Testis-specific Fankl gene in knockdown mice produces oligospermia via apoptosis

Fankl is exclusively expressed in the testis from the meiosis phase to the haploid phase of spermatogenesis. In this study, we examined the function of Fankl by establishing a Fankl-knockdown transgenic mouse model. The apoptotic statuses of the testes of the transgenic mice were tested using the terminal deoxynucleotidyl transferase dUTP nick end labeling （TUNEL） method. The FANK1 consensus DNA-binding sequence was identified using cyclic amplification of sequence target （CAST） analysis. Differentially expressed genes were examined using microarray analysis. A reduction in sperm number and an increase in apoptotic spermatocytes were observed in Fankl-knockdown mice, and the apoptotic cells were found to be primarily spermatogonia and spermatocytes. The CAST results demonstrated that the consensus DNA-binding sequence was AAAAAG, in which the percentage occurrence of each base at each position ranged from 55 to 86%. This sequence was present in the promoter regions of 10 differentially expressed genes that were examined using microarray analysis. In total, 17 genes were differentially expressed with changes in their expression levels greater than twofold. The abnormal expression of Fankl target genes that were regulated directly or indirectly by Fankl reduced the number of sperm in the knockdown mice. Coconut(Cocos nucifera L.)is a key tropical economic tree valued for its fruit flavor,particularly 2-acetyl-1-pyrroline(2AP),a vital aroma *** enhance high-aromatic coconut breeding efforts,it is essential to deeply understand the hereditary factors governing the production of *** this study,a genome-wide association analysis identifies 32 loci that exhibit significant associations with 2AP content based on single nucleotide polymorphism(SNP)variations from 168 aromatic coconut germplasm *** analysis then pinpoints 22 candidate genes near significant loci involved in 2AP *** encoded by these genes are involved in amino acid metabolism,glycolysis,and secondary *** these,Asparagine synthetase coding gene ASN1,Gamma-glutamylcysteine synthetase coding gene GSH1,and UbiA prenyltransferase coding gene UBIA are enriched in the linkage region constructed by significant locus Chr04_*** particular,the SNP mutation of CnASN1 leads to amino acid changes in the functional region of the coding protein,potentially resulting in differences in 2AP content among haplotype *** variations in related candidate genes,particularly the gene CnASN1,provides molecular markers closely associated with 2AP synthesis for coconut breeding and offers further insights into the metabolic mechanisms of 2AP.

A 12-metal Zn（Ⅱ）-Nd（Ⅲ） cluster 1（sizes:1.8 nm×2.0 nm×2.0 nm） was synthesized from a long-chain type Schiff base *** displays ratiometric fluorescence response to neopterin（Neo） with high selectivity and sensitivity,which can be expressed by the equation I545nm/I1060nm=A·[Neo]2+B·[Neo]+C.1 is used to quantitatively test Neo concentrations in fetal calf serum（FCS） and urine,and the recovery ranges are 98.57%-103.82% and 99.25%-103.50%,respectively,while the relative standard deviations（RSDs） are 7.89%-9.46% and 1.85%-4.16%,*** limits of detection of 1 to Neo in FCS and urine are 0.034 and 0.021 μmol/L,respectively.

This study introduces an innovative method for synthesizing mordenite(MOR)by employing a combination of water glass and fumed silica as the silica *** zeolite produced through this method exhibits a smaller grain size,a larger specific surface area,and a greater number of Brønsted acid sites compared to the conventionally synthesized mordenite using silica ***,the chemical environment of framework Al in MOR zeolites is influenced by different silica sources,leading to varied acid properties between the two MOR zeolites,which results in an enhanced activity of dimethyl ether carbonylation from 50%to over 83%.

Data-based neural controls for an unknown continuous-time multi-input system with integral reinforcement

Integral reinforcement learning (IRL) is an effective tool for solving optimal control problems of nonlinear systems, and it has been widely utilized in optimal controller design for solving discrete-time nonlinearity. However, solving the Hamilton-Jacobi-Bellman (HJB) equations for nonlinear systems requires precise and complicated dynamics. Moreover, the research and application of IRL in continuous-time (CT) systems must be further improved. To develop the IRL of a CT nonlinear system, a data-based adaptive neural dynamic programming (ANDP) method is proposed to investigate the optimal control problem of uncertain CT multi-input systems such that the knowledge of the dynamics in the HJB equation is unnecessary. First, the multi-input model is approximated using a neural network (NN), which can be utilized to design an integral reinforcement signal. Subsequently, two criterion networks and one action network are constructed based on the integral reinforcement signal. A nonzero-sum Nash equilibrium can be reached by learning the optimal strategies of the multi-input model. In this scheme, the NN weights are constantly updated using an adaptive algorithm. The weight convergence and the system stability are analyzed in detail. The optimal control problem of a multi-input nonlinear CT system is effectively solved using the ANDP scheme, and the results are verified by a simulation study.

High quantum yield yellow emission carbon dots for the construction of blue light blocking films

White light illumination is essential in daily life, however, the substantial amount of blue light it contains can damage human eyes. Therefore, it is important to block this high-energy blue light to protect visual health. In this study, yellow-emitting carbon dots(CDs) with a quantum yield exceeding 94% were synthesized using citric acid and urea. These CDs effectively absorb blue light. By incorporating them into polystyrene, multiple films termed CDs-based blue light blocking films(CBFs) were developed, each offering different levels of blue light absorption. These CBFs exhibited excellent transparency and efficient blue light filtering capabilities. This study highlights the potential of high quantum yield CDs, which specifically absorb blue light, as foundational materials for developing light-blocking solutions against highenergy short-wavelength light.
