Vimentin as a potential target for diverse nervous system diseases

Vimentin is a major type Ⅲ intermediate filament protein that plays important roles in several basic cellular functions including cell migration, proliferation, and division. Although vimentin is a cytoplasmic protein, it also exists in the extracellular matrix and at the cell surface. Previous studies have shown that vimentin may exert multiple physiological effects in different nervous system injuries and diseases. For example, the studies of vimentin in spinal cord injury and stroke mainly focus on the formation of reactive astrocytes. Reduced glial scar, increased axonal regeneration, and improved motor function have been noted after spinal cord injury in vimentin and glial fibrillary acidic protein knockout(GFAPVIM) mice. However, attenuated glial scar formation in post-stroke in GFAP–/– VIM–/– mice resulted in abnormal neuronal network restoration and worse neurological recovery. These opposite results have been attributed to the multiple roles of glial scar in different temporal and spatial conditions. In addition, extracellular vimentin may be a neurotrophic factor that promotes axonal extension by interaction with the insulin-like growth factor 1 receptor. In the pathogenesis of bacterial meningitis, cell surface vimentin is a meningitis facilitator, acting as a receptor of multiple pathogenic bacteria, including E. coli K1, Listeria monocytogenes, and group B streptococcus. Compared with wild type mice, VIMmice are less susceptible to bacterial infection and exhibit a reduced inflammatory response, suggesting that vimentin is necessary to induce the pathogenesis of meningitis. Recently published literature showed that vimentin serves as a double-edged sword in the nervous system, regulating axonal regrowth, myelination, apoptosis, and neuroinflammation. Analysis on phase selection and microstructure evolution in directionally solidified Zn-Al-Mg-Ce alloy

In the process of hot-dip Zn-Al-Mg alloy coating,the plating solution dissipates heat in the direction perpendicular to the steel plate,which is considered to be a process of directional solidification.To understand the relationship between microstructure and cooling rate of Zn-Al-Mg alloys,both the phase constitution and microstructure characteristic length scales of Zn-9.5Al-3Mg-0.01Ce(wt.%)alloy were investigated by the directional solidification experiments at different growth velocities(V=40,80,160,250μm·s^(-1)).The experimental results show that the microstructure of directionally solidified Zn-9.5Al-3Mg-0.01Ce alloy is composed of primary Al dendrites and(Zn-Al-Mg2Zn11)ternary eutectics at the growth velocities ranging from 40 to 250μm·s^(-1).The primary Al dendrites are aligned regularly along the growth direction,accompanied with obvious secondary dendrites.The relationship between the microstructure length scale and the thermal parameters of solidification is obtained:λ1=374.66V-0.383,andλ2=167.5V-0.563(λ1is the primary dendrite arm spacing,andλ2 is the secondary dendrit arm spacing).In addition,through the interface response function(IRF)and the nucleation and constitutional undercooling(NCU),the phase selection of Zn-9.5Al-3Mg-0.01Ce is obtained:(Zn+Al+Mg2Zn11)ternary eutectics in the Zn-9.5Al-3Mg-0.01Ce alloy will be replaced by ternary eutectics(Zn+Al+MgZn2)when the growth rate is lower than 7.53μm·s^(-1).

Effects of cooling rate on microstructure and microhardness of directionally solidified Galvalume alloy

The influences of cooling rate on the phase constitution,microstructural length scale,and microhardness of directionally solidified Galvalume(Zn-55Al-1.6Si)alloy were investigated by directional solidification experiments at different withdrawal speeds(5,10,20,50,100,200,and 400μm·s^(-1)).The results show that the microstructure of directionally solidified Galvalume alloys is composed of primary Al dendrites,Si-rich phase and(Zn-Al-Si)ternary eutectics at the withdrawal speed ranging from 5 to 400μm·s^(-1).As the withdrawal speed increases,the segregation of Si element intensifies,resulting in an increase in the area fraction of the Si-rich phase.In addition,the primary Al dendrites show significant refinement with an increase in the withdrawal speed.The relationship between the primary dendrite arm spacing(λ_(1))and the thermal parameters of solidification is obtained:λ_(1)=127.3V^(-0.31).Moreover,as the withdrawal speed increases from 5 to 400μm·s^(-1),the microhardness of the alloy increases from 90 HV to 151 HV.This is a combined effect of grain refinement and second-phase strengthening.

Teneligliptin mitigates diabetic cardiomyopathy by inhibiting activation of the NLRP3 inflammasome

BACKGROUND Diabetic cardiomyopathy(DCM),which is a complication of diabetes,poses a great threat to public health.Recent studies have confirmed the role of NLRP3(NOD-like receptor protein 3)activation in DCM development through the inflammatory response.Teneligliptin is an oral hypoglycemic dipeptidyl peptidase-IV inhibitor used to treat diabetes.Teneligliptin has recently been reported to have anti-inflammatory and protective effects on myocardial cells.AIM To examine the therapeutic effects of teneligliptin on DCM in diabetic mice.METHODS Streptozotocin was administered to induce diabetes in mice,followed by treatment with 30 mg/kg teneligliptin.RESULTS Marked increases in cardiomyocyte area and cardiac hypertrophy indicator heart weight/tibia length reductions in fractional shortening,ejection fraction,and heart rate;increases in creatine kinase-MB(CK-MB),aspartate transaminase(AST),and lactate dehydrogenase(LDH)levels;and upregulated NADPH oxidase 4 were observed in diabetic mice,all of which were significantly reversed by teneligliptin.Moreover,NLRP3 inflammasome activation and increased release of interleukin-1βin diabetic mice were inhibited by teneligliptin.Primary mouse cardiomyocytes were treated with high glucose(30 mmol/L)with or without teneligliptin(2.5 or 5μM)for 24 h.NLRP3 inflammasome activation.Increases in CKMB,AST,and LDH levels in glucose-stimulated cardiomyocytes were markedly inhibited by teneligliptin,and AMP(p-adenosine 5‘-monophosphate)-p-AMPK(activated protein kinase)levels were increased.Furthermore,the beneficial effects of teneligliptin on hyperglycaemia-induced cardiomyocytes were abolished by the AMPK signaling inhibitor compound C.CONCLUSION Overall,teneligliptin mitigated DCM by mitigating activation of the NLRP3 inflammasome.

Experimental investigation on the cuttings formation process and its relationship with cutting force in single PDC cutter tests

The single polycrystalline diamond compact(PDC)cutter test is widely used to investigate the mecha-nism of rock-breaking.The generated cuttings and cutting force are important indexes reflecting the rock failure process.However,they were treated as two separate parameters in previous publications.In this study,through a series of rock block cutting tests,the relationship between them was investigated to obtain an in-depth understanding of the formation of cuttings.In addition,to validate the standpoints obtained in the aforementioned experiments,rock sheet cutting tests were conducted and the rock failure process was monitored by a high-speed camera frame by frame.The cutting force was recorded with the same sampling rate as the camera.By this design,every sampled point of cutting force can match a picture captured by the camera,which reflects the interaction between the rock and the cutter.The results indicate that the increase in cutting depth results in a transition of rock failure modes.At shallow cutting depth,ductile failure dominates and all the cuttings are produced by the compression of the cutter.The corresponding cutting force fluctuates slightly.However,beyond the critical depth,brittle failure occurs and chunk-like cuttings appear,which leads to a sharp decrease in cutting force.After that,the generation of new surface results in a significant decrease in actual cutting depth,a parameter proposed to reflect the interaction between the rock and the cutter.Consequently,ductile failure dominates again and a slight fluctuation of cutting force can be detected.As the cutter moves to the rock,the actual cutting depth gradually increases,which results in the subsequent generation of chunk-like cuttings.It is accompanied by an obvious cutting force drop.That is,ductile failure and brittle failure,one following another,present at large cutting depth.The transition of rock failure mode can be correlated with the variation of cutting force.Based on the results of this paper,the real-time monitoring of torque ma

Reaction characteristics and kinetics of gallium in chlorination roasting of copper tailings using calcium chloride

The mineral composition of copper tailings was examined, and the phase analysis of gallium was conducted for recovery of the rare scattered metal gallium from copper tailings. The reaction characteristics and kinetics of gallium in the chlorination roasting of copper tailings were investigated in a tube furnace apparatus under different conditions, including roasting temperature, quantity of chlorination agent,roasting time, and airflow. Calcium chloride was chosen for the chlorination reaction because it is highly effective for volatilization of metals. The results show that gallium mainly exists in limonites in the form of oxides, and increases in roasting temperature, quantity of chlorination agent, and roasting time are conducive to increasing the chloridizing volatilization rate of gallium. The tailings gradually melt when the roasting temperature exceeds 900℃, resulting in a decrease in the chloridizing volatilization rate of gallium. The chloridizing volatilization rate of gallium reaches 78.86%at roasting temperature of 900℃ for 40 min, airflow of 0.1 m^(3)·h^(-1), and calcium content of 33.33 wt%. Study of the reaction kinetics of gallium in chlorination roasting shows that the values of activation energy both for chemically controlled anddiffusion-controlled reactionsare44.64and11.93 kJ·mol^(-1), respectively. This indicates that the chemical reaction serves an important function in the chlorination volatilization of gallium. Taken together, the results reveal the usefulness of tailings as a source of rare metals and provide a basis for gallium recovery and environmentally friendly disposal of copper tailings.

Horizontal structure of convergent wind shear associated with sporadic E layers over East Asia

At present,the main detection instruments for observing sporadic E(Es)layers are ground-based radars,dense networks of ground-based global navigation satellite system(GNSS)receivers,and GNSS radio occultation,but they cannot capture the whole picture of the horizontal structure of Es layers.This study employs the Whole Atmosphere Community Climate Model with thermosphere and ionosphere eXtension model(WACCM-X 2.1)to derive the horizontal structure of the ion convergence region(HSICR)to explore the shapes of the large-scale Es layers over East Asia for the period from June 1 to August 31,2008.The simulation produced the various shapes of the HSICRs elongated in the northwest-southeast,northeast-southwest,or composed of individual small patches.The close connection between Es layer critical frequency(foEs)and vertical ion convergence indicates that the HSICR is a good candidate for revealing and explaining the horizontal structure of the large-scale Es layers.

AIM:To investigate health-related quality of life(HRQoL) and psychological outcomes in 256 adults who had undergone liver transplantation(LT).METHODS:A stratified random sampling method was used in this follow-up multicenter study to select a representative sample of recipients undergoing either living donor liver transplantation(LDLT) or deceased donor liver transplantation(DDLT).HRQoL was measured by using the Chinese version of Medical Outcome Study Short Form-36(SF-36),and psychological outcomes by using the beck anxiety inventory(BAI) and the self-rating depression scale(SDS).Clinical and demographic data were collected from the records of the Chinese Liver Transplant Registry and via questionnaires.RESULTS:A total of 256 patients were sampled,including 66(25.8%) receiving LDLT and 190(74.2%) undergoing DDLT;15(5.9%) recipients had anxiety and four(1.6%) developed severe depression after the operation.Compared with LDLT recipients,DDLT patients had higher scores in general health(60.33 ± 16.97 vs 66.86 ± 18.42,P = 0.012),role-physical(63.64 ± 42.55 vs 74.47 ± 36.46,P = 0.048),roleemotional(61.11 ± 44.37 vs 78.95 ± 34.31,P = 0.001),social functioning(78.60 ± 22.76 vs 88.16 ± 21.85,P = 0.003),vitality(70.30 ± 15.76 vs 75.95 ± 16.40,P = 0.016),mental health(65.88 ± 12.94 vs 71.85 ± 15.45,P = 0.005),physical component summary scale(PCS,60.07 ± 7.36 vs 62.58 ± 6.88,P = 0.013) and mental component summary scale(MCS,52.65 ± 7.66 vs 55.95 ± 10.14,P = 0.016).Recipients > 45 years old at the time of transplant scored higher in vitality(77.33 ± 15.64 vs 72.52 ± 16.66,P = 0.020),mental health(73.64 ± 15.06 vs 68.00 ± 14.65,P = 0.003) and MCS(56.61 ± 10.00 vs 54.05 ± 9.30,P = 0.037) than those aged ≤ 45 years.MCS was poorer in recipients with than in those without complications(52.92 ± 12.21 vs 56.06 ± 8.16,P = 0.017).Regarding MCS(55.10 ± 9.66 vs 50.0 ± 10.0,P < 0.05) and PCS(61.93 ± 7.08 vs 50.0 ± 10.0,P < 0.05),recipients scored better than the Sichuan general and had improved overall QoL compared to patients with chr
