Objective: Anemoside B4(AB4), the most abundant triterpenoidal saponin isolated from Pulsatilla chinensis, inhibited influenza virus FM1 or Klebsiella pneumoniae-induced pneumonia. However, the anti-SARS-CoV-2 effect of AB4 has not been unraveled. Therefore, this study aimed to determine the antiviral activity and potential mechanism of AB4 in inhibiting human coronavirus SARS-CoV-2 in vivo and in vitro.Methods: The cytotoxicity of AB4 was evaluated using the Cell Counting Kit-8(CCK8) assay. SARS-CoV-2 infected HEK293T, HPAEpiC, and Vero E6 cells were used for in vitro assays. The antiviral effect of AB4 in vivo was evaluated by SARS-CoV-2-infected hACE2-IRES-luc transgenic mouse model. Furthermore,label-free quantitative proteomics and bioinformatic analysis were performed to explore the potential antiviral mechanism of action of AB4. Type Ⅰ IFN signaling-associated proteins were assessed using Western blotting or immumohistochemical staining.Results: The data showed that AB4 reduced the propagation of SARS-CoV-2 along with the decreased Nucleocapsid protein(N), Spike protein(S), and 3C-like protease(3CLpro) in HEK293T cells. In vivo antiviral activity data revealed that AB4 inhibited viral replication and relieved pneumonia in a SARS-CoV-2 infected mouse model. We further disclosed that the antiviral activity of AB4 was associated with the enhanced interferon(IFN)-β response via the activation of retinoic acid-inducible gene Ⅰ(RIG-1) like receptor(RLP) pathways. Additionally, label-free quantitative proteomic analyses discovered that 17 proteins were significantly altered by AB4 in the SARS-CoV-2 coronavirus infections cells. These proteins mainly clustered in RNA metabolism.Conclusion: Our results indicated that AB4 inhibited SARS-CoV-2 replication through the RLR pathways and moderated the RNA metabolism, suggesting that it would be a potential lead compound for the development of anti-SARS-CoV-2 drugs.

Time-triggered State-machine Reliable Software Architecture for Micro Turbine Engine Control

Time-triggered （TT） embedded software pattern is well accepted in aerospace industry for its high reliability. Fi-nite-state-machine （FSM） design method is widely used for its high efficiency and predictable behavior. In this paper, the time-triggered and state-machine combination software architecture is implemented for a 25 kg thrust micro turbine engine （MTE） used for unmanned aerial vehicle （UAV） system; also model-based-design development workflow for airworthiness software directive DO-178B is utilized. Experimental results show that time-triggered state-machine software architecture and development method could shorten the system development time, reduce the system test cost and make the turbine engine easily comply with the airworthiness rules.

Measurements of heat transfer and pressure in a trailing edge cavity of a turbine blade

An experimental investigation is conducted to obtain the heat transfer and pressure drop data for an integral trailing edge cavity test section that simulates a novel turbine blade＇s internal cooling passage with bleed holes. Local heat transfer is measured on both the suction and pressure sides by a transient liquid crystal technique, while pressures at six positions are recorded by pressure calibrators. Moreover, flow characteristic and its effect on heat transfer are analyzed for conditions with or without bleed flow. The experimental results show that, in the cases with bleed flow, local heat transfer on the pressure side exceeds that on the suction side in the first and second channels. In the cases without bleed flow, in the first and third channels, local heat transfer on the suction side weakens whilst it increases significantly on the pressure side. For the second channel, non-bleed condition leads to a more balanced heat transfer distribution between the upstream and downstream channel. Besides, after the bleed holes are blocked, heat transfer in the first bend region on the suction side declines sharply, while the opposite phenomenon occurs for the second bend region on the pressure side. In both bleed and non-bleed cases, the total pressure of six measurement positions decreases continuously along the channel at the same Reynolds number and it promotes for higher Reynolds number. Among all the measurement points, under the same flow rate condition, the highest speed occurs at Position 5, which also shows the maximum difference between the total and static pressures. When the bleed holes are blocked, the total pressure at each measurement position appears to increase.

Research Progress of Seafloor Pockmarks in Spatio-Temporal Distribution and Classification

Seafloor pockmarks are important indicators of submarine methane seepages and slope instabilities.In order to promote the understanding of submarine pockmarks and their relationship with sediment instabilities and climate changes,here we summarize the research results of pockmarks in the spatio-temporal distributions and shaping factors.Most of pockmarks occur along active or passive continental margins during the last 25 kyr B.P..Circular and ellipse are the most common forms of pockmarks,whereas pockmarks in a special crescent or elongated shape are indicators of slope instabilities,and ring-shape pockmarks are endemic to the gas hydrate zones.Further researches should be focused on the trigger mechanism of climate changes based on the pockmarks in the high latitudes formed during the deglaciation periods,and the role of gas hydrates in the seafloor evolution should be elucidated.In addition,the feature of pockmarks at their early stage(e.g.,developing gas chimneys and gas driving sedimentary doming)and the relations between pockmarks and mass movements,mud diapirs could be further studied to clarify the influences of rapid methane release from submarine sediments on the atmospheric carbon contents.

根据2021年4月至2022年1月4个航次渔业资源和环境数据调查资料,对瑞安海域三疣梭子蟹(Portunus trituberculatus)时空分布及其与影响因素的关系进行研究,包括其海域优势度、群落结构组成、资源密度分布等,同时运用广义可加模型(GAM)分析了资源密度与影响因素的关系。结果表明:瑞安海域中,三疣梭子蟹在春季和夏季为重要种(100≤IRI 3)。三疣梭子蟹平均资源密度范围为16.57~422.60 kg/km^(2),平均资源密度呈现以秋季最高,春季最低,秋季是春季的25.5倍。GAM分析表明,模型解释率为80.2%,季节、表层水温、底层水温、底层盐度和表层叶绿素a浓度对三疣梭子蟹资源密度具有显著影响(p <0.05),其中季节影响最大。通过本研究,为掌握瑞安海域三疣梭子蟹资源动态变化规律提供了参考,进而为三疣梭子蟹增殖放流提供理论支撑。

Visualization Experiments of a Specific Fuel Flow Through Quartz-glass Tubes Under both Sub- and Supercritical Conditions

The present work is a visualization study of a typical kerosene （RP-3） flowing through vertical and horizontal quartz-glass tubes under both sub- and supercritical conditions by a high speed camera. The experiments are accomplished at temperatures of 300-730 K under pressures from 0.107-5 MPa. Six distinctive two-phase flow patterns are observed in upward flow and the critical point of RP-3 is identified as critical pressure pc=2.33 MPa and critical temperature Tc=645.04 K and it is found that when the fluid pressure exceeds 2.33 MPa the flow can be considered as a single phase flow. The critical opalescence phenomenon of RP-3 is observed when the temperature is between 643.16 K and 648.61 K and the pressure is between 2.308 MPa and 2.366 MPa. The region filled by the critical opalescence in the upward flow is clearly larger than that in the downward flow due to the interaction between the buoyancy force and fluid inertia. Morecover, obvious layered flow phenomenon is observed in horizontal flow under supercritical pressures due to the differences of gravity and density.

为解决低进气量条件下油气涡轮功率与效率提升困难的问题,本文结合裂解油气物性特点,针对性地提出了局部进气裂解油气向心涡轮设计方案。通过数值仿真详细分析了向心式油气涡轮在设计工况下的内部流动特征,并总结了非设计工况下轮周效率随压比、转速以及正癸烷裂解度的变化规律与影响机制。仿真结果表明,本文所设计油气涡轮在0.5 kg/s进气量条件下可实现38.8 kW功率输出,轮周效率为68%,能够满足油气涡轮气动设计需求。根据结果分析可知,随着压比的增大,轮周效率呈现出先增大后减小的趋势:低压比条件下轮周效率增大主要与转子入口攻角损失有关;高压比条件下轮周效率降低则主要是由于激波损失及其引发的附面层分离损失不断增大导致的。研究表明,轮周效率随转速、裂解度发生变化时与转子入口攻角有直接关系,其中,油气工质裂解度每提高20%,涡轮轮周效率可提高13%左右。

从涡轮结构形式的角度探究了油气涡轮轴向力平衡问题。建立了半开式、开式与闭式3种局部进气裂解油气向心涡轮,通过数值仿真对比分析了设计工况下3种油气涡轮的气动性能与轴向力,并归纳总结了非设计工况下总轴向力随压比的变化规律。仿真结果表明:半开式、开式与闭式油气涡轮的总体气动性能相近但轴向力表现有明显的差别。当涡轮压比发生变化时,开式油气涡轮轴向力稳定性最好,半开式油气涡轮轴向力稳定性最差,闭式油气涡轮轴向力始终是3种油气涡轮中最小的。由结果分析可知,当涡轮压比低于3时,应避免选用半开式油气涡轮,而涡轮压比变化较大时,则宜采用开式油气涡轮,此外,在采用闭式油气涡轮时需要轴承预留足够的轴向载荷裕度。

Carbonate Sequence Stratigraphy of a Back-Arc Basin: A Case Study of the Qom Formation in the Kashan Area, Central Iran

The Qom Formation comprises Oligo-Miocene deposits from a marine succession distributed in the Central Basin of Iran. It is composed of five members designated as A-F. Little previous work exists on the sequence stratigraphy. Based on an integrated study of sequence stratigraphy with outcrop data, wells and regional seismic profiles, the Qom Formation is interpreted as a carbonate succession deposited in a mid.Tertiary back-arc basin. There are two second-order sequences （designated as SS1 and SS2） and five third-order sequences （designated as S1-S5）. Five distinct systems tracts including transgressive, highstand, forced regressive, slope margin and Iowstand have been recognized. The relationship between the sequences and lithologic sub-units has been collated and defined （S1 to S5 individually corresponding to A-C1, C2--C4, D-E, the lower and upper portions of F）; a relative sea level change curve and the sequence stratigraphic framework have been established and described in detail. The coincidence of relative sea level change between that of the determined back-arc basin and the world indicates that the sedimentary cycles of the Qom Formation are mainly controlled by eustatic cycles. The variable combination of the systems tracts and special tectonic-depositional setting causally underpin multiple sequence stratigraphic framework styles seen in the carbonates of the back-arc basin revealing：（1） a continental margin basin that developed some form of barrier, characterized by the development of multiple cycles of carbonate-evaporites; （2） a flat carbonate ramp, which occurred on the southern shelf formed by the lack of clastic supply from nearby magmatic islands plus mixed siliciclastics and carbonates that occurred on the northern shelf due to a sufficient clastics supply from the land; and （3） a forced regressive stratigraphic stacking pattern that occured on the southern shelf and in basin lows due to the uplifting of the southern shelf.
