An approach was described to produce a wound area in vitro using laminar flow technique to selectively remove cells in micro fluidic channels. Cell migration which plays an important role in the process of wound healing can be observed dynamically by this method. In order to prove our system, we have studied its properties by injecting NIH-3T3 cells into main channel and added several reagents to observe the mediatory influence. The results reveal that fibroblast growth factor-2（FGF-2）, insulin-like growth factor 1（IGF-Ⅰ）, platelet-derived growth factor BB（PDGF-BB） and ascorbic acid（Vc） groups can promote cell migration compared with the control group. But the average migration distance was diminished in dexamethasone group. It can be concluded that this system can be used to analyze the process of wound healing.

Background: During the coronavirus disease 2019(COVID-19)pandemic,seasonal influenza activity declined globally and remained below previous seasonal levels,but intensified in China since 2021.Preventive measures to COVID-19 accompanied by different epidemic characteristics of influenza in different regions of the world.To better respond to influenza outbreaks under the COVID-19 pandemic,we analyzed the epidemiology,antigenic and genetic characteristics,and antiviral susceptibility of influenza viruses in the mainland of China during 2020–2021.Methods: Respiratory specimens from influenza like illness cases were collected by sentinel hospitals and sent to network laboratories in Chinese National Influenza Surveillance Network.Antigenic mutation analysis of influenza virus isolates was performed by hemagglutination inhibition assay.Next-generation sequencing was used for genetic analyses.We also conducted molecular characterization and phylogenetic analysis of circulating influenza viruses.Viruses were tested for resistance to antiviral medications using phenotypic and/or sequence-based methods.Results: In the mainland of China,influenza activity recovered in 2021 compared with that in 2020 and intensified during the traditional influenza winter season,but it did not exceed the peak in previous years.Almost all viruses isolated during the study period were of the B/Victoria lineage and were characterized by genetic diversity,with the subgroup 1A.3a.2 viruses currently predominated.37.8%viruses tested were antigenically similar to reference viruses representing the components of the vaccine for the 2020–2021 and 2021–2022 Northern Hemisphere influenza seasons.In addition,China has a unique subgroup of 1A.3a.1 viruses.All viruses tested were sensitive to neuraminidase inhibitors and endonuclease inhibitors,except two B/Victoria lineage viruses identified to have reduced sensitivity to neuraminidase inhibitors.Conclusions: Influenza activity increased in the mainland of China in 2021,and caused flu season in the winte

Dear Editor We present a case of spontaneous dislocation of a toric implantable collamer lens(TICL)resulting in corneal endothelial decompensation 8y after implantation.Since 2006,by helping people take off their glasses,the Visian TICL(Staar Surgical Co,Monrovia,CA,USA)have gained popularity in China.Though complications of intraocular pressure(IOP)elevation,cataract formation and endothelial damage exist as relevant problems,implantable collamer lens(ICL)implantation has been generally accepted as a viable option to correct moderate to high ametropia[1-2].

Hydrofunctionalization of alkenes and alkynes is increasingly important in preparing and modifying carbon-centered organic molecules such as pharmaceuticals,agrochemicals,andmaterial precursors.The development of transition-metal-catalysts has revolutionized the hydrofunctionalization of unsaturated hydrocarbons,greatly improving the synthetic efficiency and selectivity of these reactions.In recent years,notable progress has been achieved in low-cost metal-catalyzed alkene and alkyne hydrofunctionalization reactions following the utilization of traditional precious metal catalysts,including palladium,platinum,and rhodium.Earth-abundant cobalt catalysts,which are low in toxicity and exhibit unique reactivity and selectivity,are widely implemented in academic and industrial research.In this review,we highlight recent examples of cobalt-catalyzed hydrofunctionalization of alkenes and alkynes to provide an overview of hydrofunctionalization methods,particularly hydroalkylation reactions.Hydroalkylation reactions greatly expand the scope and utility of cobalt-catalyzed couplings of carbon—carbon bonds.We also highlight the mechanistic considerations and selective determinants involved.We hope our discussion will be informative for future developments in cobalt catalysis and hydrofunctionalization reactions.

Neural stem cells promote neuroplasticity: a promising therapeutic strategy for the treatment of Alzheimer’s disease

Recent studies have demonstrated that neuroplasticity,such as synaptic plasticity and neurogenesis,exists throughout the normal lifespan but declines with age and is significantly impaired in individuals with Alzheimer’s disease.Hence,promoting neuroplasticity may represent an effective strategy with which Alzheimer’s disease can be alleviated.Due to their significant ability to self-renew,differentiate,and migrate,neural stem cells play an essential role in reversing synaptic and neuronal damage,reducing the pathology of Alzheimer’s disease,including amyloid-β,tau protein,and neuroinflammation,and secreting neurotrophic factors and growth factors that are related to plasticity.These events can promote synaptic plasticity and neurogenesis to repair the microenvironment of the mammalian brain.Consequently,neural stem cells are considered to represent a potential regenerative therapy with which to improve Alzheimer’s disease and other neurodegenerative diseases.In this review,we discuss how neural stem cells regulate neuroplasticity and optimize their effects to enhance their potential for treating Alzheimer’s disease in the clinic.

Objective Renal fibrosis is the ultimate pathway of various forms of acute and chronic kidney damage.Notably,the knockout of transient receptor potential channel 6(TRPC6)has shown promise in alleviating renal fibrosis.However,the regulatory impact of TRPC6 on renal fibrosis remains unclear.Methods In vivo,TRPC6 knockout(TRPC6−/−)mice and age-matched 129 SvEv(WT)mice underwent unilateral renal ischemia-reperfusion(uIR)injury surgery on the left renal pedicle or sham operation.Kidneys and serum were collected on days 7,14,21,and 28 after euthanasia.In vitro,primary tubular epithelial cells(PTECs)were isolated from TRPC6−/−and WT mice,followed by treatment with transforming growth factorβ1(TGFβ1)for 72 h.The anti-fibrotic effect of TRPC6−/−and the underlying mechanisms were assessed through hematoxylin-eosin staining,Masson staining,immunostaining,qRT-PCR,and Western blotting.Results Increased TRPC6 expression was observed in uIR mice and PTECs treated with TGFβ1.TRPC6−/−alleviated renal fibrosis by reducing the expression of fibrotic markers(Col-1,α-SMA,and vimentin),as well as decreasing the apoptosis and inflammation of PTECs during fibrotic progression both in vivo and in vitro.Additionally,we found that the phosphatidylinositol 3-kinase(PI3K)/protein kinase B(AKT)/glycogen synthase kinase 3 beta(GSK3β)signaling pathway,a pivotal player in renal fibrosis,was down-regulated following TRPC6 deletion.Conclusion These results suggest that the ablation of TRPC6 may mitigate renal fibrosis by inhibiting the apoptosis and inflammation of PTECs through down-regulation of the PI3K/AKT/GSK3βpathway.Targeting TRPC6 could be a novel therapeutic strategy for preventing chronic kidney disease.

The available modelling data shortage issue makes it difficult to guarantee the performance of data-driven building energy prediction(BEP)models for both the newly built buildings and existing information-poor buildings.Both knowledge transfer learning(KTL)and data incremental learning(DIL)can address the data shortage issue of such buildings.For new building scenarios with continuous data accumulation,the performance of BEP models has not been fully investigated considering the data accumulation dynamics.DIL,which can learn dynamic features from accumulated data adapting to the developing trend of new building time-series data and extend BEP model's knowledge,has been rarely studied.Previous studies have shown that the performance of KTL models trained with fixed data can be further improved in scenarios with dynamically changing data.Hence,this study proposes an improved transfer learning cross-BEP strategy continuously updated using the coarse data incremental(CDI)manner.The hybrid KTL-DIL strategy(LSTM-DANN-CDI)uses domain adversarial neural network(DANN)for KLT and long short-term memory(LSTM)as the Baseline BEP model.Performance evaluation is conducted to systematically qualify the effectiveness and applicability of KTL and improved KTL-DIL.Real-world data from six-type 36 buildings of six types are adopted to evaluate the performance of KTL and KTL-DIL in data-driven BEP tasks considering factors like the model increment time interval,the available target and source building data volumes.Compared with LSTM,results indicate that KTL(LSTM-DANN)and the proposed KTL-DIL(LSTM-DANN-CDI)can significantly improve the BEP performance for new buildings with limited data.Compared with the pure KTL strategy LSTM-DANN,the improved KTL-DIL strategy LSTM-DANN-CDI has better prediction performance with an average performance improvement ratio of 60%.

BACKGROUND Podocyte apoptosis plays a vital role in proteinuria pathogenesis in diabetic nephropathy(DN).The regulatory relationship between long noncoding RNAs(lncRNAs)and podocyte apoptosis has recently become another research hot spot in the DN field.AIM To investigate whether lncRNA protein-disulfide isomerase-associated 3(Pdia3)could regulate podocyte apoptosis through miR-139-3p and revealed the underlying mechanism.METHODS Using normal glucose or high glucose(HG)-cultured podocytes,the cellular functions and exact mechanisms underlying the regulatory effects of lncRNA Pdia3 on podocyte apoptosis and endoplasmic reticulum stress(ERS)were explored.LncRNA Pdia3 and miR-139-3p expression were measured through quantitative real-time polymerase chain reaction.Relative cell viability was detected through the cell counting kit-8 colorimetric assay.The podocyte apoptosis rate in each group was measured through flow cytometry.The interaction between lncRNA Pdia3 and miR-139-3p was examined through the dual luciferase reporter assay.Finally,western blotting was performed to detect the effect of lncRNA Pdia3 on podocyte apoptosis and ERS via miR-139-3p.RESULTS The expression of lncRNA Pdia3 was significantly downregulated in HG-cultured podocytes.Next,lncRNA Pdia3 was involved in HG-induced podocyte apoptosis.Furthermore,the dual luciferase reporter assay confirmed the direct interaction between lncRNA Pdia3 and miR-139-3p.LncRNA Pdia3 overexpression attenuated podocyte apoptosis and ERS through miR-139-3p in HG-cultured podocytes.CONCLUSION Taken together,this study demonstrated that lncRNA Pdia3 overexpression could attenuate HG-induced podocyte apoptosis and ERS by acting as a competing endogenous RNA of miR-139-3p,which might provide a potential therapeutic target for DN.

High-precision parallel computing model of solute transport based on GPU acceleration

The scenario simulation analysis of water environmental emergencies is very important for risk prevention and control,and emergency response.To quickly and accurately simulate the transport and diffusion process of high-intensity pollutants during sudden environmental water pollution events,in this study,a high-precision pollution transport and diffusion model for unstructured grids based on Compute Unified Device Architecture(CUDA)is proposed.The finite volume method of a total variation diminishing limiter with the Kong proposed r-factor is used to reduce numerical diffusion and oscillation errors in the simulation of pollutants under sharp concentration conditions,and graphics processing unit acceleration technology is used to improve computational efficiency.The advection diffusion process of the model is verified numerically using two benchmark cases,and the efficiency of the model is evaluated using an engineering example.The results demonstrate that the model perform well in the simulation of material transport in the presence of sharp concentration.Additionally,it has high computational efficiency.The acceleration ratio is 46 times the single-thread acceleration effect of the original model.The efficiency of the accelerated model meet the requirements of an engineering application,and the rapid early warning and assessment of water pollution accidents is achieved.
