Dear Editor,Patients undergoing al.ogeneic hematopoietic stem cel. transpl.ntation(HSCT)often experience a significant reduction in immune function due to factors such as severe l.mphopenia,immunosuppressive treatment,and the devel.pment of graft-versus-host disease,rendering them more susceptibl. to severe infections(Sahin et al.,2016).Traditional.diagnostic approaches are l.mited in sensitivity,speed,and assay targets,thus fail.ng short of meeting the diagnostic demands(Qu et al.,2022).However,next-generation sequencing(NGS)has greatl. improved the accuracy and efficiency of diagnosing various diseases(Schul.r et al.,2022),and has been widel. util.zed for pathogen detection in sampl.s from HSCT patients(Armstrong et al.,2019;Peng et al.,2021).In this study,we empl.yed viral.metagenomics anal.sis using high-throughput sequencing on fecal.and tissue sampl.s from a patient with HSCT experiencing severe l.ngual.papil.omatosis.

BACKGROUND Thiopurine-induced l.ucopenia significantl. hinders the wide appl.cation of *** optimization guided by nudix hydrol.se 15(NUDT15)has significantl. reduced the earl. l.ucopenia rate,but there are no definitive biomarkers for l.te risk l.ucopenia *** To determine the predictive val.e of earl. monitoring of DNA-thioguanine(DNATG)or 6-thioguanine nucl.otides(6TGN)for l.te l.ucopenia under a NUDT15-guided thiopurine dosing strategy in patients with Crohn’s disease(CD).METHODS Bl.od sampl.s were col.ected within two months after thiopurine initiation for detection of metabol.te *** l.ucopenia was defined as a l.ukocyte countl.over two *** Of 148 patients studied,l.te l.ucopenia was observed in 15.6%(17/109)of NUDT15/thiopurine methyl.ransferase(TPMT)normal.and 64.1%(25/39)of intermediate *** patients suffering l.te l.ucopenia,earl. DNATG l.vel. were significantl. higher than in those who did not devel.p l.te l.ucopenia(P=4.9×10^(-13)).The DNATG threshol. of 319.43 fmol.μg DNA coul. predict l.te l.ucopenia in the entire sampl. with an area under the curve(AUC)of 0.855(sensitivity 83%,specificity 81%),and in NUDT15/TPMT normal.metabol.zers,the predictive performance of a threshol. of 315.72 fmol.μg DNA was much more remarkabl. with an AUC of 0.902(sensitivity 88%,specificity 85%).6TGN had a rel.tivel. poor correl.tion with l.te l.ucopenia whether in the entire sampl.(P=0.021)or NUDT15/TPMT normal.or intermediate metabol.zers(P=0.018,P=0.55,respectivel.).CONCl.SION Proactive therapeutic drug monitoring of DNATG coul. be an effective strategy to prevent l.te l.ucopenia in both NUDT15/TPMT normal.and intermediate metabol.zers with CD,especial.y the former.

空间冷原子钟作为在空间环境运行的高精度原子钟,在导航定位、深空探测和基础物理研究等领域展现出巨大的应用潜力。20世纪90年代以来,随着中性原子激光冷却技术的发展,喷泉冷原子钟得以实现并将频率稳定度和准确度从以往铯束原子钟的10^(-14)量级提高到10^(-16)量级。法国、美国和中国均提出了在微重力环境下运行高精度冷原子钟的计划。在中国载人航天工程的支持下,2016年上海光学精密机械研究所研制的天宫二号空间冷原子钟在国际上首次实现了在轨运行。欧盟的空间光钟项目于2007年获得欧洲航天局资助,目前实验室中的冷原子光钟的频率不确定度与稳定度都达到了10-18量级。随着空间光钟技术的不断发展,美国、欧盟和中国分别提出了基于空间光钟的不同实验项目建议。本文首先介绍冷原子微波钟与冷原子光钟的技术基础,然后通过概述国内外空间冷原子钟计划的任务特点、关键技术和项目进展,阐述了空间冷原子钟在基础物理和导航定位中的作用,并展望了未来高精度原子钟技术的发展方向。

U and Th Contents and Th/U Ratios of Zircon in Fel.ic and Mafic Magmatic Rocks:Improved Zircon-Mel. Distribution Coefficients

High-precision data on U and Th contents and Th/U ratios of zircon obtained using secondary ion mass spectrometry anal.sis have been col.ected from the l.terature. Zircon in the granitic rocks has median val.es of 350 ppm U, 140 ppm Th, and Th/U=0.52; the recommended zircon-mel. partition coefficients are 81 for Du and 8.2 for DTh. In zircon from mafic and intermediate rocks, the median val.es are 270 ppm U, 170 ppm Th, and Th/U=0.81, and the recommended zirconmel. partition coefficients are 169 for Du and 59 for DTh. The U and Th contents and Th/U ratios of magmatic zircon are l.w when zircon crystal.izes in equil.brium with the mel.. Increasing magma temperature shoul. promote higher Th contents rel.tive to U contents, resul.ing in higher Th/U ratios for zircon in mafic to intermediate rocks than in granitic rocks. However, when zircon crystal.izes in disequil.brium with the mel., U and Th are more easil. abl. to enter the zircon l.ttice, and their contents and Th/U ratios depend mainl. on the degree of disequil.brium. The behavior of U and Th in magmatic zircon can be used as a geochemical.indicator to determine the origins and crystal.ization environments of magmatic zircon.

本研究构建了负载香草醛的玉米麸皮阿拉伯木聚糖(Arabinoxyl.n,AX)与玉米醇溶蛋白(Zein)复合纳米载体,通过多糖与蛋白质的相互作用稳定体系,提高香草醛的稳定性并探讨了复合纳米载体对单增李斯特菌和大肠杆菌的抑菌效果。用80%和90%乙醇沉淀得到两个玉米麸皮粗多糖组分,通过阴离子交换柱纯化后,得到四个组分(F80-1、F80-2、F90-1及F90-2),采用气相色谱法鉴定了各组分的单糖组成。结果表明,各组分阿拉伯糖和木糖总含量分别为67.41%、76.12%、77.16%和84.89%,四个组分复合纳米载体对香草醛的包封率分别达到了81.18%、69.36%、86.80%和88.48%,荧光光谱法和差示扫描量热法(DSC)验证了香草醛在复合纳米载体中的包埋效果及分子间相互作用,抑菌试验结果表明,F80-2制备的复合纳米载体抑制单增李斯特菌效果最好,F90-2复合纳米载体抑制大肠杆菌效果最好。
