Hidden natural products are representative of defensive strategies produced in vivo in diseased plants,a process that is induced by the plant immune *** first transcriptome library of uninfected and pathogen infected Hibiscus tiliaceus stems was constructed by transcriptome sequencing technology,genes related to cadinene-type sesquiterpenoid biosynthesis were screened and combined with ultra-performance liquid chromatography-quadrupole-time of flight mass spectrometry(UPLC-QTOF-MS)analysis data,which indicated pathological tissue had potential to produce novel carbon skeletons of cadinane sesquiterpenoid ***,two cadinane-derived sesquiterpenoid dimers with unprecedented carbon skeletons,hibisceusanols A(1)and B(2)were isolated for the first time from the stems of *** induced by plant-microbial *** structures and absolute configurations were unambiguously established by spectroscopy,advanced chemistry development(ACD)and electronic circular dichroism(ECD)*** 1 and 2 exhibited significant antitumor activity in vitro with half maximal inhibitory concentration(IC_(5)0)values of 2.3–7.2μmol/*** anticancer effect was generated via the induction of Hep G2 cell apoptosis by inhibiting the phosphatidylinositol 3-kinase(PI3K)pathway.

Metabolic dysfunction-associated steatotic liver disease(MASLD),formerly known as non-alcoholic fatty liver disease(NAFLD),is increasingly prevalent in Asia,1 an area also endemic for chronic hepatitis B(CHB).Our previous study showed that the coexistence of CHB and hepatic steatosis(HS)has reached 36.5%in Asia,2 with a similar figure observed globally.3 This coexistence may exacerbate the progression of hepatic fibrosis.

In this study,we aim to assess dynamical downscaling simulations by utilizing a novel bias-corrected global climate model(GCM)data to drive a regional climate model(RCM)over the Asia-western North Pacific *** simulations were conducted with a 25-km grid spacing for the period 1980–*** first simulation(WRF_ERA5)was driven by the European Centre for Medium-Range Weather Forecasts Reanalysis 5(ERA5)dataset and served as the validation *** original GCM dataset(MPI-ESM1-2-HR model)was used to drive the second simulation(WRF_GCM),while the third simulation(WRF_GCMbc)was driven by the bias-corrected GCM *** bias-corrected GCM data has an ERA5-based mean and interannual variance and long-term trends derived from the ensemble mean of 18 CMIP6 *** demonstrate that the WRF_GCMbc significantly reduced the root-mean-square errors(RMSEs)of the climatological mean of downscaled variables,including temperature,precipitation,snow,wind,relative humidity,and planetary boundary layer height by 50%–90%compared to the WRF_***,the RMSEs of interannual-tointerdecadal variances of downscaled variables were reduced by 30%–60%.Furthermore,the WRF_GCMbc better captured the annual cycle of the monsoon circulation and intraseasonal and day-to-day *** leading empirical orthogonal function(EOF)shows a monopole precipitation mode in the WRF_*** contrast,the WRF_GCMbc successfully reproduced the observed tri-pole mode of summer precipitation over eastern *** improvement could be attributed to a better-simulated location of the western North Pacific subtropical high in the WRF_GCMbc after GCM bias correction.

Photodynamic therapy(PDT)has emerged as a significant cancer therapy ***,cation-based organic small molecule aggregation-induced emission(AIE)photosensitizers(PSs)attract the wide atten-tion of many scientists,due to improved reactive oxygen species(ROS)production after ***,such PSs tend to localize only the mitochondria,limiting the death way of tumor cells(usu-ally apoptosis)during PDT process,which may affect the therapeutic effect under some ***,we designed a novel water-soluble three positive charge PS,TPAN-18F,which could be distributed uniformly in cell cytoplasm and had distribution in different sub-organelles(mitochondria,endoplasmic reticulum,lysosome).The experimental results showed that TPAN-18F-based PDT process can not only disrupt mitochondrial functions(reducing ATP production and destroying mitochondrial membrane po-tential),but also elevate the intracellular lipid peroxides(LPOs)level,which evoke the non-apoptotic death manner of tumor ***,in vivo studies showed that TPAN-18F-based PDT could effectively inhibit tumor ***,we believe that the construction of TPAN-18F is suggestive for tumor non-apoptotic therapy.

Despite the synergy of immune checkpoint blockade(ICB) therapy and photodynamic therapy(PDT) holds great promise as countermeasures against breast cancer, exploring long-term or flexible short-time therapeutic strategies in “cold” tumors remains a great challenge. Here, we present a polyunsaturated fatty acid-doped liposomal hydrogel Lp(DHA)@CP Gel loaded with photosensitizer chlorin e6(Ce6) and programmed death-ligand 1 antibody(αPD-L1) for flexible local photoimmunotherapy with merely singledosed administration. The presence of polyunsaturated fatty acid(docosahexaenoic acid, DHA) doped in particle membrane endows liposomes with flexibly reactive oxygen species(ROS)-responsive release capability, which was attributed to the presence of abundant unsaturated groups. The αPD-L1 was repeatedly induced to in situ release in response to the PDT under photo-exposure. The immunogenic cell death(ICD) effect of PDT evoked “cold” breast tumor to “hot” one, and then assisted the cascade releasedαPD-L1 to synergistically boost the immunotherapy. After a single dose of peritumoral administration of Lp(DHA)@CP Gel, the on-demand treatment can maximize patient compliance and safety by adjusting therapeutic behaviors via a photo on-off switch. This work presents a flexible medication platform,showing promise in improving the objective response rate of ICB therapy and minimizing its systemic toxicity.
