Assessing genetic diversity of wild populations of Japanese flounderusing AFLP markers

Amplified fragment length polymorphism （AFLP） analysis was used to evaluate the genetic diversity of four wild geographical populations of Japanese flounder （Paralichthys olivaceus）. A total of 775 loci （58.32% of which was polymorphic） in the range between 100 and 1 300 base pairs were detected from 110 individuals using seven primer combinations. The percentage of polymorphic loci detected by single primer combination for each population was calculated, ranging from 19.59% to 53.33%. Genetic similarities within and among the populations were calculated from the binary matrices of presence - absence. Phylogenetic tree of four populations was constructed by using the UPGMA method using PHYLIP Version 3.5. According to intrapopulation genetic similarities, CW population displayed the highest genetic diversity value and KY population had the lowest genetic diversity value. The distance between CW and CF populations was the farthest, which was possibly resulted from the farthest distance of Weihai of Shandong and Fujian of China compared with the geographical distance between other locations of populations. The subpopulation differentiation value （G.,） is 0. 356 5, showing a certain extent of differentiation among the four geographical populations. AFLP technology was confirmed to be an effective tool to assess within- and among-population genetic diversity of Japanese flounder. The present survey provided significant insights for research in the Japanese flounder breeding program.

Influence of composition and annealing temperature on structure and magnetic properties of amorphous and nanocrystalline Fe78.4-xCoxSi9B9Nb2.6Cu1 （x=27.4, 40.0, 51.0, 78.4） alloys was investigated by X-ray diffraction （XRD） and the temperature dependence of permeability. According to the initial crystallization temperature （Tx1） from differential scanning calorimetry （DSC） curves of as-quenched amorphous alloys, 490-700 ℃ isothermal annealing was carded out to obtain the characteristic nanocrystalline structure. Furthermore, the soft magnetic properties were measured by temperature evolution of magnetic permeability to obtain the correlation between Co content, annealing temperature and magnetic permeability. The results show that, on the one hand, the annealing temperature exerts a significant effect on phase structure and initial permeability （μi）. The higher-temperature （from 550 to 610 ℃） annealed Co content nanocrystalline samples can remain higher μi at elevated temperature. On the other hand, partial substitution Fe by Co can improve the high-temperature magnetic stability;however, the room-temperature permeability of higher Co content alloys decreases obviously at the same time. This phenomenon was analyzed from the viewpoint of the saturation magnetic induction （Bs）, magnetic anisotropy （〈K〉） and magnetostriction （λs）.

Soil microbes play essential roles in the biogeochemical processes of organic carbon and nutrient cycling. Many studies have reported various short-term effects of fertilization on soil microbes. However, less is known about the effects of longterm fertilization regimes on the rhizosphere. Therefore, the objective of this study was to explore how the soil microbial communities in the rhizosphere respond to different long-term fertilization strategies. Based on a 21-year field treatment experiment in Guizhou, China, we extracted phospholipid fatty acids(PLFAs) to determine the microbial community structure in both the non-rhizosphere(NR) and rhizosphere(R). Six treatments were included: no fertilizer(CK), mineral nitrogen fertilizer(N), N with potassium(NK), phosphorus with K(PK), NPK, and NPK combined with manure(MNPK). The results showed that total PLFAs under unbalanced mineral fertilization(N, NK and PK) were decreased by 45% on average in the NR compared with CK, whereas MNPK increased fungi and G~– bacteria abundance significantly in both the NR(by 33 and 23%) and R(by 15 and 20%), respectively. In addition, all microbial groups in the R under these treatments(N, NK and PK) were significantly increased relative to those in the NR, except for the ratio of F/B and G^+/G~–, which might be due to the high nutrient availability in the R. Soil pH and SOC significantly regulated the soil microbial community and structure, explaining 51 and 20% of the variation in the NR, respectively. However, the rhizosphere microbial community structure was only significantly affected by soil pH(31%). We concluded that the soil microbial community in the NR was more strongly affected by long-term fertilization than that in the R due to the rhizosphere effect in the agricultural ecosystem. Rhizosphere nutrient conditions and buffering capacity could help microbial communities resist the change from the long-term fertilization.
