Clinicopathological significance and immunotherapeutic outcome of claudin 18.2 expression in advanced gastric cancer:A retrospective study

Objective: Immunotherapeutic outcomes and clinical characteristics of claudin 18 isoform 2 positive(CLDN18.2-positive) gastric cancer(GC) vary in different clinical studies, making it difficult to optimize antiCLDN18.2 therapy. We conducted a retrospective analysis to explore the association of CLDN18.2 expression with clinicopathological characteristics and immunotherapeutic outcomes in GC.Methods: A total of 536 advanced GC patients from 2019 to 2021 in the CT041-CG4006 and CT041-ST-01clinical trials were included in the analysis. CLDN18.2 expression on ≥40% of tumor cells(2+, 40%) and CLDN18.2 expression on ≥70% of tumor cells(2+, 70%) were considered the two levels of positively expressed GC. The clinicopathological characteristics and immunotherapy outcomes of GC patients were analyzed according to CLDN18.2 expression status.Results: CLDN18.2 was expressed in 57.6%(cut-off: 2+, 40%) and 48.9%(cut-off: 2+, 70%) of patients.Programmed death-ligand 1(PD-L1) and CLDN18.2 were co-expressed in 19.8% [combined positive score(CPS)≥1, CLDN18.2(cut-off: 2+, 40%)] and 17.2% [CPS≥5, CLDN18.2(cut-off: 2+, 70%)] of patients.CLDN18.2 expression positively correlated with younger age, female sex, non-gastroesophageal junction(nonGEJ), and diffuse phenotype(P<0.001). HER2 and PD-L1 expression were significantly lower in CLDN18.2-positive GC(both P<0.05). Uterine adnexa metastasis(P<0.001) was more frequent and liver metastasis(P<0.001)was less common in CLDN18.2-positive GC. Overall survival and immunotherapy-related progression-free survival(ir PFS) were inferior in the CLDN18.2-positive group.Conclusions: CLDN18.2-positive GC is associated with poor prognosis and worse immunotherapeutic outcomes. The combination of anti-CLDN18.2 therapy, anti-PD-L1/PD-1 therapy, and chemotherapy for GC requires further investigation.

Slickwater fracturing fluids are widely used in the development of unconventional oil and gas resources due to the advantages of low cost,low formation damage and high drag reduction performance.However,their performance is severely affected at high temperatures.Drag reducing agent is the key to determine the drag reducing performance of slickwater.In this work,in order to further improve the temperature resistance of slickwater,a temperature-resistant polymeric drag reducing agent(PDRA)was synthesized and used as the basis for preparing the temperature-resistant slickwater.The slickwater system was prepared with the compositions of 0.2 wt%PDRA,0.05 wt%drainage aid nonylphenol polyoxyethylene ether phosphate(NPEP)and 0.5 wt%anti-expansion agent polyepichlorohydrindimethylamine(PDM).The drag reduction ability,rheology properties,temperature and shear resistance ability,and core damage property of slickwater were systematically studied and evaluated.In contrast to on-site drag reducing agent(DRA)and HPAM,the temperature-resistant slickwater demonstrates enhanced drag reduction efficacy at 90℃,exhibiting superior temperature and shear resistance ability.Notably,the drag reduction retention rate for the slickwater achieved an impressive 90.52%after a 30-min shearing period.Additionally,the core damage is only 5.53%.We expect that this study can broaden the application of slickwater in high-temperature reservoirs and provide a theoretical basis for field applications.

Amyotrophic lateral sclerosis(ALS)is a fatal neurodegenerative disease characterized by progressive degeneration of motor neurons,and it demonstrates high clinical heterogeneity and complex genetic architecture.A variation within TRMT2B(c.1356G>T;p.K452N)was identified to be associated with ALS in a family comprising two patients with juvenile ALS(JALS).Two missense variations and one splicing variation were identified in 10 patients with ALS in a cohort with 910 patients with ALS,and three more variants were identified in a public ALS database including 3317 patients with ALS.A decreased number of mitochondria,swollen mitochondria,lower expression of ND1,decreased mitochondrial complex I activities,lower mitochondrial aerobic respiration,and a high level of ROS were observed functionally in patient-originated lymphoblastoid cell lines and TRMT2B interfering HEK293 cells.Further,TRMT2B variations overexpression cells also displayed decreased ND1.In conclusion,a novel JALS-associated gene called TRMT2B was identified,thus broadening the clinical and genetic spectrum of ALS.

Objective Studies on the relationship between iodine,vitamin A(VA),and vitamin D(VD)and thyroid function are limited.This study aimed to analyze iodine and thyroid-stimulating hormone(TSH)status and their possible relationships with VA,VD,and other factors in postpartum women.Methods A total of 1,311 mothers(896 lactating and 415 non-lactating)from Hebei,Zhejiang,and Guangxi provinces were included in this study.The urinary iodine concentration(UIC),TSH,VA,and VD were measured.Results The median UIC of total and lactating participants were 142.00µg/L and 139.95µg/L,respectively.The median TSH,VA,and VD levels in all the participants were 1.89 mIU/L,0.44μg/mL,and 24.04 ng/mL,respectively.No differences in the UIC were found between lactating and non-lactating mothers.UIC and TSH levels were significantly different among the three provinces.The rural UIC was higher than the urban UIC.Obese mothers had a higher UIC and a higher prevalence of excessive TSH.Higher UICs and TSHs levels were observed in both the VD deficiency and insufficiency groups than in the VD-sufficient group.After adjustment,no linear correlation was observed between UIC and VA/VD.No interaction was found between vitamins A/D and UIC on TSH levels.Conclusion The mothers in the present study had no iodine deficiency.Region,area type,BMI,and VD may be related to the iodine status or TSH levels.

A prospective microbiome analysis of the impact of physical exercise and blueberry supplementation on the gut microbiome of obesity-induced hypertensive in college students

The prevalence of high blood pressure is on the rise among younger individuals， and recent findings have linked exercise to changes in the diversity and configuration of gut microbiota. However， specific modifications to one's diet， such as incorporating blueberries while engaging in physical activity， can also influence this association. To explore the effects of exercise and blueberry consumption on gut bacteria， we conducted an eight-week study involving healthy and hypertensive college students. Our microbiome analysis revealed slight shifts in gut bacteria composition in response to increased physical activity， with significant changes in the diversity of the g＿[Eubacterium]＿eligens＿group observed in hypertensive college students who regularly consumed blueberries. Although exercise and blueberry supplementation had a negligible impact on the gut microbiota diversity of healthy college students， it significantly boosted the production of butyric acid bacteria in hypertensive young adults. These results suggest that the changes in gut bacteria variety and abundance brought about by blueberries may be tied to alterations in the body composition of patients with hypertension. This study aims to reveal the impact of blueberry on the hypertensive students from a unique perspective， suggesting the impact of taking blueberry while taking exercises.

Lithium–sulfur(Li–S)batteries promise high-energy-density potential to exceed the commercialized lithiumion batteries but suffer from limited cycling lifespan due to the side reactions between lithium polysulfides(LiPSs)and Li metal anodes.Herein,a three-way electrolyte with ternary solvents is proposed to enable high-energy-density and long-cycling Li–S pouch cells.Concretely,ternary solvents composed of 1,2-dimethoxyethane,di-isopropyl sulfide,and 1,3,5-trioxane are employed to guarantee smooth cathode kinetics,inhibit the parasitic reactions,and construct a robust solid electrolyte interphase,respectively.The cycling lifespan of Li–S coin cells with 50μm Li anodes and 4.0 mg cm^(−2) sulfur cathodes is prolonged from88 to 222 cycles using the three-way electrolyte.Nano-heterogeneous solvation structure of LiPSs and organic-rich solid electrolyte interphase are identified to improve the cycling stability of Li metal anodes.Consequently,a 3.0 Ah-level Li–S pouch cell with the three-way electrolyte realizes a high energy density of 405 Wh kg^(−1) and undergoes 27 cycles.Thiswork affords a three-way electrolyte recipe for suppressing the side reactions of LiPSs and inspires rational electrolyte design for practical high-energy-density and long-cycling Li–S batteries.

Analysis of explosion wave interactions and rock breaking effects during dual initiation

In blasting engineering, the location and number of detonation points, to a certain degree, regulate the propagation direction ofthe explosion stress wave and blasting effect. Herein, we examine the explosion wave field and rock breaking effect in terms of shockwave collision, stress change of the blast hole wall in the collision zone, and crack propagation in the collision zone. The produced shockwave on the collision surface has an intensity surpassing the sum of the intensities of the two colliding explosion shock waves. At the collisionlocation, the kinetic energy is transformed into potential energy with a reduction in particle velocity at the wave front and the wavefront pressure increases. The expansion form of the superposed shock wave is dumbbell-shaped, the shock wave velocity in the collisionarea is greater than the radial shock wave velocity, and the average propagation angle of the explosion shock waves is approximately 60°.Accordingly, a fitted relationship between blast hole wall stress and explosion wave propagation angle in the superposition area is plotted.Under the experimental conditions, the superimposed explosion wave stress of the blast hole wall is approximately 1.73 times the singleexplosionwave incident stress. The results of the model test and numerical simulations reveal that large-scale radial fracture cracks weregenerated on the blast hole wall in the superimposed area, and the width of the crack increased. The width of the large-scale radial fracturecracks formed by a strong impact is approximately 5% of the blast hole length. According to the characteristics of blast hole wallcompression, the mean peak pressures of the strongly superimposed area are approximately 1.48 and 1.84 times those of the weakly superimposedand nonsuperimposed areas, respectively.

Low Resource Chinese Geological Text Named Entity Recognition Based on Prompt Learning

Geological reports are a significant accomplishment for geologists involved in geological investigations and scientific research as they contain rich data and textual information.With the rapid development of science and technology,a large number of textual reports have accumulated in the field of geology.However,many non-hot topics and non-English speaking regions are neglected in mainstream geoscience databases for geological information mining,making it more challenging for some researchers to extract necessary information from these texts.Natural Language Processing(NLP)has obvious advantages in processing large amounts of textual data.The objective of this paper is to identify geological named entities from Chinese geological texts using NLP techniques.We propose the RoBERTa-Prompt-Tuning-NER method,which leverages the concept of Prompt Learning and requires only a small amount of annotated data to train superior models for recognizing geological named entities in low-resource dataset configurations.The RoBERTa layer captures context-based information and longer-distance dependencies through dynamic word vectors.Finally,we conducted experiments on the constructed Geological Named Entity Recognition(GNER)dataset.Our experimental results show that the proposed model achieves the highest F1 score of 80.64%among the four baseline algorithms,demonstrating the reliability and robustness of using the model for Named Entity Recognition of geological texts.
