Modification of adipose mesenchymal stem cells-derived small extracellular vesicles with fibrin-targeting peptide CREKA for enhanced bone repair

The process of bone repair is highly regulated by a large number of bioactive factors.Thus,a“cocktail”of bioactive factors supplemented to the defect sites is desirable for bone repair.In this regard,small extracellular vesicles(sEVs)derived from mesenchymal stem cells hold great potential in tissue repair.Nevertheless,the poor homing and retention of sEVs greatly limited their possible clinical application.In the present work,DMPE-PEG-CREKA was inserted into the membrane of sEVs released from adipose-derived mesenchymal stem cells to obtain CREKA functionalized sEVs(CREKA-sEVs),which could target fibrin to accumulate and retain in bone defects.Our results showed that CREKA-sEVs,like sEVs,promoted the osteogenic differentiation of BMSCs,the angiogenic property of HUVECs,and modulated the polarization of macrophages in vitro.Furthermore,due to the improved fibrin-binding and retention capacity of CREKA-sEVs,they enhanced the bone repair substantially in the rat femoral defect model.This study provided a new strategy to improve the therapeutic efficiency of sEVs and showed that CREKA-sEVs had great application value in bone tissue repair.

目的分析某院门急诊利巴韦林注射液的使用情况,加强合理用药,为临床安全用药提供参考。方法在医院HIS系统中检索出使用了利巴韦林注射液的门急诊处方,回顾性研究2021年4~8月利巴韦林注射液在院门急诊的用量、患者年龄、诊断及与其他药物联用等情况,调查利巴韦林注射液在抗病毒治疗中的合理使用情况。结果共检索出含有利巴韦林注射液的处方565张,利巴韦林的应用于诊断为急性咽炎、急性上呼吸道感染、急性扁桃体炎占比达63.54%;诊断为咳嗽、头痛、头晕、腹痛占比达27.61%。结论利巴韦林注射液的应用存在严重的不合理用药情况,药学部门应加强医院临床用药管控,门急诊医生应严格控制抗病毒药的使用,按照药品说明书规定用药。

A HMM-based Mandarin Chinese Singing Voice Synthesis System

We propose a mandarin Chinese singing voice synthesis system, in which hidden Markov model(HMM)-based speech synthesis technique is used. A mandarin Chinese singing voice corpus is recorded and musical contextual features are well designed for training. F0 and spectrum of singing voice are simultaneously modeled with context-dependent HMMs. There is a new problem, F0 of singing voice is always sparse because of large amount of context, i.e., tempo and pitch of note, key, time signature and etc. So the features hardly ever appeared in the training data cannot be well obtained. To address this problem,difference between F0 of singing voice and that of musical score(DF0) is modeled by a single Viterbi training. To overcome the over-smoothing of the generated F0 contour, syllable level F0 model based on discrete cosine transforms(DCT) is applied, F0 contour is generated by integrating two-level statistical models.The experimental results demonstrate that the proposed system outperforms the baseline system in both objective and subjective evaluations. The proposed system can generate a more natural F0 contour. Furthermore, the syllable level F0 model can make singing voice more expressive.

针对Cu^(2+)合成探针的污染问题,以天然染料为检测Cu^(2+)探针,通过络合变色反应实现Cu^(2+)的可视化检测。利用吸收光谱法与比色法研究了天然染料的抗干扰性、耐酸碱性、溶解性及Cu^(2+)检测效果。结果表明,黄芩、苏木与紫胶红都能与Cu^(2+)产生变色反应,但仅有紫胶红能够选择性识别Cu^(2+),产生由红到黄的明显变色,吸收光谱蓝移57 nm;且紫胶红具有良好的耐酸碱性及水溶性,能够识别到20μmol/L的Cu^(2+);Cu^(2+)浓度与变色溶液相对吸光度呈良好线性关系,R^(2)=0.97295。

The importance of coil conductivity and eddy current effects in the analysis of electromagnetic forming process

Coil design and performance analysis are critical for the electromagnetic forming(EMF)process.However,previous studies have paid little attention to the coil con-ductivity,and the influence of eddy current effects in coil conductors on the forming process has not been well understood.This work aims to address these problems through numerical and experimental investigations.It is found that,due to the eddy current effects,the uneven current distribution appears in the coil conductors that should be considered when analysing the forming process,especially for the cases with large heights of coil conductors.Meanwhile,an important and interesting discovery is that the eddy current effects can greatly reduce the sensitivity of the coil conductivity with respect to the workpiece deformation,the Joule heating and the temperature rise in the coil.For instance,compared to a forming system with a copper coil,the forming height of the workpiece in the forming system with an AA5083‐O coil is reduced by less than 5%under the same discharge energy,while the copper conductivity is 3.3 times that of the latter.These results are helpful to understand the EMF process and achieve the optimal design of coils.

Unconventional oil and gas resources have become the most important and realistic field for increasing China’s domestic oil and gas reserves and production.At present,the production scale does not match the massive amount of resources and the rapid growth of proven geological reserves.The challenges of technology,cost,management,and methodology restrict large-scale and economic development.Based on successful practices,a"one engine with six gears"system engineering methodology is put forward,which includes life-cycle management,overall synergy,interdisciplinary cross-service integration,marketoriented operation,socialized support,digitalized management,and low-carbon and green development.The methodology has been proved to be effective in multiple unconventional oil and gas national demonstration areas,including the Jimusar continental shale oil demonstration area.Disruptive views are introduced-namely,that unconventional oil and gas do not necessarily yield a low return,nor do they necessarily have a low recovery factor.A determination to achieve economic benefit must be a pervasive underlying goal for managers and experts.Return and recovery factors,as primary focuses,must be adhered to during China’s development of unconventional oil and gas.The required methodology transformation includes a revolution in management systems to significantly decrease cost and increase production,resulting in technological innovation.

Methylselenized glucose:An efficient organoselenium fertilizer enhancing the selenium content in wheat grains

Selenium(Se)is an essential mineral element for human and other animals,and has been proved to improve plant growth and development and tolerance to different abiotic stresses.Selenium biofortification is considered to be a key strategy to increase the selenium content of edible parts of crops,which is helpful for improving human health.In this work,foliar fertilization with different concentrations and selenium forms was carried out on two wheat varieties at the flowering stage to compare the selenium enrichment effect of Na_(2)SeO_(3),methylselenized selenocysteine(MSC),methylselenized glucose(MSG)and methylselenized lactide(MSL)in wheat grains.Surprisingly,MSG was found to be the preferable fertilizer.After the application of MSG,the highest selenium content in wheat gains reached 6 mg/kg in this experiment,and the average selenium content was 2-4 times versus that of Na_(2)SeO_(3)application.Since MSG has high utilization rate and is easily available at relatively low cost,it can be employed as a potential selenium source for selenium biofortification to enhance the added value of agricultural industry.

Focal adhesion kinase signaling is necessary for the hydrogen sulfide-enhanced proliferation,migration,and invasion of HTR8/SVneo human trophoblasts

Objective:Hydrogen sulfide(H_(2)S)has been elucidated that it promotes migration and invasion in human placenta trophoblasts.However,the signaling pathway underlying H_(2)S-based regulation of trophoblasts remains unknown.Hence,we investigated the potential effect of sodium hydrosulfide(NaHS),an exogenous H_(2)S donor,on extravillous trophoblasts.Methods:The Cell Counting Kit-8 was used to detect the proliferative activity of trophoblasts and to screen the optimal concentration of NaHS.The migration and invasion of HTR8/SVneo cells were measured by Transwell assays.Gene expression was determined by quantitative real-time PCR analysis.Protein expression was determined by western blot.Results:We found that NaHS could promote the proliferation,migration,and invasion of HTR8/SVneo cells.The phosphorylation of focal adhesion kinase(FAK),Src,and extracellular signal-regulated kinase(ERK)were activated by NaHS.Moreover,NaHS also upregulated the expression of matrix metalloproteinase-2(MMP-2)and MMP-9,downregulated the expression of E-cadherin in HTR8/SVneo cells.The application of NaHS could increase the expression of cystathionine-β-synthase.Conclusion:Both FAK-Src signaling and the upstream signaling cascade of ERK activation play a significant important role in NaHS-induced proliferation,migration,and invasion via upregulating activity of MMP-2,MMP-9,and downregulating E-cadherin in HTR8/SVneo cells.These novel findings may provide a strong foundation for the clinical application of H_(2)S donor drugs.
