Bacillus anthracis lethal toxin(LT)is a determinant of lethal *** function in myeloid cells is required for bacterial dissemination,and LT itself can directly trigger dysfunction of the cardiovascular *** interplay between LT and the host responses is important in the pathogenesis,but our knowledge on this interplay remains *** necrosis factor-a(TNF-a)is a pleiotropic pro-inflammatory cytokine induced by bacterial *** LT accumulates and cytokines,predominantly TNF,amass during *** infection,co-treatment of TNF+LT in mice was used to mimic in vivo conditions for LT to function in inflamed *** marrow transplantation and genetically engineered mice showed unexpectedly that the death of intestinal epithelial cells(IECs)rather than that of hematopoietic cells led to LT+TNF-induced *** of p38a mitogen-activated protein kinase(MAPK)signaling by LT in IECs promoted TNF-induced apoptosis and necroptosis of IECs,leading to intestinal damage and mouse ***,p38a inhibition by LT enhanced TNF-mediated cell death in human colon epithelial HT-29 *** intestinal damage is one of the leading causes of lethality in anthrax patients,the IEC damage caused by LT+TNF would most likely be a mechanism underneath this clinical manifestation and could be a target for interventions.

Nrf2-mediated ferroptosis of spermatogenic cells involved in male reproductive toxicity induced by polystyrene nanoplastics in mice

Geophysical fluid dynamics(GFD)is an interdisciplinary field that studies the large-scale motion of fluids in the natural *** a wide range of applications such as weather forecasts and climate prediction,GFD employs various research approaches including in-situ observations,satellite measurements,numerical simulations,theoretical analysis,artificial intelligence,and physical model experiments in *** these approaches,rotating tank experiments provide a valuable tool for simulating naturally-occurring fluid motions in *** proportional scaling and proper techniques,scientists can reproduce multi-scale physical processes of stratified fluids in the rotation system,which allows for the simulation of essential characteristics of fluid motions in the atmosphere and *** this review,rotating tanks of various scales in the world are introduced,as these tanks have been actively used to explore fundamental scientific questions in ocean and atmosphere *** illustrate the GFD experiments,three representative cases are presented to demonstrate the frontier achievements in the the GFD study by using rotating tank experiments:mesoscale eddies in the ocean,convection processes,and plume *** references for the experimental procedures are *** studies are encouraged to further explore the utilization of rotating tanks with improvements in experimental design and integration of other research *** is a promising direction of GFD to help enhance our understanding of the complex nature of fluid motions in the natural world and to address the challenges posed by global environmental changes.

High-Performance Anion Exchange Membrane fuel Cells Enabled by Nitrogen Configuration Optimization in Graphene-Coated Nickel for Enhanced Hydrogen Oxidation

Anion exchange membrane fuel cell （AEMFC） technology is attracting intensive attention,due to its great potential by using non-precious-metal catalysts （NPMCs） in the cathode and cheap bipolar plate materials in alkaline ***,in such case,the kinetics of hydrogen oxidation reaction （HOR） in the anode is two orders of magnitude sluggish than that of acidic electrolytes,which is recognized as the grand challenge in this ***,we report the rationally designed Ni nanoparticles encapsulated by N-doped graphene layers （Ni@NG） using a facile pyrolysis *** on the density functional theory calculations and electrochemical performance analysis,it is witnessed that the rich Pyridinic-N within the graphene shell optimizes the binding energy of the intermediates,thus enabling the fundamentally enhanced activity for HOR with robust *** a proof of concept,the resultant Ni@NG sample as the anode with a low loading(1.8 mg cm-2) in AEMFCs delivers a high peak power density of500 mW cm-2,outperforming all of those of NPMC-based analogs ever reported.

Cell-free RNA(cf RNA) allows assessment of health, status, and phenotype of a variety of human organs and is a potential biomarker to non-invasively diagnose numerous diseases. Nevertheless, there is a lack of highly efficient and bias-free cf RNA isolation technologies due to the low abundance and instability of cf ***, we developed a reproducible and high-efficiency isolation technology for different types of cell-free nucleic acids(containing cf RNA and viral RNA) in serum/plasma based on the inclusion of nucleic acids by metal-organic framework(MOF) materials, which greatly improved the isolation efficiency and was able to preserve RNA integrity compared with the most widely used research kit method. Importantly, the quality of cf RNA extracted by the MOF method is about 10-fold that of the kit method, and the MOF method isolates more than three times as many different RNA types as the kit method. The whole transcriptome mapping characteristics of cf RNA in serum from patients with liver cancer was described and a cf RNA signature with six cf RNAs was identified to diagnose liver cancer with high diagnostic efficiency(area under curve = 0.905 in the independent validation cohort) using this MOF method. Thus, this new MOF isolation technique will advance the field of liquid biopsy, with the potential to diagnose liver cancer.

Without intervention,a considerable proportion of patients with metabolism‐associated fatty liver disease(MAFLD)will progress from simple steatosis to metabolism‐associated steatohepatitis(MASH),liver fibrosis,and even hepatocellular ***,the molecular mechanisms that control progressive MAFLD have yet to be fully ***,we unraveled that the expression of the N6-methyladenosine(m6A)methyltransferase METTL14 is remarkably downregulated in the livers of both patients and several murine models of MAFLD,whereas hepatocyte-specific depletion of this methyltransferase aggravated lipid accumulation,liver injury,and ***,hepatic Mettl14 overexpression alleviated the above pathophysiological changes in mice fed on a high-fat diet(HFD).Notably,in vivo and in vitro mechanistic studies indicated that METTL14 downregulation decreased the level of GLS2 by affecting the translation efficiency mediated by YTHDF1 in an m6A-depedent manner,which might help to form an oxidative stress microenvironment and accordingly recruit Cx3cr1^(+)Ccr2^(+)monocyte-derived macrophages(Mo-macs).In detail,Cx3cr1^(+)Ccr2^(+)Mo-macs can be categorized into M1-like macrophages and S100A4-positive macrophages and then further activate hepatic stellate cells(HSCs)to promote liver *** experiments revealed that CX3CR1 can activate the transcription of S100A4 via CX3CR1/MyD88/NF-κB signaling pathway in Cx3cr1^(+)Ccr2^(+)*** of METTL14 or GLS2,or interfering with this signal transduction pathway such as inhibiting MyD88 could ameliorate liver injuries and *** together,these findings indicate potential therapies for the treatment of MAFLD progression.

Assessing the Performance of a Dynamical Downscaling Simulation Driven by a Bias-Corrected CMIP6 Dataset for Asian Climate

In this study,we aim to assess dynamical downscaling simulations by utilizing a novel bias-corrected global climate model(GCM)data to drive a regional climate model(RCM)over the Asia-western North Pacific *** simulations were conducted with a 25-km grid spacing for the period 1980–*** first simulation(WRF_ERA5)was driven by the European Centre for Medium-Range Weather Forecasts Reanalysis 5(ERA5)dataset and served as the validation *** original GCM dataset(MPI-ESM1-2-HR model)was used to drive the second simulation(WRF_GCM),while the third simulation(WRF_GCMbc)was driven by the bias-corrected GCM *** bias-corrected GCM data has an ERA5-based mean and interannual variance and long-term trends derived from the ensemble mean of 18 CMIP6 *** demonstrate that the WRF_GCMbc significantly reduced the root-mean-square errors(RMSEs)of the climatological mean of downscaled variables,including temperature,precipitation,snow,wind,relative humidity,and planetary boundary layer height by 50%–90%compared to the WRF_***,the RMSEs of interannual-tointerdecadal variances of downscaled variables were reduced by 30%–60%.furthermore,the WRF_GCMbc better captured the annual cycle of the monsoon circulation and intraseasonal and day-to-day *** leading empirical orthogonal function(EOF)shows a monopole precipitation mode in the WRF_*** contrast,the WRF_GCMbc successfully reproduced the observed tri-pole mode of summer precipitation over eastern *** improvement could be attributed to a better-simulated location of the western North Pacific subtropical high in the WRF_GCMbc after GCM bias correction.

The increase in energy consumption and its collateral damage on the environment has encouraged the development of environment-friendly ceramic materials with good energy storage *** this work,(1-x)Na_(0.5)Bi_(0.5)TiO_(3)-xCa(Mg_(1/3)Nb_(2/3))O_(3) ceramics were synthesized by the solid-state reaction *** 0.88Na_(0.5)Bi_(0.5)TiO_(3)-0.12Ca(Mg_(1/3)Nb_(2/3))O_(3) ceramic exhibited a high recoverable energy storage density of 8.1 J/cm3 and energy storage efficiency of 82.4% at 550 kV/*** introduction of Ca(Mg_(1/3)Nb_(2/3))O_(3) reduced the grain size and increased the band gap,thereby enhancing the breakdown field strength of the ceramic *** method also resulted in good temperature stability(20–140℃),frequency stability(1–200 Hz),and fatigue stability over 106 *** addition,an ultrahigh power density of 187 MW/cm^(3) and a fast charge-discharge rate(t0.9=57.2 ns)can be obtained *** element method analysis revealed that the decrease of grain size was beneficial to the increase of breakdown field ***,the 0.88Na_(0.5)Bi_(0.5)TiO_(3)-0.12Ca(Mg_(1/3)Nb_(2/3))O_(3) ceramics resulted in high energy storage properties with good stability and were promising environment-friendly materials for advanced pulsed power systems applications.
