RNA interference(RNAi)is useful for selective gene *** P4503A4(CYP3A4),which metabolizes approximately 50% of drugs in clinical use,plays an important role in drug *** this study,we aimed to develop a short hairpin RNA(shRNA)to modulate CYP3A4 *** new shRNAs(S1,S2 and S3)were designed to target the coding sequence(CDS)of CYP3A4,cloned into a shRNA expression vector,and tested in different *** mixture of three shRNAs produced optimal reduction(55%)in CYP3A4 CDS-luciferase activity in both CHL and HEK293 *** CYP3A4 expression in HepG2 cells was decreased about 50%at both mRNA and protein level after transfection of the mixture of three *** contrast,CYP3A5 gene expression was not altered by the shRNAs,supporting the selectivity of CYP3A4 *** addition,HepG2 cells transfected with CYP3A4 shRNAs were less sensitive to Ginkgolic acids,whose toxic metabolites are produced by *** results demonstrate that vector-based shRNAs could modulate CYP3A4 expression in cells through their actions on CYP3A4 CDS,and CYP3A4 shRNAs may be utilized to define the role of CYP3A4 in drug metabolism and toxicity.

This study delved into the formulaic patterns of liver cancer treatment outlined in“The Complete Book of Good Tumor Prescriptions”and explored the action mechanisms of core traditional Chinese medicine(TCM)***,liver cancer treatment formulas were extracted from the aforementioned book,and Excel 2017 was employed for calculating the frequency,properties,and meridians associated with the *** SPSS Modeler 18.0 was utilized for the analysis of TCM association rules,and Cytoscape 3.7.2 was employed for the visualization of these ***,network pharmacology was utilized to analyze the action mechanisms of core TCM combinations,leveraging TCMSP,TCMID,Genecards,DAVID,and other *** results revealed the inclusion of 131 prescriptions encompassing 303 *** core TCMs predominantly comprised those invigorating blood circulation and removing blood stasis,clearing heat,and tonifying *** flavor of the TCMs was chiefly bitter or pungent,with a prevailing cold property entering the liver and spleen *** analysis indicated that the support of Angelicae sinensis radix-Trionycis carapax was the *** pharmacology predictions indicated that Angelicae sinensis radix-Trionycis carapax possessed 20 target genes associated with anti-liver cancer properties,including IL-6,MAPK3,and *** survival analysis demonstrated high expression of MAPK3 and SRC in liver cancer patients,correlating with a poor *** analysis identified major anti-liver cancer pathways for Angelicae sinensis radix-Trionycis carapax,encompassing the cancer pathway,TNF signaling pathway,and prolactin signaling *** summary,this study elucidated that TCMs for liver cancer treatment primarily consisted of invigorating blood circulation and removing blood stasis,antipyretic,and tonifying *** core TCM pair,Angelicae sinensis radix-Trionycis carapax,appeared pivotal in this *** underlying mechanism might

High-performance Cu/Graphene composite wire synergistically strengthened by nano Cr_(3)C_(2) phase was directly synthesized via hot press sintering followed by severe cold plastic deformation, using liquid paraffin and CuCr alloy powder as the raw materials. Since graphene is in situ formed under the catalysis of copper powder during the sintering process, the problem that graphene is easy to agglomerate and difficult to disperse uniformly in the copper matrix has been solved. The nano Cr_(3)C_(2)-particles nailed at the interface favor to improve the interface bonding. The Cu/Graphene composite possesses high electrical conductivity, hardness, and plasticity. The composite wire exhibits high electrical conductivity of 96.93% IACS, great tensile strength of 488MPa, and excellent resistance to softening. Even after annealing at 400℃ for 1 h, the tensile strength can still reach 268 MPa with a conductivity of about 99.14% *** wire's temperature coefficient of resistance(TCR) is largely reduced to 0.0035/℃ due to the complex structure,which leads the wire to present low resistivity at higher temperatures. Such Cu/Graphene composite wire with excellent comprehensive performance has a good application prospect in high-power density motors.

Background:Associated liver partition and portal vein ligation for staged hepatectomy(ALPPS)is increasingly used for the treatment of primarily unresectable colorectal liver metastases(CRLM)because of the insufficient future liver remnant(FLR).The aim of this study was to share our experience with ALPPS for advanced CRLM and review the role of ALPPS in the management of patients with ***:Consecutive patients(n=10)with CRLM who underwent ALPPS at our center between December 2014 and October 2021 were retrospectively ***:The median age of patients was 58 years(range 49–70 years).Five patients had metachronous CRLM and the other 5 patients had synchronous *** patients had unilateral CRLM and the other 5 patients had bilateral *** patients previously underwent neoadjuvant *** FLR volume increased by 45.3%(35.8%–61.6%)over 14 days(range 7–21 days).The absolute and relative kinetic growth rates were 17.9 mL/day(range 7.5–32.1 mL/day)and 4.6%/day(range 2.3%–8.8%/day),*** morbidity rates were 20%(2/10)and 40%(4/10)after ALPPS stage 1 and stage 2,*** 30-day and 90-day mortality rates were 0%and 10%,respectively.R0 resection was achieved for all patients,and the median disease-free survival was 18.7 ***:ALPPS is an effective treatment option for advanced *** enables rapid hypertrophy of the FLR and achieves a high R0 resection rate with an acceptable oncological ***,ALPPS should be reserved for selected patients because of its relatively high morbidity and mortality.

The new generation of artificial intelligence(AI)research initiated by Chinese scholars conforms to the needs of a new information environment changes,and strives to advance traditional artificial intelligence(AI 1.0)to a new stage of AI *** one of the important components of AI,collective intelligence(CI 1.0),i.e.,swarm intelligence,is developing to the stage of CI 2.0(crowd intelligence).Through in-depth analysis and informative argumentation,it is found that an incompatibility exists between CI 1.0 and CI ***,CI 1.5 is introduced to build a bridge between the above two stages,which is based on biocollaborative behavioral *** 1.5 is the transition from CI 1.0 to CI 2.0,which contributes to the compatibility of the two ***,a new interpretation of the meta-synthesis of wisdom proposed by Qian Xuesen is *** meta-synthesis of wisdom,as an improvement of crowd intelligence,is an advanced stage of bionic intelligence,i.e.,CI *** is pointed out that the dual-wheel drive of large language models and big data with deep uncertainty is an evolutionary path from CI 2.0 to CI 3.0,and some elaboration is *** a result,we propose four development stages(CI 1.0,CI 1.5,CI 2.0,and CI 3.0),which form a complete framework for the development of *** different stages are progressively improved and have good *** to the dominant role of cooperation in the development stages of CI,three types of cooperation in CI are discussed:indirect regulatory cooperation in lower organisms,direct communicative cooperation in higher organisms,and shared intention based collaboration in *** division is the main form of achieving cooperation and,for this reason,this paper investigates the relationship between the complexity of behavior and types of labor ***,based on the overall understanding of the four development stages of CI,the future development direction and research issues of CI are explored.

The paper addresses the decentralized optimal control and stabilization problems for interconnected systems subject to asymmetric *** with previous work,a closed-loop optimal solution to the control problem and sufficient and necessary conditions for the stabilization problem of the interconnected systems are given for the first *** main challenge lies in three aspects:Firstly,the asymmetric information results in coupling between control and estimation and failure of the separation ***,two extra unknown variables are generated by asymmetric information(different information filtration)when solving forward-backward stochastic difference ***,the existence of additive noise makes the study of mean-square boundedness an *** adopted technique is proving and assuming the linear form of controllers and establishing the equivalence between the two systems with and without additive noise.A dual-motor parallel drive system is presented to demonstrate the validity of the proposed algorithm.
