Antibacterial mechanism of kojic acid and tea polyphenols against Escherichia coli O157:H7 through transcriptomic analysis

Escherichia coli O157:H7 is one of the major foodborne pathogenic bacterial that cause infectious diseases in *** previous found that a combination of kojic acid and tea polyphenols exhibited better activity against *** O157:H7 than using either *** study aimed to explore responses underlying the antibacterial mechanisms of kojic acid and tea polyphenols from the gene *** functional enrichment analysis by comparing kojic acid and tea polyphenols individually or synergistically against *** O157:H7 found that acid resistance systems in kojic acid were activated,and the cell membrane and genomic DNA were destructed in the cells,resulting in“oxygen starvation”.The oxidative stress response triggered by tea polyphenols inhibited both sulfur uptake and the synthesis of ATP,which affected the bacteria's life metabolic ***,we found that kojic acid combined with tea polyphenols hindered the uptake of iron that played an essential role in the synthesis of DNA,respiration,tricarboxylic acid *** results suggested that the iron uptake pathways may represent a novel approach for kojic acid and tea polyphenols synergistically against *** O157:H7 and provided a theoretical basis for bacterial pathogen control in the food industry.

Ribophagy, the cellular process wherein ribosomes are selectively self-digested through autophagy,plays a pivotal role in maintaining ribosome turnover. Understanding the molecular regulatory mechanisms governing ribophagy is pivotal to uncover its significance. Consequently, the establishment of methods for detecting ribophagy becomes important. In this protocol, we have optimized, enriched,and advanced existing ribophagy detection techniques, including immunoblotting, fluorescence microscopy, and transmission electron microscopy(TEM), to precisely monitor and quantify ribophagic events. Particularly noteworthy is the introduction of TEM technology for yeast ribophagy detection. In summary, the delineated methods are applicable for detecting ribophagy in both yeast and mammals,laying a solid foundation for further exploring the physiological importance of ribophagy and its potential implications in diverse cellular environments.

A REAL-TIME MATHEMATICAL MODEL FOR TWO DIMENSIONAL TIDAL FLOW AND WATER QUALITY

A real-time mathematical model for two dimensional tidal flow and water quality is presented in this paper. The control-volume-based-finite-difference method and the 'power interpolation distribution' advocated by Patankar [4] have been employed, and new boundary condition for tidal flow is recommended. The model is un- conditionally stable and convergent, and able to deal with irregular estuarine topography and movable boundary problems. Practical application of the model is illustrated by an example for the Swatou Bay. A fair agreement be- tween the values measured and computed demonstrates the validity of the model developed.

Preliminary Response of Soil Fauna to Simulated N Deposition in Three Typical Subtropical Forests

A field-scale experiment arranged in a complete randomized block design with three N addition treatments including a control (no addition of N), a low N (5 g m-2 year-1), and a medium N (10 g m-2 year-1) was performed in each of the three typical forests, a pine (Pinus massoniana Lamb.) forest (PF), a pine-broadleaf mixed forest (MF) and a mature monsoon evergreen broadleaf forest (MEBF), of the Dinghushan Biosphere Reserve in subtropical china to study the response of soil fauna community to additions of N. Higher NH4+ and NO3- concentrations and a lower soil pH occurred in the medium N treatment of MEBF, whereas the NO3- concentration was the lowest in PF after the additions of N. The response of the density, group abundance and diversity index of soil fauna to addition of N varied with the forest type, and all these variables decreased with increasing N under MEBF but the trend was opposite under PF. The N treatments had no significant effects on these variables under MF. Compared with the control plots, the medium N treatment had significant negative effect on soil fauna under MEBF. The group abundance of soil fauna increased significantly with additions of higher N rates under PF. These results suggested that the response of soil fauna to N deposition varied with the forest type and N deposition rate, and soil N status is one of the important factors affecting the response of soil fauna to N deposition.

Regional patterns of particulate spectral absorption in the Pearl River estuary

Spectral absorption coefficients of the total particulate, ap(l ), nonalgal particulate, ad(l ), and phyto-plankton pigment, aph(l ), in the Pearl River estuary and its vicinity waters were determined using the quantitative filter technique. The particulate absorption ap(443) ranged from 0.04 to 1.82 m-1, with the corresponding aph(443) ranging from 0.016 to 0.484 m-1. Two typical spectral patterns are found for the total particulate absorption. For the first typi-cal spectral pattern, the total particulate absorption spectra are similar to that of nonalgal particulate, with values of absorption coefficient decreasing with wavelength. In con-trast, for the second spectral pattern the spectral absorptions by total particulate are very similar to that of phytoplankton pigment. The spectral dependency of absorption by nonalgal particulate follows an exponential increase toward short wavelengths, with an average slope of 0.012±0.002 nm-1. The nonalgal absorption and the fraction of the nonalgal particulate absorption to the total particulate absorption exhibit a distinct trend of decreasing with salinity of the sur-face water. Phytoplankton pigment absorption exhibits a clear trend of increasing nonlinearly with chlorophyll a con-centration. The relationships between the phytoplankton pigment absorption and chlorophyll a concentration can be described by power law, with the determination coefficient r2 of 0.82. But only weak relationships between ap(l ) and chlorophyll a concentration are observed, with the determination coefficient r2 of 0.42. The relatively large scatter around ap(443) versus chl-a relationship would be attributed to the effects of loading of the nonalgal particulate absorption. Our analysis indicated that such relationships similar to that for Case Ⅰ waters can be applicable to optically complex Case Ⅱ waters if the effects caused by nonalgal are corrected. The Formation of Yanshanian Granitic Magma in Dabie Mountains:Dehydration or Aquifer Melting-with Tiantangzhai and Jiuzihe Granites as Examples

There are large-areas of late Yanshanian granites in the Dabie Orogen, but the formation mode of the relevant granitic magma is still under discussion. Taking the typical Yanshanian granites in the North Dabie Block - The TG and JG as examples, the Writers endeavor to discuss contain aspects of the formation mode of the granitic magma by studying the characteristics of the chemical compositions of biotite and amphibole of the granites and petrological features of enclaves in the intrusions, in the light of recent results of experimental petrology on dehydration melting and aquifer melting. It is suggested that: (1) Some biotites in the Tiantangzhai and the JGs and their enclaves were residual minerals; (2) Some of the amphiboles could be new-born minerals, and others, residual ones. (3) the banded bodies in granites were residuals of anatexis. (4) The TG and JG were formed by aquifer melting, and the South Dabie Block could be the important source region of water.

LITHOSPHERE-ASTHENOSPHERE INTERACTION: A COMPARATIVE STUDY ON CENOZOIC AND MESOZOIC BASALTS AROUND BOHAI AREA

Mesozoic and Cenozoic basalitc rnagmatism within the basins around Bohai area displays temporal variations in petrochemistry,trace element and isotope geochmistry. Two stages of evolution are recognized. The preextension magmatism (134 - 91 Ma,Jiyang basin) is characterised by small volumes of alkaline basalts and potassic lamprophyres. These rocks are interpreted as results of very small degrees of partial melting of the lithosphere and volatile-rich metasome within the lithosphere induced by initial thermal perturbation. The second stage (74- 27 Ma) coincides with the lithospheric extension. It, is noted that the alkalinity and Dea of basalts increased systematically from 74 to 27 Ma. Major element chemistry,trace element modelling antl experimental data indicate that there is an increase in depth of melting and a decrease in degree of partial melting from 74 to 27 Ma. Such a temporal change in source character and in the depth of origin is best understood when magmatism is considered within the framework of a lithosphere-asthenos phere interaction model where melting is initiated by discrete thermal events related lithospheric extension.

The Late Permian Emeishan large igneous province (ELIP), with an areal extent of over 500,000 km, at the western margin of Yangtze craton, is increasingly regarded as the result of the impingement of a mantle plume onto the lithosphere. However, petrogenesis of the continental flood basalt remains controversial. The best-exposed lava succession in the western ELIP is studied in order to further constrain their petrogenesis and plume-lithosphere interaction. The basaltic lava flows of the ELIP are geochemically classified into low-Ti (LT) and high-Ti (HT) types. The LT type lavas exhibit low Ti/Y (500), εNd(t) (-1.17～0.43 ) but lower Mg# (31～53) and (Sr/Sr) (0.705- 0706). The LT basalts can be further subdivided into: LT1 and LT2. LT1 lavas exhibit relatively higher Mg# (51～67) and (Sr/Sr) ratio (0.706- 0.707), lower Nb/La ratio (1.1;εNd(t)= -1.17～0.43). Detailed stratigraphic work indicates that there was a temporal progression from LT1 to LT2 to HT-type magmas. This compositional shift cannot be explained simply in terms of a declining extent of crustal contamination of a mantle-derived melt with time. Instead, it seems that the LT and HT type magmas originated from distinct mantle sources and parental magmas. Geochemical features of the early stage LT1 lavas are indicative of a significant contribution from the enriched continental lithosphere mantle, whereas the compositional shift from LT1 to LT2 reveals a trend from predominantly shallower lithospheric mantle to deeper mantle with time. Late stage HT magma formed from a deeper mantle source that may possibly be a mantle plume.
