Stromal thrombospondin 1 suppresses angiogenesis in oral submucous fibrosis

A decline in mucosal vascularity is a histological hallmark of oral submucous fibrosis (OSF), a premalignant disease that is largely induced by betel quid chewing. However, the lack of available models has challenged studies of angiogenesis in OSF. Here, we found that the expression of thrombospondin 1 (THBS1), an endogenous angiostatic protein, was elevated in the stroma of tissues with OSF. Using a fibroblast-attached organoid (FAO) model, the overexpression of THBS1 in OSF was stably recapitulated in vitro. In the FAO model,treatment with arecoline, a major pathogenic component in areca nuts, enhanced the secretion of transforming growth factor (TGF)-β1 by epithelial cells, which then promoted the expression of THBS1 in fibroblasts. Furthermore, human umbilical vein endothelial cells (HUVECs)were incorporated into the FAO to mimic the vascularized component. Overexpression of THBS1 in fibroblasts drastically suppressed the sprouting ability of endothelial cells in vascularized FAOs (v FAOs). Consistently, treatment with arecoline reduced the expression of CD31in v FAOs, and this effect was attenuated when the endothelial cells were preincubated with neutralizing antibody of CD36, a receptor of THBS1. Finally, in an arecoline-induced rat OSF model, THBS1 inhibition alleviated collagen deposition and the decline in vascularity in vivo. Overall, we exploited an assembled organoid model to study OSF pathogenesis and provide a rationale for targeting THBS1.

Research progress and challenges in stem cell therapy for diabetic foot: Bibliometric analysis and perspectives

BACKGROUND Stem cell therapy has shown great potential for treating diabetic foot(DF).AIM To conduct a bibliometric analysis of studies on the use of stem cell therapy for DF over the past two decades,with the aim of depicting the current global research landscape,identifying the most influential research hotspots,and providing insights for future research directions.METHODS We searched the Web of Science Core Collection database for all relevant studies on the use of stem cell therapy in DF.Bibliometric analysis was carried out using CiteSpace,VOSviewer,and R(4.3.1)to identify the most notable studies.RESULTS A search was conducted to identify publications related to the use of stem cells for DF treatment.A total of 542 articles published from 2000 to 2023 were identified.The United States had published the most papers on this subject.In this field,Iran’s Shahid Beheshti University Medical Sciences demonstrated the highest productivity.Furthermore,Dr.Bayat from the same university has been an outstanding researcher in this field.Stem Cell Research&Therapy is the journal with the highest number of publications in this field.The main keywords were“diabetic foot ulcers,”“wound healing,”and“angiogenesis.”CONCLUSION This study systematically illustrated the advances in the use of stem cell therapy to treat DF over the past 23 years.Current research findings suggested that the hotspots in this field include stem cell dressings,exosomes,wound healing,and adipose-derived stem cells.Future research should also focus on the clinical translation of stem cell therapies for DF.

Injection of a Self-propelled Polymer into a Small Circular Cavity

The injection of a polymer chain into a small circular cavity under tangential self-propelled force is studied by using Langevin dynamics simulation.Results indicate that the injection dynamics of the active polymer shows strong correlation with the polymer conformation inside the cavity depending on the polymer rigidity(k_(b)).The injection timeτvaries nonmonotonously with increasing k_(b),and reaches its minimum at k_(b)^(*).When k_(b) is small(k_(b)≪k_(b)^(*)),the polymer is nearly random coil in the cavity,and spends a long time at the final stage of the injection process due to the large repulsion between monomers inside the cavity.When k_(b) is moderate(k_(b)∼k_(b)^(*)),the part of polymer inside the cavity forms spiral configuration under the tangential active force,and the whole polymer moves synchronously with a constant velocity during the injection process,leading to a small injection time.When k_(b) is large(k_(b)≫k_(b)^(*)),the polymer is nearly straight at the initial stage of the injection process,and takes a long time to bend itself,leading to a large injection time.

Adsorption limitation investigation on olefins for Cu–BTC

To utilize Cu-benzene-1,3,5-tricarboxylate(Cu-BTC)adsorbed lubricant oils in the self-lubricating field,the adsorption properties of Cu-BTC on different 1-olefins must be clarified.In this work,1-hexene,1-octene,1-nonene,1-decene,1-undecene,and 1-dodecene were studied by the Monte Carlo method and experimentally.The adsorption limit of Cu-BTC for n-olefins was determined as 1-undecene by the adsorption isotherms.This suggested a limit for even straight-chain molecules to the adsorption of Cu-BTC.The maximum ratio of the olefin length of the largest pore diameter(L/D)of Cu-BTC was approximately 1.57.Furthermore,theoretical calculations(radial distribution function(RDF))and experiments(infrared(IR)spectra)confirmed the interaction of n-olefin adsorbates and the Cu-BTC framework occurred between the-CH=of olefins and the Cu and O atoms of the Cu-BTC framework.This work adds to the understanding and investigation of the adsorption of liquid lubricants using Cu-BTC as a metal-organic framework(MOF).

Synergistic Promotion of Electrical, Mechanical and Thermal Properties for Silicone Rubber-based Field Grading Material via Compound Modification

Use of nonlinearconductive SiC/silicone rubber(SR)field grading material(FGM)can improve the local field concentration of composite insulators.Adding large volume fraction and large-size SiC particles(SiCp)into SR can obtain a good field grading effect,but it is accompanied by the deterioration of mechanical properties.Compounding SiC with different shapes can solve this contradiction.By incorporating one-dimensional SiC whiskers(SiCw)to synergize with granular SiCp,SiC/SR FGM with better field-dependent conductivity,mechanical properties and thermal conductivity than large-size SiCp and large volume fraction filling case can be obtained by using smaller size SiCp and lower filling contents.The simulations of 500 kv line insulators show that the modified SiC/SR FGM can reduce the maximum field strength along the insulator surface and at sheath-core rod interfaces by 55%and 71.4%,respectively.The combined application of FGM and grading ring can achieve a complementary effect.Using FGM to partially replace the role of the grading rings,the field strength indicators can still meet the operational requirements after the tube radius and shielding depth of the grading rings at both ends are reduced by 36.2%and 40%separately,which is a benefit to alleviating the problems of high weight and large volume faced by traditional field grading methods.

Interactive mechanism and friction modelling of transient tribological phenomena in metal forming processes: A review

The accurate representation of tribological boundary conditions at the tool–workpiece interface is crucial for analysis and optimization of formability,material flow,and surface quality of components during metal forming processes.It has been found that these tribological conditions vary spatially and historically with process parameters and contact conditions.These time-dependent tribological behaviours are also known as transient tribological phenomena,which are widely observed during forming processes and many other manufacturing application scenarios.However,constant friction values are usually assigned to represent complex and dynamic interfacial conditions,which would introduce deviations in the relevant predictions.In this paper,transient tribological phenomena and the contemporary understanding of the interaction between friction and wear are reviewed,and it has been found that these phenomena are induced by the transitions of friction mechanisms and highly dependent on complex loading conditions at the interface.Friction modelling techniques for transient behaviours for metal forming applications are also reviewed.To accurately describe the evolutionary friction values and corresponding wear during forming,the advanced interactive friction modelling has been established for different application scenarios,including lubricated condition,dry sliding condition(metal-on-metal contact),and coated system.

The Solar Polar-orbit Observatory(SPO),proposed by Chinese scientists,is designed to observe the solar polar regions in an unprecedented way with a spacecraft traveling in a large solar inclination angle and a small ellipticity.However,one of the most significant challenges lies in ultra-long-distance data transmission,particularly for the Magnetic and Helioseismic Imager(MHI),which is the most important payload and generates the largest volume of data in SPO.In this paper,we propose a tailored lossless data compression method based on the measurement mode and characteristics of MHI data.The background out of the solar disk is removed to decrease the pixel number of an image under compression.Multiple predictive coding methods are combined to eliminate the redundancy utilizing the correlation(space,spectrum,and polarization)in data set,improving the compression ratio.Experimental results demonstrate that our method achieves an average compression ratio of 3.67.The compression time is also less than the general observation period.The method exhibits strong feasibility and can be easily adapted to MHI.

The role of primary cilia in mechanical transmission of osteocyte based on a 3D finite element model

The primary cilium,as a mechanical receptor of osteocytes,participates in the regulation of osteocyte mechanosensitivity.However,how the length of osteocyte primary cilia changes with fluid shear stress(FSS)are unclear,and how the mechanical transmission within osteocytes altered by primary cilia is not well understood yet.Therefore,the ciliary length changes of osteocyte under 15dyn/cm2 of FSS were experimentally detected,and then 3D finite element models of osteocyte primary cilia containing the basal body and axoneme were built.The results showed that(1)The ciliary length of the CON group,FSS 1h,and FSS 6h were 3.71±1.34μm,3.79±1.04μm,and 1.24±0.73μm respectively,indicating the different durations of FSS might lead to the adaptive changes of cilium length.The calculations showed(2)when the ciliary length became shorter with the ciliary angle stayed the same,the deformation and stress of the cell membrane and membrane skeleton was increased.However,the deformation and stress of the cilia membrane,basal body,the rotation angles of basal body were decreased,and those of cytoplasm,cytoskeleton,actin cortex and nucleus were also decreased;(3)With the decrease of the ciliary angle,the deformation and stress of the cilia membrane,basal body,as well as the rotation angles of basal body were increased.Those of the cytoplasm,cytoskeleton,actin cortex,and nucleus were also increased except the cell membrane and membrane skeleton.The calculation results suggested the length and angle of the primary cilia,the deformation and stress of intracellular structures in osteocyte were altered with ciliary basal body,indicated the connection between the basal body and cytoskeleton may be a key factor that affected the mechanical transport in osteocytes across the cell membrane.This finally promoted the adaptive change of ciliary length under FSS.

Macroautophagy(referred to as autophagy hereafter)is a major intracellular lysosomal degradation pathway that is responsible for the degradation of misfolded/damaged proteins and organelles.Previous studies showed that autophagy protects against acetaminophen(APAP)-induced injury(AILI)via selective removal of damaged mitochondria and APAP protein adducts.The lysosome is a critical organelle sitting at the end stage of autophagy for autophagic degradation via fusion with autophagosomes.In the present study,we showed that transcription factor EB(TFEB),a master transcription factor for lysosomal biogenesis,was impaired by APAP resulting in decreased lysosomal biogenesis in mouse livers.Genetic loss-of and gain-of function of hepatic TFEB exacerbated or protected against AILI,respectively.Mechanistically,overexpression of TFEB increased clearance of APAP protein adducts and mitochondria biogenesis as well as SQSTM1/p62-dependent non-canonical nuclear factor erythroid 2-related factor 2(NRF2)activation to protect against AILI.We also performed an unbiased cell-based imaging high-throughput chemical screening on TFEB and identified a group of TFEB agonists.Among these agonists,salinomycin,an anticoccidial and antibacterial agent,activated TFEB and protected against AILI in mice.In conclusion,genetic and pharmacological activating TFEB may be a promising approach for protecting against AILI.

Design and application of an autonomous Master Control System for a multi-layer magnetic and helioseismic telescope

With the growing significance of space weather forecasting,multi-layer magnetic and helioseismic telescopes are emerging as a key area of research.However,owing to the diverse operational processes and sophisticated hardware configurations of these devices,there is an urgent need for efficient autonomous observation capabilities.An autonomous Master Control System(MCS)can ensure efficient performance,data consistency,and stability,and the prototype presented here adopts a microservices architecture,breaking down the hardware into multiple subsystems and converting their functions into individual services.A central decision-making system leads the operations,supported by three auxiliary systems and three device control systems.Through inter-subsystem service calls,it achieves rapid imaging and spectroscopic monitoring.To verify system stability and observation efficiency,the system was tested on the Solar Full-disk Multi-layer Magnetograph.Experimental results verify this system can operate automatically for 4 consecutive months,acquire photospheric vector magnetic and Doppler velocity fields within a 15-minute interval,and measure chromospheric longitudinal magnetic and Doppler velocity fields in under 180 s.This ensures consistent and stable solar monitoring and serves as a practical methodological benchmark for the development of similar devices.
