MFG-E8 Alleviates Cognitive Impairments Induced by Chronic Cerebral Hypoperfusion by Phagocytosing Myelin Debris and Promoting Remyelination

Chronic cerebral hypoperfusion is one of the pathophysiological mechanisms contributing to cognitive decline by causing white matter injury.Microglia phagocytosing myelin debris in a timely manner can promote remyelination and contribute to the repair of white matter.However,milk fat globule-epidermal growth factor-factor 8(MFG-E8),a microglial phagocytosis-related protein,has not been well studied in hypoperfusion-related cognitive dysfunction.We found that the expression of MFG-E8 was significantly decreased in the brain of mice after bilateral carotid artery stenosis(BCAS).MFG-E8 knockout mice demonstrated more severe BCAS-induced cognitive impairments in the behavioral tests.In addition,we discovered that the deletion of MFG-E8 aggravated white matter damage and the destruction of myelin microstructure through fluorescent staining and electron microscopy.Meanwhile,MFG-E8 overexpression by AAV improved white matter injury and increased the number of mature oligodendrocytes after BCAS.Moreover,in vitro and in vivo experiments showed that MFG-E8 could enhance the phagocytic function of microglia via theαVβ3/αVβ5/Rac1 pathway and IGF-1 production to promote the differentiation of oligodendrocyte progenitor cells into mature oligodendrocytes.Interestingly,we found that MFG-E8 was mainly derived from astrocytes,not microglia.Our findings suggest that MFG-E8 is a potential therapeutic target for cognitive impairments following cerebral hypoperfusion.

Moyamoya disease(MMD)is a chronic occlusive cerebrovascular disease with the development of a network of abnormal vessels.Immune inflammation is associated with the occurrence and development of MMD.However,the mechanisms underlying the formation of the abnormal vascular network remain unclear.Twenty-eight patients with MMD,26 ischemic stroke patients,and 26 unrelated healthy volunteers were enrolled in this study The data showed that the levels of granulocyte-macrophage colony-stimulating factor(GM-CSF)were higher in MMD patients than in healthy controls(P<0.01),and GM-CSF was mainly from Th1 and Th17 cells in MMD.We found that increased GM-CSF drove monocytes to secrete a series of cytokines associated with angiogenesis,inflammation,and chemotaxis.In summary,our findings demonstrate for the first time the important involvement of GM-CSF in MMD and that GM-CSF is an important factor in the formation of abnormal vascular networks in MMD.

Astrocytic Gap Junctions Contribute to Aberrant Neuronal Synchronization in a Mouse Model of MeCP2 Duplication Syndrome

Abnormal synchronous neuronal activity has been widely detected by brain imaging of autistic patients,but its underlying neural mechanism remains unclear.Compared with wild-type mice,our in vivo two-photon imaging showed that transgenic(Tgl)mice over-expressing human autism risk gene MeCP2 exhibited higher neuronal synchrony in the young but lower synchrony in the adult stage.Whole-cell recording of neuronal pairs in brain slices revealed that higher neuronal synchrony in young postnatal Tgl mice was atributed mainly to more prevalent giant slow inward currents(SICs).Both in vivo and slice imaging further demonstrated more dynamic activity and higher synchrony in astrocytes from young Tgl mice.Blocking astrocytic gap junctions markedly decreased the generation of SICs and overall cell synchrony in the Tgl brain.Furthermore,the expression level of Cx43 protein and the coupling efficiency of astrocyte gap junctions remained unchanged in Tgi mice.Thus,astrocytic gap junctions facilitate but do not act as a direct trigger for the abnormal neuronal synchrony in young Tgl mice,revealing the potential role of the astrocyte network in the pathogenesis of MeCP2 duplication syndrome.

目的探讨铸瓷粘接桥修复深覆[牙合]伴单颗前牙缺失患者的临床疗效。方法选取2023年1月—2023年6月安徽医专附属口腔医院就诊的30例深覆[牙合]伴前牙单颗缺失的患者作为研究对象,唇侧制作铸瓷粘接桥,术后1周和6个月分别检查修复体脱落、修复体折裂、边缘密合性、继发龋、边缘着色、牙周健康状况等。结果术后1周修复效果较好,无修复体脱落及折断,边缘密合性好、无着色,牙周健康。术后6个月检查见2例修复体脱落,重新粘结后继续使用,1例探针可探及边缘,牙龈轻度炎症,其余27例未见异常,未见修复体脱落和折断,边缘密合性好,基牙无龋坏,牙周健康。结论铸瓷粘接桥修深覆[牙合]伴单颗前牙缺失患者短期临床效果较好。

Chronic cerebral hypoperfusion leads to white matter injury(WMI),which subsequently causes neurodegeneration and even cognitive impairment.However,due to the lack of treatment specifically for WMI,novel recognized and effective therapeutic strategies are urgently needed.In this study,we found that honokiol and magnolol,two compounds derived from Magnolia officinalis,significantly facilitated the differentiation of primary oligodendrocyte precursor cells(OPCs)into mature oligodendrocytes,with a more prominent effect of the former compound.Moreover,our results demonstrated that honokiol treatment improved myelin injury,induced mature oligodendrocyte protein expression,attenuated cognitive decline,promoted oligodendrocyte regeneration,and inhibited astrocytic activation in the bilateral carotid artery stenosis model.Mechanistically,honokiol increased the phosphorylation of serine/threonine kinase(Akt)and mammalian target of rapamycin(mTOR)by activating cannabinoid receptor 1 during OPC differentiation.Collectively,our study indicates that honokiol might serve as a potential treatment for WMI in chronic cerebral ischemia.

Research on the evolutionary game of the supervision of civil aviation dangerous goods transportation training

This study is carried out to promote the precise supervision of dangerous goods transportation training,improve the efficiency of civil aviation supervision and further ensure the safety of air transportation.First,from the perspective of behaviour interaction and interest demands,evolutionary game theory is used to construct a tripartite evolutionary game model of dangerous goods transportation training institutions,the Civil Aviation Administration of China(CAAC)and the public.Then,the evolutionary game equilibrium of the system is further analysed under the joint action of the three parties.Finally,the influences of important parameters of the model on the behavioural strategy selection of the three parties are investigated via MATLAB numerical simulation.The conclusions reveal that the system has three evolutionarily stable strategies under different parameters,namely(non-compliant training,supervision,non-participation in supervision),(non-compliant training,supervision,participation in supervision)and(compliant training,supervision,non-participation in supervision).Moreover,the CAAC supervision cost,the fine amount,the supervision cost of public participation,the supervision success rate and the reporting reward amount are the main parameters that affect the behavioural strategy selection of the tripartite game players.The conclusions and suggestions of this study provide a decision-making basis and guidance for the supervision and management of civil aviation dangerous goods transportation training.

FERONIA-mediated TIR1/AFB2 oxidation stimulates auxin signaling in Arabidopsis

The phytohormone auxin plays a pivotal role in governing plant growth and development.Although the TRANSPORT INHIBITOR RESPONSE1/AUXIN SIGNALING F-BOX(TIR1/AFB)receptors function in both the nucleus and cytoplasm,the mechanism governing the distribution of TIR1/AFBs between these cellular compartments remains unknown.In this study,we demonstrate that auxin-mediated oxidation of TIR1/AFB2 is essential for their targeting to the nucleus.We showed that small active molecules,reactive oxygen species(ROS)and nitric oxide(NO),are indispensable for the nucleo-cytoplasmic distribution of TIR1/AFB2 in trichoblasts and root hairs.Further studies revealed that this process is regulated by the FERONIA receptor kinase–NADPH oxidase signaling pathway.Interestingly,ROS and NO initiate oxidative modifications in TIR1C140/516 and AFB2C135/511,facilitating their subsequent nuclear import.The oxidized forms of TIR1C140/516 and AFB2C135/511 play a crucial role in enhancing the function of TIR1 and AFB2 in transcriptional auxin responses.Collectively,our study reveals a novel mechanism by which auxin stimulates the transport of TIR1/AFB2 from the cytoplasm to the nucleus,orchestrated by the FERONIA–ROS signaling pathway.

Comparisons of Wave Force Model Effects on the Structural Responses and Fatigue Loads of a Semi-Submersible Floating Wind Turbine

The selection of wave force models will significantly impact the structural responses of floating wind turbines.In this study,comparisons of wave force model effects on the structural responses and fatigue loads of a semi-submersible floating wind turbine(SFWT)were conducted.Simulations were performed by employing the Morison equation(ME)with linear or second-order wave kinematics and potential flow theory(PFT)with first-or second-order wave forces.A comparison of regular waves,irregular waves,and coupled wind/waves analyses with the experimental data showed that many of the simulation results and experimental data are relatively consistent.However,notable discrepancies are found in the response amplitude operators for platform heave,tower base bending moment,and tension in mooring lines.PFT models give more satisfactory results of heave but more significant discrepan-cies in tower base bending moment than the ME models.In irregular wave analyses,low-frequency resonances were captured by PFT models with second-order difference-frequency terms,and high-frequency resonances were captured by the ME models or PFT models with second-order sum-frequency terms.These force models capture the response frequencies but do not reasonably predict the response amplitudes.The coupled wind/waves analyses showed more satisfactory results than the wave-only analyses.However,an important detail to note is that this satisfactory result is based on the overprediction of wind-induced responses.

A Task Scheduling Algorithm Based on Clustering Pre-processing in Space-Based Information Network

With the diversification of space-based information network task requirements and the dramatic increase in demand, the efficient scheduling of various tasks in space-based information network becomes a new challenge. To address the problems of a limited number of resources and resource heterogeneity in the space-based information network, we propose a bilateral pre-processing model for tasks and resources in the scheduling pre-processing stage. We use an improved fuzzy clustering method to cluster tasks and resources and design coding rules and matching methods to match similar categories to improve the clustering effect. We propose a space-based information network task scheduling strategy based on an ant colony simulated annealing algorithm for the problems of high latency of space-based information network communication and high resource dynamics. The strategy can efficiently complete the task and resource matching and improve the task scheduling performance. The experimental results show that our proposed task scheduling strategy has less task execution time and higher resource utilization than other algorithms under the same experimental conditions. It has significantly improved scheduling performance.

Trophic niche and adaptation in highland lizards:sex has greater influences than species matching

The plateau environments are typically arid,cool,and high altitude,posing formidable challenges to wildlife survival due to resource scarcity and harsh conditions.Unraveling ecological adaptability in severe conditions requires a deeper understanding of the niche characteristics of plateau species.Trophic niche,which is a comprehensive indicator describing the energy acquisition strategy of animals,remains relatively understudied in plateau species.Here,by combining stable isotopes and morphological data,we quantified the trophic niches of two allopatric lizard species(Phrynocephalus vlangalii and P.erythrurus)that live in the hinterland of the Qinghai–Tibetan Plateau,and explored how their trophic niches correlate with morphological and environmental factors.While both trophic niche and morphological traits were similar between species,noteworthy distinctions were observed between male and female Phrynocephalus lizards.The morphological traits associated with predation(i.***.limb length and head size)and reproduction(i.***.abdomen length),annual mean temperature,and sex played influential roles in shifting trophic niches.These results imply that sexual dimorphism may facilitate inter-sex divergence in resource utilization,leading to trophic niche variations in the highland lizards.Furthermore,extreme environmental stress can constrain interspecific divergence in morphological and trophic traits.Our findings illustrate the dynamic variations of trophic niches in highland lizards,contributing to a more comprehensive understanding of the adaptation strategies employed by lizard species in plateau environments.
