TLR2 signaling subpathways regulate TLR9 signaling for the effective induction of IL-12 upon stimulation by heat-killed Brucella abortus

Brucella abortus is a Gram-negative intracellular bacterium that induces MyD88-dependent IL-12 production in dentritic cells （DCs） and a subsequent protective Thl immune response. Previous studies have shown that the Toll-like receptor 2 （TLR2） is required for tumor-necrosis factor （TNF） production, whereas TLR9 is responsible for IL-12 induction in DCs after exposure to heat-killed Brucella abortus（HKBA）. TLR2 is located on the cell surface and is required for optimal microorganism-induced phagocytosis by innate immune cells; thus, phagocytosis is an indispensable preliminary step for bacterial genomic DNA recognition by TLR9 in late-endosomal compartments. Here, we hypothesized that TLR2-triggered signals after HKBA stimulation might cross-regulate TLR9 signaling through the indirect modulation of the phagocytic function of DCs or the direct modulation of cytokine gene expression. Our results indicate that HKBA phagocytosis was TLR2-dependent and an essential step for IL-12p40 induction. In addition, HKBA exposure triggered the TLR2-mediated activation of both p38 and extracellular signal-regulated kinase 1/2 （ERKI/2）. Interestingly, although p38 was required for HKBA phagocytosis and phagosome maturation, ERK1/2 did not affect these processes but negatively regulated IL-12 production, Although p38 inhibitors tempered both TNF and IL.12 responses to HKBA, pre-treatment with an ERKI/2 inhibitor significantly increased IL-12p40 and abrogated TNF production in HKBA-stimulated DCs. Further experiments showed that the signaling events that mediated ERK1/2 activation after TLR2 triggering also required HKBA-induced Ras activation. Furthermore, Ras-guanine nucleotide-releasing protein 1 （RasGRP1） mediated the TLR2-induced ERKI/2 activation and inhibition of IL-12p40 production. Immunotherapy in combination with chemotherapy for Peutz-Jeghers syndrome with advanced cervical cancer:A case report

BACKGROUND Peutz-Jeghers syndrome(PJS)is a rare autosomal dominant disorder,and female patients may develop gynecologic *** prognosis for such patients is poor and the specific pathogenesis remains ***,there are currently no uniform treatment *** SUMMARY Herein,we introduce the case of a 45-year-old female who was diagnosed with PJS for 45 years and cervical cancer for 3 *** pathological examination showed metastases in the right external iliac lymph *** patient was initially treated with a combination of doxorubicin and carboplatin chemotherapy and pelvic magnetic resonance showed that the metastases had ***,we performed whole exome sequencing in this patient and identified the relevant causative *** addition to the chemotherapy regimen,sindilizumab was administered and the patient was followed *** 4 cycles of treatment,the metastases were substantially reduced and were not enlarged after six months of *** case report suggests that patients with PJS combined with cervical cancer may have a sustained response to immunecombination chemotherapy *** Clinicians should be aware of the importance of immunotherapy in patients with PJS combined with advanced cervical cancer.

Semi-Varying Coefficient Panel Data Model with Technical Indicators Predicts Stock Returns in Financial Market

Accurately predicting stock returns is a conundrum in financial *** this conundrum can bring huge economic benefits for investors and also attract the attention of all circles of *** this paper the authors combine semi-varying coefficient model with technical analysis and statistical learning,and propose semi-varying coefficient panel data model with individual effects to explore the dynamic relations between the stock returns from five companies:CVX,DFS,EMN,LYB,and MET and five technical indicators:CCI,EMV,MOM,ln ATR,ln RSI as well as closing price(ln CP),combine semi-parametric fixed effects estimator,semi-parametric random effects estimator with the testing procedure to distinguish fixed effects(FE) from random effects(RE),and finally apply the estimated dynamic relations and the testing set to predict stock returns in December 2020 for the five *** proposed method can accommodate the varying relationship and the interactive relationship between the different technical indicators,and further enhance the prediction accuracy to stock returns.

Enhancing adhesive bond strength of CFRP/titanium joints through NaOH anodising and resin pre-coating treatments with optimised anodising conditions

Adhesively Bonded Carbon Fibre Reinforced Plastic(CFRP)and titanium alloy have been extensively used as a hybrid structure in modern aircrafts due to their excellent combination of mechanical properties and chemical *** study utilised NaOH anodising method to create micro-rough titanium surfaces for enhancing adhesive bonding between titanium alloy and CFRP laminates.A special and simple technique named Resin Pre-Coating(RPC)was also employed to improve the surface wetting of anodised titanium and grinded CFRP *** influences of anodising temperature and duration on the surface morphology,wettability and adhesive bond strength were *** single lap shear test results showed that the bond strength of specimens anodised at 20℃for 15 min improved by 135.9%and 95.4%,respectively,in comparison with that of acid pickled and grinded specimens(without RPC treatment).Although increasing the anodising temperature and duration produced rougher titanium surfaces,the adhesively bonded joints were not strong enough due to relatively friable titanium oxide layers.

Effects of Additives on the Microstructure and Tribology Performance of Ta-12W Alloy Micro-Arc Oxidation Coating

Oxide ceramic coatings were fabricated on tantalum alloys by micro-arc oxidation (MAO) to improve their hardness and tribological properties. The MAO coatings were manufactured in a mixed silicatephosphate electrolyte containing NaF and/or EDTA (ethylene diamine tetraacetic acid). The surface morphology,cross-sectional view, chemical composition, hardness, and wear performance of the coatings were analysed. As revealed by the scanning electron microscopy, silica-rich nodules appear on the MAO coating obtained in the silicate-phosphate electrolyte, but the formation of nodules is inhibited with NaF and/or EDTA in the ***, they reduce the roughness and improve the compactness of the coatings, which are composed of Ta_(2)O_(5),(Ta, O), and TaO. A thick and hard coating is obtained in the NaF-containing electrolyte, and the tribology performance is effectively improved. With additives, the nodule structure is detached from the coating surface and dissolved in the electrolyte. By using NaF as an electrolyte additive, the abrasion performance of the MAO coating is enhanced by decreasing the nodule structure, increasing the size of micropores, and improving the coating hardness.

Interregional Value-Added Tax in the Era of E-Commerce: Tax Policy Design Based on Big Data from Online Retailing

The value-added tax(VAT)in China is levied and allocated based on the origin *** the background of the increasing substitution of online retail for traditional offline retail,this mechanism will exacerbate the disparity of regional tax revenue,and intensify tax competition among local ***,reconsidering the allocation mechanism of value-added tax in China can be an important policy decision,and it is influenced by various economic and social ***,we utilize large-scale retail transaction data from an e-commerce platform to measure regional disparities in retail and consumption among different regions and then reveals present tax policy results in revenue imbalance in different ***,we establish a model based on game theory to illustrate how the origin principle leads to fierce tax competition among ***,by establishing and solving tax allocation models between local governments and the central government,this study simulates and calculates the degree of revenue imbalance under different scenarios and attempts to propose policy *** results indicate that implementing the destination principle will reduce regional tax ***,adjusting the allocation ratio between the central government and local governments based on city levels is advantageous for further reducing regional tax revenue disparities.

Formation of Superthin Galaxies in IllustrisTNG

Superthin galaxies are observed to have stellar disks with extremely small minor-to-major axis *** this work,we investigate the formation of superthin galaxies in the TNG100 *** trace the merger history and investigate the evolution of galaxy properties of a selected sample of superthin galaxies and a control sample of galaxies that share the same joint probability distribution in the stellar-mass and color *** making comparisons between the two galaxy samples,we find that present-day superthin galaxies had similar morphologies as the control sample counterparts at higher redshifts,but have developed extended flat“superthin”morphologies since z~*** this latter evolution stage,superthin galaxies undergo an overwhelmingly higher frequency of prograde mergers(with orbit-spin angleθ_(orb)≤40°).Accordingly the spins of their dark matter halos have grown significantly and become noticeably higher than those of their normal disk *** further results in the buildup of their stellar disks at larger distances much beyond the regimes of normal disk *** also discuss the formation scenario of those superthin galaxies that live in larger dark matter halos as satellite galaxies therein.
