Background:Nasopharyngeal carcinoma(NPC)exhibits a significant prevalence in the southern regions of China,and paclitaxel(PTX)is frequently employed as a medication for managing advanced NPC.However,drug resistance is typically accompanied by a poor prognosis.Exploring the synergistic potential of combining multiple chemotherapeutic agents may represent a promising avenue for optimizing treatment efficacy.Methods:This study investigated whether 3-Methyladenine(3-MA)could potentiated the effect of PTX and its potential molecular mechanism.Samples were divided into the following categories:Negative control(NC)with the solvent dimethyl sulfoxide(DMSO,0.5%v/v),PTX(400 nM),3-MA(4 mM),and PTX(400 nM)+3-MA(4 mM).The viability of NPC cells was assessed using both the cell counting kit-8(CCK-8)assay and the colony formation assay.Microscopic observation was performed to identify morphological cell changes.Flow cytometry was used to assess cell cycle status,mitochondrial membrane potential(MMP),and apoptotic cells.Western blotting was conducted to quantify the protein expression.Results:3-MA enhanced PTX-specific inhibition of NPC cell proliferation.PTX,either alone or in combination with 3-MA,caused cell cycle halt at the G2/M phase in the majority of NPC cells,and the combination treatment of PTX with 3-MA induced a higher rate of NPC cell death compared to PTX alone.Western blotting results revealed the combination of PTX with 3-MA heightened activation of cyclin-dependent kinase 1(CDK1),a key molecule in shifting cells from mitotic arrest to apoptosis,led to a reduction in Myeloid Cell Leukemia 1(MCL-1)expression and an increase in Poly(ADP-ribose)polymerase(PARP)cleavage.Conclusion:The concurrent administration of PTX with 3-MA effectively enhances PTX’s inhibitory impact on NPC and activates the apoptosis signal regulated by CDK1.

NFXL1 functions as a transcriptional activator required for thermotolerance at reproductive stage in Arabidopsis

Plants are highly susceptible to abiotic stresses,particularly heat stress during the reproductive stage.However,the specific molecular mechanisms underlying this sensitivity remain largely unknown.In the current study,we demonstrate that the Nuclear Transcription Factor,X-box Binding Protein 1-Like 1(NFXL1),directly regulates the expression of DEHYDRATION-RESPONSIVE ELEMENT-BINDING PROTEIN 2A(DREB2A),which is crucial for reproductive thermotolerance in Arabidopsis.NFXL1 is upregulated by heat stress,and its mutation leads to a reduction in silique length(seed number)under heat stress conditions.RNA-Seq analysis reveals that NFXL1 has a global impact on the expression of heat stress responsive genes,including DREB2A,Heat Shock Factor A3(HSFA3)and Heat Shock Protein 17.6(HSP17.6)in flower buds.Interestingly,NFXL1 is enriched in the promoter region of DREB2A,but not of either HSFA3 or HSP17.6.Further experiments using electrophoretic mobility shift assay have confirmed that NFXL1 directly binds to the DNA fragment derived from the DREB2A promoter.Moreover,effector-reporter assays have shown that NFXL1 activates the DREB2A promoter.The DREB2A mutants are also heat stress sensitive at the reproductive stage,and DEREB2A is epistatic to NFXL1 in regulating thermotolerance in flower buds.It is known that HSFA3,a direct target of DREB2A,regulates the expression of heat shock proteins genes under heat stress conditions.Thus,our findings establish NFXL1 as a critical upstream regulator of DREB2A in the transcriptional cassette responsible for heat stress responses required for reproductive thermotolerance in Arabidopsis.

Evaluation of transmission risk of respiratory particles under different ventilation strategies in an elevator

People in elevators are at risk of respiratory infection because the elevator cabin is crowded and has poor ventilation.The exhaled particles may be inhaled by the susceptible person,deposited on the surface and suspended in the elevator,which can result in direct and indirect transmission.However,whether the air vent designs adopted in the elevator can effectively reduce the transmission risk of respiratory particles remains unknown.In this study,the dispersion of particles under four common ventilation strategies used in the commercial elevator was investigated by proven computational fluid dynamics(CFD)simulations.The flow field was simulated with the RNG κ-ξ turbulence model and the Lagrangian method was adopted to track particle trajectories.The effects of air vent layout and airflow rate on particle transmission were analyzed.We found that more than 50% of exhaled particles(average value)were suspended in the cabin and difficult to discharge under the investigated ventilation strategies.The deposited fraction of particles on the susceptible person reached up to 39.14% for infiltration ventilation,which led to a high risk of contact infection.Increasing the ventilation rate could not significantly reduce the inhalation proportion of particles due to the poor airflow distribution inside the elevator.A more proper ventilation strategy should be explored for the elevator to control transmission risk.

Spatial patterns of zooplankton abundance,biovolume,and size structure in response to environmental variables:a case study in the Yellow Sea and East China Sea

The Yellow Sea(YS)and East China Sea(ECS)are highly dynamic marginal seas of the northwestern Pacific Ocean.To gain an in-depth understanding of zooplankton community structure,zooplankton abundance,biovolume,and size structure in summer 2017 in the YS and ECS were assessed using ZooScan imaging analysis.Zooplankton abundance and biovolume ranged 2.94–1187.14 inds./m^(3)and 3.13–3438.51 mm^(3)/m^(3),respectively.Based on the biovolume data of the categorized size classes of 26 identified taxonomic groups,the zooplankton community was classified into five groups,and each group was coupled with distinctive oceanographic features.Under the influence of the Yellow Sea Cold Water Mass,the Yellow Sea offshore group featured the lowest bottom temperature(10.84±3.42℃)and the most abundant Calanoids(mainly in the 2–3 mm size class).In the Yellow Sea inshore group,Hydrozoans showed the largest biovolume and dominated in the 3–4-mm and>5-mm size classes.The East China Sea offshore group,which was affected by the Kuroshio Branch Current,featured high temperature and salinity,and the lowest bottom dissolved oxygen(2.58±0.5 mg/L).The lowest values of zooplankton abundance and biovolume in the East China Sea offshore group might be attributed to the bottom dissolved oxygen contents.The East China Sea inshore group,which was mainly influenced by the Zhejiang-Fujian Coastal Current and Changjiang Diluted Water,was characterized by high chlorophyll a and the largest biovolume of carnivorous Siphonophores(280.82±303.37 mm^(3)/m^(3)).The Changjiang River estuary offshore group showed the most abundant Cyclopoids,which might be associated with the less turbid water mass in this region.Seawater temperature was considered the most important factor in shaping the size compositions of Calanoids in different groups.

Heavy metal(loid)s(HMs)pollution has become a common and complex problem in industrial parks due to rapid industrialization and urbanization.Here,soil and groundwater were sampled from a retired industrial park to investigate the pollution characteristics of HMs.Results show that Ni,Pb,Cr,Zn,Cd,and Cu were the typical HMs in the soil.Source analysis with the positive matrix factorization model indicates that HMs in the topsoil stemmed from industrial activities,traffic emission,and natural source,and the groundwater HMs originated from industrial activities,groundwater-soil interaction,groundwater-rock interaction,and atmosphere deposition.The sequential extraction of soil HMs reveals that As and Hg were mainly distributed in the residue fraction,while Ni,Pb,Cr,Zn,Cd,and Cu mainly existed in the mobile fraction.Most HMs either in the total concentration or in the bioavailable fraction preferred to retain in soil as indicated by their high soil-water partitioning coefficients(K_(d)),and the K_(d) values were correlated with soil pH,groundwater redox potential,and dissolved oxygen.The relative stable soil-groundwater circumstance and the low active fraction contents limited the vertical migration of soil HMs and their release to groundwater.These findings increase our knowledge about HMs pollution characteristics of traditional industrial parks and provide a protocol for HMs pollution scrutinizing in large zones.

With the rapid development of electric power systems,load estimation plays an important role in system operation and planning.Usually,load estimation techniques contain traditional,time series,regression analysis-based,and machine learning-based estimation.Since the machine learning-based method can lead to better performance,in this paper,a deep learning-based load estimation algorithm using image fingerprint and attention mechanism is proposed.First,an image fingerprint construction is proposed for training data.After the data preprocessing,the training data matrix is constructed by the cyclic shift and cubic spline interpolation.Then,the linear mapping and the gray-color transformation method are proposed to form the color image fingerprint.Second,a convolutional neural network(CNN)combined with an attentionmechanism is proposed for training performance improvement.At last,an experiment is carried out to evaluate the estimation performance.Compared with the support vector machine method,CNN method and long short-term memory method,the proposed algorithm has the best load estimation performance.

Elastohydrodynamic lubrication(EHL)point contact occurs between two rough surfaces at the mesoscopic level,while the interaction of rough surfaces involves contact between asperities at the microscale level.In most cases,the contact between asperities within an interface takes the form of lateral contact rather than peak contact.Regions devoid of contact asperities are filled with lubricating oil.However,conventional models often oversimplify lateral contact forms as interactions between asperities and a smooth,rigid plane.These simplifications fail to accurately represent the true contact conditions and can lead to inaccuracies in the analysis of EHL’s contact performance.To address this issue,we have developed a novel EHL interface model comprising two rough surfaces.This model allows us to explore the influence of asperity height,contact angle,and contact azimuth angle on EHL interface performance.

Dear Editor,Tuberculosis(TB)is an infectious disease which has been considered as a global public health emergency problem during the past 25y.The World Health Organization has estimated that approximately one-third of the world’s population are infected by TB.However,only 10%of patients with TB is symptomatic.TB can affect multiple organs throughout the body.Eighty percent of those infected are pulmonary TB.Ocular TB can occur with or without lung or other systemic TB.It is a rare extrapulmonary form of the disease and occupied about 3.5%–5%of all of the TB.It should not be disregarded as its potential influence on vision loss.The intraocular and/or extraocular structures could be impacted by ocular TB[1].Uvea was the most common affected structure[2].

Currently,as the electromagnetic(EM)environment becomes increasingly complex,single-function EM materials can hardly resist the increasing electromagnetic interference(EMI),and there is an urgent need to develop multifunctional EM materials.In this work,multifunctional WSe_(2)/Co_(3)C was prepared by simple hydrothermal methods.Its dielectric performance and EM response were investigated.Efficient absorption,shielding performance,and energy conversion devices were customized.By tailoring the loading content,WSe_(2)/Co_(3)C can switch between EM absorption and EMI shielding.The maximum shielding effectiveness(SE)of WSe_(2)/Co_(3)C reached 36 dB,and high reflection loss(RL)of-60.28 dB and wide effective absorption bandwidth(EAB)of 6.16 GHz can be obtained at low thickness.The multiple EM attenuation mechanisms brought by the combination of two-dimensional(2D)WSe_(2) and magnetic Co_(3)C are considered to be the main reason for the enhanced EM attenuation ability.The WSe_(2)/Co_(3)C composite provides a viable candidate for developing multifunctional EM materials in 2–18 GHz.
