Collapse of a Deep Excavated Foundation Pit in the Soft Soils by 3-D FEM

In view of the collapse of a deep excavated foundation pit of the Xianghu subway underground station in Hangzhou of China,the main features of the accident are analyzed,and the induced factors of the accident are summarized. Then,a 3-D FEM analysis model is created to demonstrate the soil-support structures interaction system,and the effect of the main factors,such as the volume replacement ratio of the bottom soil reinforcing,the asymmetric ground overload,the embedded depth of the diaphragm wall,the shear strength of the bottom soils disturbed by the construction,and the excessive excavation of the bottom soil,are analyzed and compared. The results show that the ineffective original reinforcement plan for the bottom soft soil is the most prominent factor for the accident,and the disturbance effect of the deep excavation on the shear strength of the bottom soft soil is another significant factor for the accident. Meanwhile,if the reinforcement of the bottom soft soil is canceled,an appropriate extension of the diaphragm retaining walls to the under lying harder soil layer can also effectively prevent the collapse of the deep excavated foundation pit. In addition,the partly excessive excavation in the process has a great influence on the axial force of the most nearby horizontal support but few effect on the stability of the diaphragm wall. Thus,the excessive excavation of the bottom soils should not be the direct inducing factor for the accident. To the asymmetric ground overload,it should be the main factor inducing the different damage conditions of the diaphragm walls on different sides. According to the numerical modeling and actual engineering accident condition,the development process of the accident is also identified.

在基因治疗中,实现目的基因的调控表达是非常重要的。然而,传统基因载体的无调控地持续或不适当的表达会影响治疗效果,甚至可能带来致命的副作用。在本研究中,我们构建了一种带有DsRed红色荧光蛋白报告基因并可经RU486诱导的真核表达载体,并在体外评估了其调控表达作用。利用分子生物学技术,将DsRed基因和启动子,以及RU486系统构建成单一的质粒载体PDC-RURED,为减少RU486调控元件和基因表达元件之间的相互干扰,在两者之间加入1.6kb的绝缘子。经PCR检测和限制性酶切分析及序列测定均证实了载体的正确性。在转染HEK293细胞后,运用荧光显微镜和流式细胞技术证实了该载体的调控能力。没有RU486时,几乎没有红色荧光蛋白的表达,而加入诱导剂RU486后,最高可以实现红色荧光蛋白的40余倍的表达。实验结果表明构建的可经RU486诱导的新型真核表达载体可以实现对目的基因的表达时间和表达水平的调控,为进一步的基因调控研究和和基因治疗提供了良好的工具。

软土层对地铁狭长深基坑地表沉降的影响研究

随着我国城市轨道交通的快速发展,地铁地下车站狭长深基坑工程越来越多,尤其是长江中下游和沿海地区城市的狭长深基坑大多开挖于深厚软土场地,对该类深基坑的施工安全造成了严重的威胁和工程事故。鉴于此,本文基于Midas商用软件,建立了土体-围护结构相互作用体系的三维有限元分析模型,通过现场实测与数值计算结果的对比分析,验证了数值分析模型的可行性。通过改变软土层厚度、埋深和开挖深度等不同组合的108个计算工况,系统分析了软土层厚度和埋深变化对地铁狭长深基坑周围地表沉降的影响规律。结果表明:当软土层主要位于基坑底面以上侧向地基中时,软土层厚度变化对基坑周围地表沉降影响较大;地铁狭长深基坑侧向地基中软土层埋深增大时基坑周围地表沉降先变大后变小,且变化转折点为软土层顶层埋深高于基坑底面以上的一定范围内;当软土层埋深较大且较厚时,开挖深度对地表沉降的影响有增速的趋势,即当基坑开挖深度较深时软土层的影响更为明显。同时,根据数值计算结果,给出了软土层厚度和埋深等与地表沉降的统计关系。

Mechanisms behind the surprising observation of supra-thermal ions in NIF’s fusion burning plasmas

Although ignition had been achieved at the National Ignition Facility(NIF),recent observations of the experiments indicate novel physics that beyond theoretical predictions emerge,e.g.,the neutron analysis of experiments has revealed deviations from the Maxwellian distributions in ion relative kinetic energies of burning plasmas,with the surprising emergence of supra-thermal deuterium and tritium(DT)ions that fall outside the predictions of macroscopic statistical hydrodynamic *** our newly developed hybrid-particle-in-cell code,incorporating the newly-proposed model of large-angle collisions,we infer that that this could be attributed to the increased significance of large-angle collisions among DT ions andα-particles in the burning *** and unprecedented kinetic investigations into the implications of large-angle collisions in the burning plasma have yielded several key findings,including an ignition moment promotion by~10 ps,the presence of supra-thermal ions below an energy threshold,and approximately twice the expected deposition ofα-particles *** rationality of our findings is confirmed through the congruency between the neutron spectral moment analyses conducted by the NIF and our kinetic simulations,both highlighting progressively widening disparities between neutron spectral moment analyses and hydrodynamics predictions,which becomes more pronounced as the yield *** kinetic simulations with large-angle collisions not only provide novel insights for experiment interpretation but also open new research opportunities for the largely unexplored regime of the nuclear burning plasmas,which are distinguished by their exceptionally high energy densities and hold immense potential for illuminating the intricate physics that underpins the evolution of the early universe.

以实际工程为背景,运用MIDAS GTS NX工程型和ABAQUS学术型软件模拟深基坑开挖过程,将模拟与实测数据进行对比,总结MIDAS GTS NX与ABAQUS软件在模拟深基坑开挖过程中的特点。通过计算结果的对比分析发现:MIDAS GTS NX软件模拟得到的围护结构侧方位移与实测数据相比明显偏大;使用ABAQUS软件中修正剑桥模型预测深基坑侧方地基的变形时,与MIDAS GTS NX软件计算结果相比更佳;ABAQUS软件计算得到的围护结构最大侧方位移的位置与MIDAS GTS NX软件相比更高,更接近实测情况;采用MIDAS GTS NX和ABAQUS软件模拟深基坑侧方已建地铁隧道的变形时得出的位移变化趋势相似,ABAQUS软件得到的隧道衬砌变形最大值与MIDAS GTS NX软件计算结果相比更小,ABAQUS软件得到的隧道受深基坑开挖影响范围与MIDAS GTS NX软件计算结果相比范围偏大。

PbrMYB4,a R2R3-MYB protein,regulates pear stone cell lignification through activation of lignin biosynthesis genes

Pear(Pyrus bretschneideri)fruit stone cells are primarily composed of lignin and have strongly lignified cell *** presence of stone cells has a negative influence on fruit texture and taste,and thus the reduction of stone cell content in pear fruit is a key goal of breeding ***,research into the key transcription factors and regulatory networks associated with pear fruit stone cell formation have been *** here used a combination of co-expression network and expression quantitative trait locus(eQTL)analyses in 206 pear cultivars with different stone cell contents to identify relevant genes;these analyses uncovered the gene PbrMYB4,a R2R3 MYB transcription factor *** was a strong positive correlation between relative PbrMYB4 expression levels in the fruit flesh and stone cell/lignin *** of PbrMYB4 significantly increased the lignin contents,whereas silencing of PbrMYB4 had the opposite effect,decreasing the contents of ***4 overexpression in pear calli significantly promoted lignin *** Arabidopsis thaliana,PbrMYB4 overexpression resulted in increasing lignin deposition,cell wall thickness of vessels and xylary fiber,and accelerating expression level of lignin biosynthetic ***4 was found to activate 4-Coumarate:Coenzyme A Ligase(Pbr4CL1)by binding to AC-I elements in the promoter regions,as demonstrated with dual-luciferase reporter assays and a yeast one-hybrid *** results demonstrated that PbrMYB4 positively regulated lignin biosynthesis in pear fruit stone cells by activating lignin biosynthesis *** study improves our understanding of the gene regulatory networks associated with stone cell formation in pear fruit,providing guidance for molecular breeding of pear varieties with low stone cell content.

Oxygen atmosphere enhances ball milling remediation of petroleum-contaminated soil and reuse as adsorptive/catalytic materials for wastewater treatment

Turbulent Agglomeration of Microparticles in a Cylinder Wake Flow Using LES-DEM:Focusing on the Effect of the Reynolds Number
