The small-angle neutron scattering(SANS)instrument,one of the first three instruments of the china Spallation Neutron Source(CSNS),is designed to probe the microscopic and mesoscopic structures of materials in the scale range 1–100 nm.A large-area ^(3)He tube array detector has been constructed and operates at the CSNS SANS instrument since August *** consists of 12. linear position-sensitive detector tubes,each 1 m in length and 8 mm in diameter,and filled with ^(3)He gas at 2. bar to obtain a high detection *** ^(3)He tubes were divided into ten modules,providing an overall area of 1000 mm×102. mm with a high count rate *** each tube is installed independently,the detector can be quickly repaired in situ by replacing damaged *** reduce air scattering,the SANS detector must operate in a vacuum environment(0.1 mbar).An all-metal sealing technique was adopted to avoid high-voltage breakdown by ensuring a high-voltage connection and an electronic system working in an atmospheric environment.A position resolution of 7.8±0.1 mm(full width at maximum)is measured along the length of the tubes,with a high detection efficiency of 81±2. at *** over the past four years,the detector appears to perform well and with a high stability,which supports the SANS instrument to finish approximately 2.0 user scientific programs.

Our recent studies demonstrate that the focal adhesion protein Kindlin-2.is critical for chondrogenesis and early skeletal development. Here, we show that deleting Kindlin-2.from osteoblasts using the 2.3-kb mouse Col1 a1-Cre transgene minimally impacts bone mass in mice, but deleting Kindlin-2.using the 10-kb mouse Dmp1-Cre transgene, which targets osteocytes and mature osteoblasts, results in striking osteopenia in mice. Kindlin-2.loss reduces the osteoblastic population but increases the osteoclastic and adipocytic populations in the bone microenvironment. Kindlin-2.loss upregulates sclerostin in osteocytes,downregulates β-catenin in osteoblasts, and inhibits osteoblast formation and differentiation in vitro and in vivo. Upregulation ofβ-catenin in the mutant cells reverses the osteopenia induced by Kindlin-2.deficiency. Kindlin-2.loss additionally increases the expression of RANKL in osteocytes and increases osteoclast formation and bone resorption. Kindlin-2.deletion in osteocytes promotes osteoclast formation in osteocyte/bone marrow monocyte cocultures, which is significantly blocked by an anti-RANKLneutralizing antibody. Finally, Kindlin-2.loss increases osteocyte apoptosis and impairs osteocyte spreading and dendrite ***, we demonstrate an important role of Kindlin-2.in the regulation of bone homeostasis and provide a potential target for the treatment of metabolic bone diseases.

A good prognostic predictor for liver transplantation recipients with benign end-stage liver cirrhosis

BACKGROUND： Post-transplant model for predicting mortality（PMPM, calculated as-5.359＋1.988×ln（serum creatinine [mg/d L]）＋1.089×ln（total bilirubin [mg/d L]）） score has been proved to be a simple and accurate model for predicting the prognosis after liver transplantation（LT） in a single center study. Here we aim to verify this model in a large cohort of ***： A total of 2.2. patients undergoing LT with endstage liver cirrhosis from January 2.03 to December 2.10 were included in this retrospective study. Data were collected from the china Liver Transplant Registry（CLTR）. PMPM score was calculated at 2.-h and 7-d following LT. According to the PMPM score at 2.-h, all patients were divided into the low-risk group（PMPM score ≤-1.4, n=2.09） and the high-risk group（PMPM score 〉-1.4, n=2.8）. The area under receiver operator characteristic curve（AUROC） was calculated for evaluating the prognostic ***： The 1-, 3-, and 5-year patient survival rates in the low-risk group were significantly higher than those in the high-risk group（90.2.%, 88.01%, and 86.03% vs 63.16%, 59.62., and 56.43%, respectively, P〈0.001）. In the high-risk group, 131 patients had a decreased PMPM score（≤-1.4） at 7-d, and their cumulative survival rate was significantly higher than the other 87 patients with sustained high PMPM score（〉-1.4）（P〈0.001）. For predicting 3-month mortality, PMPM score showed a much higher AUROC than post-transplant MELD score（P〈0.05）.CONCLUSION： PMPM score is a simple and effective tool to predict short-term mortality after liver transplantation in patients with benign liver diseases, and an indicator for prompt salvaging treatment as well.

Effect of fragile histidine triad gene transduction on proliferation and apoptosis of human hepatocellular carcinoma cells

AIM:To evaluate the inhibitory effects of human fragile histidine triad(FHIT) gene on cell proliferation and apoptosis in human hepatocellular carcinoma line Hep3B in vitro. METHODS:A recombinant pcDNA3.1(+) /FHIT including the functional region of FHIT gene was constructed and transferred into human hepatocellular carcinoma cells in vitro. mRNA and protein expression of the FHIT gene in the transfected cells was detected by RT-PCR and Western blot,respectively. The effect of FHIT on proliferation was detected by MTT assay. Changes in cell cycle and apoptosis were assayed by flow cytometry. Five mice received subcutaneous transplantation of Hep3B-FHIT;5 mice received subcutaneous transplantation of normal Hep3B and Hep3B-C as controls. The body weight of nude mice and tumor growth were measured. RESULTS:RT-PCR and Western blot analysis showed that the expression level of FHIT-mRNA and FHIT protein was higher in Hep3B cells after infection withpcDNA3.1(+) /FHIT. The growth of Hep3B cells treated with pcDNA3.1(+) /FHIT was significantly inhibited. The pcDNA3.1(+) /FHIT-transfected Hep3B cells showed a significantly higher cell rate at G0-G1 phase and increased apoptosis in comparison with controls(P < 0.05) . The growth of transplanted tumor was inhibited markedly by FHIT. Tumors arising from the Hep3B-FHIT cells occurred much later than those arising from the Hep3B and Hep3B-C cells. The growth of Hep3B-FHIT cells was slow and the tumor volume was low. CONCLUSION:Transduction of FHIT gene inhibits the growth of human hepatocellular carcinoma cells and induces cell apoptosis in vivo and in vitro.

Carbon dots(CDs), a new building unit, have been revolutionizing the fields of biomedicine, bioimaging, and optoelectronics with their excellent physical, chemical, and biological properties. However, the difficulty of preparing excitation-dependent full-spectrum fluorescent CDs has seriously hindered their further research in fluorescence emission mechanisms and biomedicine. Here, we report full-spectrum fluorescent CDs that exhibit controlled emission changes from purple(380 nm) to red(613 nm) at room temperature by changing the excitation wavelength, and the excitation dependence was closely related to the regulation of sp2.and sp3 hybrid carbon structures by β-cyclodextrin-related groups. In addition,by regulating the content of β-cyclodextrin, the optimal quantum yields of full-spectrum fluorescent CDs were 8.97%, 8.35%, 7.90%, 9.69% and 17.4% at the excitation wavelengths of 340, 350, 390, 410 and 540 nm,respectively. Due to their excellent biocompatibility and color tunability, full-spectrum fluorescent CDs emitted bright and steady purple, blue, green, yellow, and red fluorescence in MCF-7 cells. Moreover, we optimized the imaging conditions of CDs and mitochondrial-specific dyes;and realized the mitochondrialtargeted co-localization imaging of purple, blue and green fluorescence. After that, we also explored the effect of full-spectrum fluorescent CDs in vivo fluorescence imaging through the intratumorally, subcutaneously, and caudal vein, and found that full-spectrum fluorescent CDs had good fluorescence imaging ability in vivo.

In iron-based superconductors,the(0,π) or(π,0) nematicity,which describes an electronic anisotropy with a fourfold symmetry breaking,is well established and believed to be important for understanding the superconducting ***,how exactly such a nematic order observed in the normal state can be related to the superconducting pairing is still ***,by performing angular-dependent in-plane magnetoresistivity using ultra-thin flakes in the steep superconducting transition region,we unveil a nematic superconducting order along the(π,π) direction in electron-doped BaFe_(2.x)Ni_(x)As_(2. from under-doped to heavily overdoped regimes with x=0.065- *** shows superconducting gap maxima along the(π,π) direction rotated by 45° from the nematicity along(0, π) or(π,0) direction observed in the normal state.A similar(π,π)-type nematicity is also observed in the under-doped and optimally doped hole-type Ba1-yKyFe2.As_(2.,with y=*** results suggest that the(π,π) nematic superconducting order is a universal feature that needs to be taken into account in the superconducting pairing mechanism in iron-based superconductors.
