A novel type of glucose sensor was fabricated based on a glucose oxidase(GOD)-N,N-dimethtylformamide(DMF)-[BMIm][BF4] composites modified three-dimensional ordered macroporous(3DOM) gold film electrode.The immobilized GOD exhibits a pair of well-defined reversible peaks in 50 mM pH 7.0 phosphate buffer solutions(PBS),which could be attributed to the redox of flavin adenine dinucleotide(FAD) in GOD.The research results show that ionic liquid([BMIm][BF4]),DMF and 3DOM gold film are crucial for GOD to exhibit a pair of stable and reversible peaks.It is believed that the large active area of 3DOM gold film can increase the amount of immobilized GOD.Simultaneously,the application of IL enhances the stability of GOD and facilitates the electron transfer between GOD and the electrode.The synergetic effect of DMF can help the GOD to maintain its bioactivity better.GOD immobilized on the electrode exhibits the favorable electrocatalytic property to glucose,and the prepared sensor has a linear range from 10 to 125 nM with a detection limit of 3.3 nM at a signal-to-noise ratio of 3σ.The apparent Km(Michaelis-Menten constant) for the enzymatic reaction is 0.018 mM.

Trust Evaluation Model Based on PSO and LSTM for Huge Information Environments

Due to the challenge of increasing data volume, the traditional trust model is unable to manage data with high efficiency and effectively extract useful information hidden in big data. To fully utilize big data and combine machine learning with trust evaluation, a trust evaluation model based on Long short-term memory(LSTM) is presented. The powerful learning ability,expressive ability and dynamic timing of LSTM can be applied to study data while avoiding the vanishing and exploding gradient phenomena of traditional Recurrent neural networks(RNNs) to ensure that the model can learn sequences of random length and provide accurate trust evaluation. Targeting the performance instability caused by the LSTM model’s random initialization of weights and thresholds, Particle swarm optimization(PSO), one of the intelligent algorithms, is introduced to find global optimal initial weights and thresholds.Experiments proved that the trust model proposed in this paper has high accuracy and contributes a new idea for trust evaluation in big data environments.

为探明盐酸多西环素治疗黄颡鱼维氏气单胞菌感染的防耐药给药方案,体外测定盐酸多西环素对维氏气单胞菌的最小抑菌质量浓度、最小杀菌质量浓度、防耐药突变质量浓度、耐药选择窗以及抗菌后效应等药效学参数,并根据国标渔药盐酸多西环素的使用方案,在水温(20±2)℃下,体质量(200±50)g的黄颡鱼口灌20 mg/kg的盐酸多西环素,给药后0.5、1、2、4、8、12、24、48、78 h取血、肌肉、脑组织、肝胰脏、肾脏样品,利用高效液相色谱法测定和分析盐酸多西环素在黄颡鱼体内5个组织的药代动力学参数。试验结果显示,盐酸多西环素对维氏气单胞菌的最小抑菌质量浓度为0.5μg/mL,最小杀菌质量浓度为1.0μg/mL,防耐药突变质量浓度为4.0μg/mL,耐药选择窗为0.5~4.0μg/mL,抗菌后效应分别为(0.95±0.24)、(0.79±0.58)、(1.92±0.71)h。黄颡鱼口灌20 mg/kg的盐酸多西环素后,血清、肝胰脏、肾脏、肌肉、脑中的曲线下面积(0~78 h)与最小抑菌质量浓度的比值(AUC0~78h/MIC)分别为75.5580、686.5486、557.2362、736.9682、234.9658,最大药物质量浓度与最小抑菌质量浓度的比值(ρmax/MIC)为2.4908、18.6652、15.3044、24.1590、10.9018。根据浓度依赖型药物曲线下面积(0~t)与最小抑菌质量浓度的比值(AUC0~t/MIC)>125、最大药物质量浓度与最小抑菌质量浓度的比值(ρmax/MIC)>8的有效标准,确定盐酸多西环素用药方案为20 mg/kg,每日1次。依据肌肉中的药物残留动力学数据,确定休药期至少11.2 d。综合药代动力学参数和药效学参数,试验结果可为利用国标渔药盐酸多西环素有效治疗黄颡鱼维氏气单胞菌感染提供理论依据。

农业机械化与农村劳动力转移是农民收入增长的两个重要影响因素,通过对农业机械化、农村劳动力转移与农民收入增长关系的研究,可以为“三农”问题和缩小城乡差距问题提供参考与借鉴。在现有研究基础上,使用VAR模型进行多变量分析。认为农村劳动力转移、农业机械化与农民收入增长之间存在长期均衡关系,农村劳动力转移与农民收入增长以及农业机械化与农民收入增长之间存在单向格兰杰因果关系。基于此,需要制定农村劳动力转移支持政策,发挥农村劳动力转移的示范效应,加强农业机械的推广与运用,落实农业机械化扶持政策。

我国是目前全球最大的铁素资源消费国,当前及未来一段时间,我国铁素资源需求仍将保持高位。目前,我国国内铁矿石生产不足,权益矿建设进展缓慢,废钢资源加工配送体系有待完善,存在铁素资源自给率低、铁素资源供给结构不均衡、铁素资源定价话语权缺失等问题。国家积极推动“基石计划”,加快钢铁行业铁素资源安全保障体系建设,切实维护我国产业链供应链安全稳定可控,未来需加大国内铁矿的开发力度,在防范风险前提下加快海外优质权益矿及产业链延伸开发,加强废钢及直接还原铁等替代铁矿石资源的利用。从行业规范管理、矿业权整合重组、矿业资本市场建设、矿业产业链延伸开发、铁矿石替代等方面,提出提高我国铁素资源自给率的对策建议。

目的在平痤散中药处方基础上,研制平痤滴丸,并对比研究平痤散和平痤滴丸对痤疮的作用。方法以综合评分为评价指标,考察因素为药物与基质质量比、药液温度、冷凝温度,采用Box-Behnken响应面法,对平痤滴丸的制备工艺进行优化;考察平痤散和平痤滴丸对小鼠激素水平、皮脂腺斑组织的影响,对耳肿胀的抗炎作用、对金色葡萄球菌和痤疮丙酸杆菌的抑制作用。结果最优滴丸制备工艺料液比为5∶1,药液温度为80℃,冷凝温度为0℃;平痤滴丸组体内激素水平睾酮(T)值、雌二醇(E2)值、T/E2值、小鼠耳肿胀程度及抑菌作用,均与阳性药组差异无统计学意义(P>0.05),且优于平痤散组。结论平痤滴丸组治疗痤疮效果显著,优于平痤散组,能够有效调节小鼠体内激素水平、减轻小鼠耳肿胀程度、抑制痤疮致病菌。

为了建立一种灵敏度高、特异性强并且适合现场诊断的大口黑鲈弹状病毒(Micropterus salmoides rhabdovirus,MSRV)检测方法,研究基于CRISPR-Cas13a系统并结合多酶恒温核酸快速扩增(MIRA)技术建立了一种MSRV的检测方法。实验对MSRV序列进行多重序列比对分析后,针对MSRV衣壳蛋白(CP)基因的特异性区域设计两个靶点,并通过体外转录成特异性的crRNA,同时设计合成MIRA引物序列实现目标序列的等温扩增,最后结合Cas13a蛋白、crRNA、恒温扩增体系构建检测体系,并从高效crRNA的选择、反应温度、ssRNA报告探针浓度和Cas13a与crRNA的浓度比四个方面优化了反应体系,并采用最优检测体系对大口黑鲈样本进行检测验证。结果显示,在20μL检测体系中加入200 nmol/L Cas13a、100 nmol/L crRNA1、100 nmol/L crRNA2及500 nmol/L ssRNA报告探针,在37℃的情况下能够获得最佳的检测效果。并且该检测体系可以检测到102 fM的MSRV病毒,具有良好的特异性和灵敏性,此外检测结果可直接通过紫外灯照射直接观察。研究基于CRISPR/Cas13a系统结合等温扩增MIRA技术建立的MSRV检测方法,可在室温下且不需要昂贵的仪器进行MSRV病毒检测,检测结果可以肉眼直接观察,能在多种场合下使用,研究结果为检测MSRV提供了有效方法。

The removal of a failed implant with high torque causes significant damage to the surrounding tissue,compromising bone regeneration and subsequent osseointegration in the defect area.Here,we report a case of carrier screw fracture followed by immediate implant removal,bone grafting and delayed reimplantation.A dental implant with a fractured central carrier screw was removed using the bur-forceps technique.The resulting three-wall bone defect was filled with granular surface demineralized freeze-dried bone allograft(SD-FDBA).Cone-beam computerized tomography was performed at 1week,6months and 15months postoperatively and standardized for quantitative evaluation.The alveolar bone width and height at 15months post-surgery were about 91%of the original values,with a slightly lower bone density,calculated using the gray value ratio.The graft site was reopened and was found to be completely healed with dense and vascularized bone along with some residual bone graft.Reimplantation followed by restoration was performed 8months later.The quality of regenerated bone following SD-FDBA grafting was adequate for osseointegration and longterm implant success.The excellent osteogenic properties of SD-FDBA are attributed to its human origin,cortical bone-like structure,partly demineralized surfaces and bone morphogenetic protein-2-containing nature.Further investigation with more cases and longer follow-up was required to confirm the final clinical effect.

Abstract Mn-based layered oxides are among the most promising cathode materials for sodium-ion batteries owing to the advantages of abundance,environmenta friendliness,low cost and high specific capacity.P2 and O'3 are two representative structures of Mn-based layered oxides.However,the P2 structure containing insufficien Na generally exhibits low initial charge capacity,while O'3structure with sufficient Na delivers high initial charge capacity but poor cycle stability.This study prepared a multitude of NaxMnO2（x=0.7,0.8,0.9） cathode materials with varying P2/O'3 ratios and further investigated their electrochemical performances.The optimized Na0.8MnO2comprising 69.9 wt%O'3 and 30.1 wt% P2 phase,exhibited relatively balanced specific capacity,Coulombic efficiency and cycle stability.Specifically,it achieved a high specific capacity of 128.9 mAh·g-1with an initia Coulombic efficiency of 98.2%in half-cell configuration The Na0.8MnO2// hard carbon full cell also achieved a high specific capacity of 126.7 mAh·g-1with an initia Coulombic efficiency of 98.9%.Moreover,the capacity fading mechanism was revealed by combining in-situ and ex-situ X-ray diffraction.The findings of this study provide theoretical guidance for further modification design of Mnbased layered cathodes.

Background:Hepatocellular carcinoma(HCC)has a poor long-term prognosis.The competition of circular RNAs(circRNAs)with endogenous RNA is a novel tool for predicting HCC prognosis.Based on the alterations of circRNA regulatory networks,the analysis of gene modules related to HCC is feasible.Methods:Multiple expression datasets and RNA element targeting prediction tools were used to construct a circRNA-microRNA-mRNA network in HCC.Gene function,pathway,and protein interaction analyses were performed for the differentially expressed genes(DEGs)in this regulatory network.In the proteinprotein interaction network,hub genes were identified and subjected to regression analysis,producing an optimized four-gene signature for prognostic risk stratification in HCC patients.Anti-HCC drugs were excavated by assessing the DEGs between the low-and high-risk groups.A circRNA-microRNA-hub gene subnetwork was constructed,in which three hallmark genes,KIF4A,CCNA2,and PBK,were subjected to functional enrichment analysis.Results:A four-gene signature(KIF4A,CCNA2,PBK,and ZWINT)that effectively estimated the overall survival and aided in prognostic risk assessment in the The Cancer Genome Atlas(TCGA)cohort and International Cancer Genome Consortium(ICGC)cohort was developed.CDK inhibitors,PI3K inhibitors,HDAC inhibitors,and EGFR inhibitors were predicted as four potential mechanisms of drug action(MOA)in high-risk HCC patients.Subsequent analysis has revealed that PBK,CCNA2,and KIF4A play a crucial role in regulating the tumor microenvironment by promoting immune cell invasion,regulating microsatellite instability(MSI),and exerting an impact on HCC progression.Conclusions:The present study highlights the role of the circRNA-related regulatory network,identifies a four-gene prognostic signature and biomarkers,and further identifies novel therapy for HCC.
