PPP1R14A is Associated with Immunotherapy Resistance in Head and Neck Squamous Cell Carcinoma Identified by Single-Cell and Bulk RNA-Sequencing

Objective To identify nivolumab resistance-related genes in patients with head and neck squamous cell carcinoma(HNSCC)using single-cell and bulk RNA-sequencing *** The single-cell and bulk RNA-sequencing data downloaded from the Gene Expression Omnibus database were analyzed to screen out differentially expressed genes(DEGs)between nivolumab resistant and nivolumab sensitive patients using R *** Least Absolute Shrinkage Selection Operator(LASSO)regression and Recursive Feature Elimination(RFE)algorithm were performed to identify key genes associated with nivolumab *** enrichment of DEGs was analyzed with Gene Ontology and Kyoto Encyclopedia of Genes and Genomes *** relationships of key genes with immune cell infiltration,differentation trajectory,dynamic gene expression profiles,and ligand-receptor interaction were *** We found 83 *** were mainly enriched in T-cell differentiation,PD-1 and PD-L1 checkpoint,and T-cell receptor *** six key genes identified using machine learning algorithms,only PPP1R14A gene was differentially expressed between the nivolumab resistant and nivolumab sensitive groups both before and after immunotherapy(P0,P<0.05),lower differentiation of infiltrated immune cells(P<0.05),and a higher degree of interaction between HLA and CD4(P<0.05).Conclusions PPP1R14A gene is closely associated with resistance to nivolumab in HNSCC ***,PPP1R14A may be a target to ameliorate nivolumab resistance of HNSCC patients.

Highly productive estuaries facilitate intense decomposition of dissolved organic matter(DOM) as a carbon ***,the specific impacts of typhoons on DOM decomposition in eutrophic bays remain *** address this issue,we investigated the spectral characteristics of DOM before and after Typhoon Ewiniar in Zhanjiang B ay,a eutrophic semi-enclosed bay in the northwestern South China *** results revealed that intense microbial decomposition of DOM occurred during the pre-typhoon period because high nutrient inputs facilitated the mobilization of DOM in the ***,the intrusion of external seawater induced by the typhoon diluted the nutrient levels in Zhanjiang B ay,reducing the impact of microbial decomposition on DOM during the post-typhoon perio ***,the net addition of DOM occurred in Zhanjiang Bay during the post-typhoon period,possibly because of the decomposition of particulate organic matter(POM) and desorption of particulate *** addition,an increase in apparent oxygen utilization,a decrease in DO saturation and the reduced level of Chl a indicated that organic matter(OM) decomposition was enhanced and OM decomposition shifted to POM decomposition in Zhanjiang Bay after the ***,our study highlighted the shift in the intense OM decomposition from DOM to POM decomposition before and after typhoons in eutrophic bays,providing new insights into the response of typhoons to biogeo chemistry.

Automatic velocity picking based on optimal key points tracking algorithm

Picking velocities from semblances manually is laborious and necessitates experience. Although various methods for automatic velocity picking have been developed, there remains a challenge in efficiently incorporating information from nearby gathers to ensure picked velocity aligns with seismic horizons while also improving picking accuracy. The conventional method of velocity picking from a semblance volume is computationally demanding, highlighting a need for a more efficient strategy. In this study, we introduce a novel method for automatic velocity picking based on multi-object tracking. This dynamic tracking process across different semblance panels can integrate information from nearby gathers effectively while maintaining computational efficiency. First, we employ accelerated density clustering on the velocity spectrum to discern cluster centers without the requirement for prior knowledge regarding the number of clusters. These cluster centers embody the maximum likelihood velocities of the main subsurface structures. Second, our proposed method tracks key points within the semblance volume. Kalman filter is adopted to adjust the tracking process, followed by interpolation on these tracked points to construct the final velocity model. Our synthetic data example demonstrates that our proposed algorithm can effectively rectify the picking errors of the clustering algorithm. We further compare the performances of the clustering method(CM), the proposed tracking method(TM), and the variational method(VM) on a field dataset from the Gulf of Mexico. The results attest that our method offers superior accuracy than CM, achieves comparable accuracy with VM, and benefits from a reduced computational cost.

Aquatic vegetation is a vital component of natural river ecosystems,playing a crucial role in maintaining ecological balance,providing habitat and improving water ***,the presence of vegetation results in increased resistance in vegetated channels compared with non-vegetated channels,rendering traditional sediment movement predictions inadequate for the ***,the concept of a vegetation influence factor,denoted by CDah,has been proposed by previous researchers to represent the effect of vegetation on sediment movement in *** this study,we focus on exploring the vegetation resistance coefficient(CD)among the vegetation influence factors,evaluating two different calculation methods for vegetation resistance coefficient,and presenting two expressions through genetic algorithm analysis to predict the incipient flow velocity of sediment in vegetated *** predicted values from the new formulae show excellent agreement with measured data,highlighting the high accuracy of the proposed methods in predicting the incipient flow velocity of *** results provide a solid theoretical basis for understanding the influence of aquatic vegetation on sediment particle movement.

A review of control strategies for proton exchange membrane(PEM)fuel cells and water electrolysers:From automation to autonomy

Proton exchange membrane (PEM) based electrochemical systems have the capability to operate in fuel cell (PEMFC) and water electrolyser (PEMWE) modes, enabling efficient hydrogen energy utilisation and green hydrogen production. In addition to the essential cell stacks, the system of PEMFC or PEMWE consists of four sub-systems for managing gas supply, power, thermal, and water, respectively. Due to the system's complexity, even a small fluctuation in a certain sub-system can result in an unexpected response, leading to a reduced performance and stability. To improve the system's robustness and responsiveness, considerable efforts have been dedicated to developing advanced control strategies. This paper comprehensively reviews various control strategies proposed in literature, revealing that traditional control methods are widely employed in PEMFC and PEMWE due to their simplicity, yet they suffer from limitations in accuracy. Conversely, advanced control methods offer high accuracy but are hindered by poor dynamic performance. This paper highlights the recent advancements in control strategies incorporating machine learning algorithms. Additionally, the paper provides a perspective on the future development of control strategies, suggesting that hybrid control methods should be used for future research to leverage the strength of both sides. Notably, it emphasises the role of artificial intelligence (AI) in advancing control strategies, demonstrating its significant potential in facilitating the transition from automation to autonomy.

Experimental investigation on pyrolysis products and pore structure characteristics of organic-rich shale heated by supercritical carbon dioxide

The efficient pyrolysis and conversion of organic matter in organic-rich shale,as well as the effective recovery of pyrolysis shale oil and gas,play a vital role in alleviating energy *** state of carbon dioxide（CO2） in the pyrolysis environment of shale reservoirs is the supercritical *** unique supercritical fluid properties not only effectively heat organic matter,displace pyrolysis products and change shale pore structure,but also achieve carbon storage to a certain *** samples were made into powder and three sizes of cores,and nitrogen（N2） and supercritical carbon dioxide（ScCO2）pyrolysis experiments were performed at different final pyrolysis *** properties and mineral characteristics of the pyrolysis products were studied based on gas chromatography analysis,Xray diffraction tests,and mass spectrometry ***,the pore structure characteristics at different regions of cores before and after pyrolysis were analyzed using N2adsorption tests to clarify the impact of fracturing degree on the pyrolysis *** results indicate that the optimal pyrolysis temperature of Longkou shale is about 430℃.Compared with N2,the oil yield of ScCO2pyrolysis is *** pyrolysis oil obtained by ScCO2extraction has more intermediate fractions and higher relative molecular *** ScCO2can effectively improve the pore diameter of shale and its effect is better than that of *** micropores are produced in shale after pyrolysis,and the macropores only are generated in ScCO2pyrolysis environments with temperatures greater than 430℃.The pore structure has different development characteristics at different pyrolysis temperatures,which are mainly affected by the pressure holding of volatile matter and products *** to the surface of the core,the pore development effect inside the core is *** the decrease in core size,the pore diameter,specific surface area,and pore volume of cores all increase after pyrolysis.

Engineering high-performance dielectric chiral shells with enhanced chiral fields for sensitive chiral biosensor

Chiral metamaterials have been a topic of significant research interest in recent years due to their potential for various applications in nanophotonic devices and chiral ***,the intrinsic Ohmic loss in surface plasmonic resonance has limited their practical use,resulting in large light dissipation and weak chiroptical ***,we report on the development of high-performance dielectric chiral shells(DCS)through a two-step Si deposition process on a self-assembled microsphere *** form DCS sample completely oyercomes the cancelation effect originated from the disorder property of the micro-sphere monolayer in macroscale,and at a wavelength of approximately 710 nm,the measured optimal chiral signal(g-factor)and transmittance can reach up to 0.7 and 0.3,*** strong chiroptical effect comes from the asymmetric circular displacement currents(i.e.,magnetic modes)enabled by the specific shell *** chiral shell geometry,electromagnetic properties,sensor sensitivity of chiral molecules and figure of merit are systematically *** DCSs demonstrate highly sensitive detection of chiral biomolecules owing to their easily accessible geometry and enhanced uniform chiral field.

Demonstration of a small‐scale power generator using supercritical CO_(2)

The supercritical CO_(2)(sCO_(2))power cycle could improve efficiencies for a wide range of thermal power *** sCO_(2)turbine generator plays an important role in the sCO_(2)power cycle by directly converting thermal energy into mechanical work and electric *** operation of the generator encounters challenges,including high temperature,high pressure,high rotational speed,and other engineering problems,such as *** studies of sCO_(2)turbines are insufficient because of the significant difficulties in turbine manufacturing and system *** most experimental investigations that primarily focus on 100 kW‐or MW‐scale power generation systems,we consider,for the first time,a small‐scale power generator using sCO_(2).A partial admission axial turbine was designed and manufactured with a rated rotational speed of 40,000 rpm,and a CO_(2)transcritical power cycle test loop was constructed to validate the performance of our manufactured generator.A resistant gas was proposed in the constructed turbine expander to solve the leakage *** dynamic and steady performances were *** results indicated that a peak electric power of 11.55 kW was achieved at 29,369 *** maximum total efficiency of the turbo‐generator was 58.98%,which was affected by both the turbine rotational speed and pressure ratio,according to the proposed performance map.

Chiral multiple-resonance thermally activated delayed fluorescence materials based on chiral spiro-axis skeleton for efficient circularly polarized electroluminescence

Chiral luminescence materials have potential applications in the field of three-dimensional displays due to their circularly polarized luminescence(CPL)***,the further development of circularly polarized organic light-emitting diodes(CP-OLEDs)needs to meet the requirements of high efficiency,high color purity,low cost,and high dissymmetry factor(gPLor gEL),chiral multiple resonance thermally activated delayed fluorescence(MR-TADF)materials are considered as candidates in these ***,based on a pair of chiral spirofluorene precursors,two pairs of high-performance chiral MR-TADF emitters((R/S)-p-Spiro-DtBuCzB and(R/S)-m-Spiro-DtBuCzB)are developed,which exhibit strong emissions peaking at 491 and 502 nm in toluene with full-width at half-maximum values of 25 and 33 nm,*** addition,small singlet–triplet energy gaps of 0.15 and 0.10 eV with high absolute photoluminescence efficiencies of 95.0%and 96.7%are observed for p-Spiro-DtBuCzB and m-Spiro-DtBuCzB molecules,*** based on p-Spiro-DtBuCzB and m-Spiro-DtBuCzB display high maximum external quantum efficiencies of 29.6%and 33.8%,*** importantly,CP-OLEDs present symmetric circularly polarized electroluminescence spectra with|gEL|factors of 3.36×10^(-4)and 7.66×10^(-4)for devices based on(R/S)-p-Spiro-DtBuCzB and(R/S)-m-Spiro-DtBuCzB enantiomers,respectively.
