Integrin binding peptides facilitate growth and interconnected vascular-like network formation of rat primary cortical vascular endothelial cells in vitro

Neovascularization and angiogenesis in the brain are important physiological processes for normal brain development and repair/regeneration following insults. Integrins are cell surface adhesion receptors mediating important function of cells such as survival, growth and development during tissue organization, differentiation and organogenesis. In this study, we used an integrin-binding array platform to identify the important types of integrins and their binding peptides that facilitate adhesion, growth, development, and vascular-like network formation of rat primary brain microvascular endothelial cells. Brain microvascular endothelial cells were isolated from rat brain on post-natal day 7. Cells were cultured in a custom-designed integrin array system containing short synthetic peptides binding to 16 types of integrins commonly expressed on cells in vertebrates. After 7 days of culture, the brain microvascular endothelial cells were processed for immunostaining with markers for endothelial cells including von Willibrand factor and platelet endothelial cell adhesion molecule. 5-Bromo-2′-dexoyuridine was added to the culture at 48 hours prior to fixation to assess cell proliferation. Among 16 integrins tested, we found that α5β1, αvβ5 and αvβ8 greatly promoted proliferation of endothelial cells in culture. To investigate the effect of integrin-binding peptides in promoting neovascularization and angiogenesis, the binding peptides to the above three types of integrins were immobilized to our custom-designed hydrogel in three-dimensional(3 D) culture of brain microvascular endothelial cells with the addition of vascular endothelial growth factor. 为揭示隧道进出口抗滑性能的空间变化规律,现采用摆式仪与路面构造深度仪,通过现场实验的方法,分别采集了水头特长隧道、西浮图特长隧道、韩信岭长隧道、燕家岭中隧道、陈家山短隧的进出口摆值与构造深度。数据分析结果表明,隧道进口的摆值减小值明显大于隧道出口,说明隧道进口的减速行为对路面抗滑性能的影响更大;隧道进口摆值降低的终点为洞外50 m,超车道的摆值明显低于行车道,表明速度对摩擦因数的影响比载重大;隧道出口摆值降低的起点为洞门外30 m。此外,短隧道的路面抗滑性能沿隧道长度方向保持稳定,中隧道与长隧道的路面抗滑性能沿隧道长度方向呈平稳-下降-上升-平稳的变化规律。

Nitrogen(N) is an essential nutrient for food production. The rapid increase in population requires high inputs of N to meet the growing food demand. If not managed well, the substantial loss of N from the food system has multiple impacts on grain yield, air and water pollution, and the economic benefits of agricultural. Multi-objective(food security, environmental sustainability and economic sustainability) synergistic consideration of N management in the food system is still lacking. This study employed strategies for optimizing N management in the food system, using Quzhou County as a typical example on the North China Plain. Firstly, a food chain approach was adopted to understand drivers and reasons behind N losses from the food ***, a top-down approach was used to define multi-objective N management, taking into consideration food security, environmental sustainability and economic sustainability. Multi-objective N management aims to reduce N losses to the environment and increase N use efficiencies,while simultaneously increasing yields and economic benefits. Thirdly, 3R(reduce-retain-recycle) N management strategies were identified for specific crops and animals through a bottom-up approach and then analyzed the potential of these strategies to achieve the multi-objectives. Finally, there is a discussion of how to engage different stakeholders to promote the technologies implementation. This study provides new insights into the synergistic achievement of multi-objective N management in the food system and the development of environmentally-friendly agriculture.

Anti-corrosion inspection engineers are responsible for 14 degassing stations, which complete anti-corrosion examinations of desalination and dehydration containers under maintenance, detecting potential corrosion, drawing up the diagram illustrations and inspection reports of Rumaila's oil field and gas system, guiding the completion of equipment sandblasting and anti-corrosion inspections, and ensuring the fulfillment of other relevant work reports. Anti-corrosion inspection engineers can be called the "health doctors" of oil and gas systems.
