Estrogenic activities of two synthetic pyrethroids and their metabolites

Synthetic pyrethroids （SPs） are among the most common pesticides in current use, and so far, several SPs have been assessed for their potential estrogenicities by various methods. Previous studies have shown that the estrogenicities partly come from their metabolites. Although considerable information is available with respect to the metabolism and environmental degradation of SPs, little is known about the estrogenicities of the metabolites. In this study, permethrin （PM） and β-cypermethrin （CP）, as well as their metabolites （3-phenoxybenzoic alcohol （PBCOH）, 3-phenoxybenzaldehyde （PBCHO） and 3-phenoxybenzoic acid （PBCOOH） were evaluated for their estrogenic activities in the MCF-7 human breast carcinoma cell line. In the MCF-7 cell proliferation assay, PM and CP exhibited significant estrogenic activities at 10-7 mol/L, comparable to 17β-estradiol （E2） of 10-9 tool/L, with the relative proliferative effect ratios of 55.4% and 56.3%, respectively. The real-time quantitative polymerase chain reaction （qRT-PCR） results confirmed the estrogenicities of PM and CP with significant alteration of pS2 and ERα mRNA levels observed at 10-6 mol/L. For the three major metabolites, PBCOH and PBCOOH exhibited estrogenic activities in all assays, while no significant estrogenic responses was observed for PBCHO compared to the vehicle control. In particular, PBCOH had even slightly stronger estrogenic activity than its parent compounds, indicating that metabolism may be one of the reasons for the estrogenicities of the SPs. Given the widespread use of SPs, the toxicological effects of parent compounds and their metabolites should be taken into consideration in the risk assessment of SPs.

Objective:To evaluate whether the efficacy of Getong Tongluo Capsule(葛酮通络胶囊,GTC,consisted of total flavone of Radix Puerariae)on improving patients'quality of life and lowering blood pressure are superior to the extract of Ginkgo biloba(EGB)for patients with convalescent-phase ischemic stroke and primary ***:This randomized,positive-drug-and placebo-controlled,double-blind trial was conducted from September 2015 to October *** 477 eligible patients from 18 hospitals in china were randomly assigned in a 2:1:1 ratio to the following interventions,twice a day for 12 weeks:(1)GTC 250 mg plus EGB-matching placebo 40 mg(237 cases,GTC group),(2)EGB 40 mg plus GTC-matching placebo 250 mg(120 cases,EGB group)or(3)GTC-matching placebo 250 mg plus EGB-matching placebo 40 mg(120 cases,placebo group).Moreover,all patients were orally administered aspirin enteric-coated tablets 100 mg,once a day fo r 12 *** primary outcome was the Barthel Index(Bl).The secondary outcomes included the control rate of blood pressure and National Institutes of Health Stroke Scale(NIHSS)*** incidence and severity of adverse events(AEs)were calculated and ***:The Bl relative independence rates,the clinical recovery rates of NIHSS,and the total effective rates of NIHSS in the GTC and EGB groups were significantly higher than the placebo group at 12 weeks after treatment(Pen the GTC and EGB groups(P>0.05).The control rate of blood pressure in the GTC group was significantly higher than the EGB and placebo groups at 12,18 and 24 weeks after treatment(P1).There were no statistically significant differences in the incidences of AEs,adverse drug reactions,or serious AEs among the 3 groups(P>0.05).Conclusion:GTC exhibited significant efficacy in improving patients'quality of life as well as neurological function and controlling hypertension.

Objective: To assess whether the TaqIB polymorphism of cholesteryl ester transfer protein (CETP) is associated with coronary artery disease (CAD) in Chinese population, we performed a meta-analysis in this paper. Methods: We searched PubMed, Embase, the Science Citation Index (SCI), the china Biological Medicine database (CBM), the china National Knowledge Infrastructure (CNKI), and the Wanfang database for relevant articles. Data were extracted, and pooled odds ratios (ORs) and 95% confidence intervals (CIs) were calculated. Results: The lit- erature search yielded 448 studies, in which 10 case-control studies including 1 694 cases and 1 456 controls matched the selection criteria. The combined B1 and B2 allele frequencies were 0.587 and 0.413, respectively. The pooled OR was 1.10 (95% CI, 0.89-1.34) for comparing the B1B1 or B1B2 carriers with B2B2 carriers, and was 1.27 (95% CI, 1.09-1.49) in the B1B1 carriers versus B2B2 or B1B2 carriers. Conclusions: In the present study, the TaqIB poly- morphism of CETP was found to be associated with CAD in the Chinese population.

目的探讨我国遗传性平滑肌瘤病和肾细胞癌综合征相关性肾癌(HLRCC-RCC)患者的临床特征以及系统治疗的有效性。方法去回顾性分析2008年1月至2021年12月在国内15家医疗中心基因诊断明确有FH胚系突变的119例HLRCC-RCC患者的临床资料。男73例,女46例;中位年龄38(13,74)岁;中位肿瘤直径6.5(1.0,20.5)cm。I~Ⅱ期38例(31.9%),Ⅲ~V期81例(68.1%)。本组119例中11例同时表现为皮肤平滑肌瘤,46例女性HLRCC-RCC患者中44例有子宫肌瘤病史。总结患者的病理特点、治疗方法,以及患者预后生存情况。结果本研究119例中,共86例患者接受手术治疗,其中根治性肾切除术70例,肾部分切除术5例,减瘤性肾切除术11例;其余33例初诊转移患者行肾穿刺活检术。基因检测结果显示119例均存在FH基因胚系突变,其中94例FH基因点突变,18例FH基因插人/缺失突变,4例FH基因剪接突变,2例FH基因大片段缺失,1例FH基因拷贝数变异。免疫组化染色结果显示113例患者2-SC强阳性,FH阴性。本组共102例患者接受系统治疗,包括44例初诊转移患者和58例术后转移患者。其中酪氨酸激酶抑制剂(TKI)联合免疫检查点抑制剂(ICI)治疗组33例,贝伐单抗联合厄洛替尼治疗组8例,TKI单药治疗组61例。所有患者均获得随访,中位随访时间17(3,82)个月。生存分析结果显示TKI联合ICI治疗组中位PFS为18(5,38)个月,中位OS未达到;贝伐单抗联合厄洛替尼治疗组中位PFS为12(5,14)个月,中位OS为30(10,32)个月;TKI单药治疗组中位PFS为10(364)个月,中位OS为44(10,74)个月。TKI联合ICI治疗组的PFS(P=0.009)和OS(P=0.006)均优于贝伐单抗联合厄洛替尼治疗组;TKI联合ICI治疗组的PFS(P=0.003)和OS(P=0.028)均优于TKI单药治疗组。结论HLRCC-RCC罕见,具有家系遗传特性,恶性程度高,预后不佳,易出现转移。免疫组化染色2-SC强阳性及FH阴性可为临床诊断提供重要依据,基因检测显示FH基因胚系突变可明确诊断。初步研究结果证实TKI联合ICI治疗转移性HLRCC-RCC的临床效果较好,但有待大样本多中心随机对照临床研究进一步验证。

Coronavirus disease 2019(COVID-19)is a public health disaster that has not been encountered for a hundred *** January 12,2020,the World Health Organization(WHO)confirmed and named the coronavirus which caused unexplained pneumonia as 2019 novel coronavirus(2019-nCoV).Since then,the disease caused by this virus had been named as coronavirus disease 2019(COVID-19).On February 11,2020,the International Committee on Taxonomy of Viruses announced that the english name of the novel coronavirus was severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)[1].At present,SARS-CoV-2 infection is still rampant *** of September 10,2021,there were about 222 million confirmed cases of COVID-19 and more than 4.5 million deaths worldwide[2].According to the American centers for Disease Control and Prevention,by July 29,2021,4.19 million pediatric COVID-19 cases had been reported in the United States。

BACKGROUND The treatment outcome of transarterial chemoembolization(TACE)in unresectable hepatocellular carcinoma(HCC)varies greatly due to the clinical heterogeneity of the ***,several prognostic systems have been proposed for risk stratification and candidate identification for first TACE and repeated TACE(re-TACE).AIM To investigate the correlations between prognostic systems and radiological response,compare the predictive abilities,and integrate them in sequence for outcome *** This nationwide multicenter retrospective cohort consisted of 1107 unresectable HCC patients in 15 Chinese tertiary hospitals from January 2010 to May *** Hepatoma Arterial-embolization Prognostic(HAP)score system and its modified versions(mHAP,mHAP2 and mHAP3),as well as the six-and-twelve criteria were compared in terms of their correlations with radiological response and overall survival(OS)prediction for first *** same analyses were conducted in 912 patients receiving re-TACE to evaluate the ART(assessment for re-treatment with TACE)and ABCR(alpha-fetoprotein,Barcelona Clinic Liver Cancer,Child-Pugh and Response)systems for post re-TACE survival(PRTS).RESULTS All the prognostic systems were correlated with radiological response achieved by first TACE,and the six-and-twelve criteria exhibited the highest correlation(Spearman R=0.39,P=0.026)and consistency(Kappa=0.14,P=0.019),with optimal performance by area under the receiver operating characteristic curve of 0.71[95%confidence interval(CI):0.68-0.74].With regard to the prediction of OS,the mHAP3 system identified patients with a favorable outcome with the highest concordance(C)-index of 0.60(95%CI:0.57-0.62)and the best area under the receiver operating characteristic curve at any time point during follow-up;whereas,PRTS was well-predicted by the ABCR system with a C-index of 0.61(95%CI:0.59-0.63),rather than ***,combining the mHAP3 and ABCR systems identified candidates suitable for TA
