ceramic sand discs. Under the same conditions,the blocking performance was superior to silica(5 μm) and calcium carbonate(2.6 μm). In addition, the mechanism of action of QW-1 was further investigated. The results show that QW-1 with amide and quaternary ammonium groups on the molecular chain can be adsorbed onto the surface of clay particles through hydrogen bonding and electrostatic interaction to form a dense blocking layer, thus preventing further intrusion of drilling fluid into the formation.

Changes in protein abundance and reversible protein phosphorylation(RPP)play important roles in regulating hypometabolism but have never been documented in overwintering frogs at high *** test the hypothesis that protein abundance and phosphorylation change in response to winter hibernation,we conducted a comprehensive and quantitative proteomic and phosphoproteomic analysis of the liver of the Xizang plateau frog,Nanorana parkeri,living on the Qinghai-Xizang *** total,5170 proteins and 5695 phosphorylation sites in 193. proteins were *** on proteomic analysis,674 differentially expressed proteins(43. up-regulated,23. down-regulated)were screened in hibernating *** versus summer *** enrichment analysis revealed that higher expressed proteins in winter were significantly enriched in immune-related signaling pathways,whereas lower expressed proteins were mainly involved in metabolic processes.A total of 4251 modified sites(4147 up-regulated,104 down-regulated)belonging to 163. phosphoproteins(1555 up-regulated,83.down-regulated)were significantly changed in the *** hibernation,RPP regulated a diverse array of proteins involved in multiple functions,including metabolic enzymatic activity,ion transport,protein turnover,signal transduction,and alternative *** changes contribute to enhancing protection,suppressing energy-consuming processes,and inducing metabolic ***,the activities of phosphofructokinase,glutamate dehydrogenase,and ATPase were all significantly lower in winter compared to *** conclusion,our results support the hypothesis and demonstrate the importance of RPP as a regulatory mechanism when animals transition into a hypometabolic state.

It has been revealed that poly I：C is a potent stimulator for NK cells, which can induce NK cell rapid activation and preferential accumulation into liver. However, the process mediating the influx of NK cells remains obscure. In this study, we found that poly I：C administration increased the portion and absolute number of NK cells in liver, but largely decreased those in spleen. There were no obvious changes of these lymphocytes in other immune organs. The results from splenic adoptive transfer and splenectomy showed that the recruited spleen NK cells contributed to the accumulation of NK cells in liver, and this process was regulated by the production of chemokines and the presence of T cells.
