NDUFA8 potentially rescues Wolbachia-induced cytoplasmic incompatibility in Laodelphax striatellus

The endosymbiont Wolbachia manipulates host reproduction by several strategies, one of the most important of which is cytoplasmic incompatibility (CI). CI can be rescued when Wolbachia-infected males mate with females infected with the same Wolbachia strain. However, the potential rescue mechanism of CI in the small brown planthopper Laodelphax striatellus is unclear. In this study, comparative transcriptome analysis was applied to explore the effect of Wolbachia on L. striatellus eggs. A total of 1387 differentially expressed genes were identified. RNA interference of 7 Wolbachia-upregulated key planthopper genes reduced egg reproduction, suggesting that Wolbachia might improve fecundity in L. striatellus by affecting these 7 genes. Suppressing the expression of another upregulated gene, NDUFA8 (encoding NADH dehydrogenase [ubiquinone] 1 α subcomplex subunit 8-like) by RNA interference significantly increased the mortality of early embryos without affecting the number of deposited eggs. Wolbachia infection upregulated the mRNA level of NDUFA8, and dsNDUFA8 treatment of Wolbachia-infected females recreated CI-like symptoms, suggesting that NDUFA8 is associated with the rescue phenotype. Because all L. striatellus populations worldwide are infected with Wolbachia, NDUFA8 is a potential pest control target.

Simiao Wan(SMW)is a traditional Chinese formula,including Atractylodis Rhizoma,Achyranthis Bidentatae Radix,Phellodendri Chinensis Cortex and Coicis Semen at the ratio of 1:1:2:2.It can be used to the treatment of diabetes.However,its bioactive compounds and underlying mechanism are unclear.This study was designed to screen the antilipolytic fraction from SMW and investigate its therapeutic mechanisms on hepatic insulin resistance.Different fractions of SMW were prepared by membrane separation combined with macroporous resin and their antilipolytic activity was screened in fasted mice.The effects of 60%ethanol elution(ESMW)on lipolysis were investigated in 3T3-L1 adipocytes stimulated by palmitic acid(PA)and high fat diet(HFD)-fed mice.In our study,ESMW was the bioactive fraction responsible for the antilipolytic activity of SMW and 13 compounds were characterized from ESMW by UHPLC-QTOF-MS/MS.ESMW suppressed protein kinase A(PKA)-hormone-sensitive lipase(HSL)related lipolysis and increased AMP-activated protein kinase(AMPK)phosphorylation in PA challenged 3T3-L1 adipocytes.AMPKa knockdown abolished the inhibitory effects of ESMW on IL-6 and HSL pSer-660,revealing that the antilipolytic and anti-inflammatory activities of ESMW are AMPK dependent.Furthermore,ESMW ameliorated insulin resistance and suppressed lipolysis in HFD-fed mice.It inhibited diacylglycerol accumulation in the liver and inhibited hepatic gluconeogenesis.Conditional medium collected from ESMW-treated 3T3-L1 cells ameliorated insulin action on hepatic gluconeogenesis in liver cells,demonstrating that the antilipolytic activity contributed to ESMW beneficial effects on hepatic glucose production.In conclusion,ESMW,as the antilipolytic fraction of SMW,inhibited PKA-HSL related lipolysis by activating AMPK,inhibiting diacylglycerol(DAG)accumulation in the liver and thereby improving insulin resistance and hepatic gluconeogenesis.

Animals that live in seasonal environments adjust their reproduction cycle to optimize seasonal forage quality.Giant pandas(Ailuropoda melanoleuca)are seasonal altitudinal migrants that feed on bamboo shoots and leaves with different nutritional quality.However,the importance of bamboo shoots to giant pandas,especially small and isolated populations,is not fully appreciated.Here,we explored whether mating time of giant pandas is shaped by bamboo shoot phenology.We also assessed the intensity of ongoing bamboo shoot harvesting by local communities in 42 giant panda reserves based on questionnaire surveys.Varying intensity and protection levels of bamboo shoot harvesting were found.From these data,we developed a priority ranking scheme of protection areas for this key food resource.Our study showed that pandas time their mating behavior to coincide with bamboo shoot phenology due to the high nutritional demands associated with mating and pregnancy.We also found that bamboo shoots were not well protected in many places.Liangshan,Daxiangling,and xiaoxiangling,containing the most isolated panda populations,were identified as the areas with the most urgent need of protection.Furthermore,equal attention should be paid to the Qiong L-B population,as this region holds considerable potential to serve as a corridor between the Minshan and Qionglai populations.To address the challenges posed by bamboo shoot harvesting,we recommend establishing more practical bamboo shoot management policies,increasing public awareness of bamboo shoot protection,and providing alternative sources of income for local communities.

Background:Abdominal pain is often observed after endoscopic retrograde cholangiopancreatography(ERCP).Few studies have focused on the risk factors of post-ERCP abdominal pain without post-ERCP pancreatitis(PEP).This study aimed to identify risk factors of post-ERCP abdominal pain without PEP and investigate characteristics of the abdominal pain in non-PEP patients.Methods:Data from patients who underwent ERCP from August 2019 to January 2020 were retrospectively collected.Characteristics of the abdominal pain after ERCP were recorded and compared between PEP and non-PEP patients.Multivariate analysis was conducted to identify risk factors of non-PEP abdominal pain.Results:A total of 1295 ERCP procedures were investigated in this study,among which 100(7.72%)patients presented post-ERCP abdominal pain without PEP and 63(4.86%)patients with PEP.Multivariate analysis found 9 risk factors of non-PEP abdominal pain:age≤65 years[odds ratio(OR):1.971],primary ERCP(OR:2.442),dilated extrahepatic bile duct(OR:1.803),no papilla opening(OR:2.095),pancreatic guidewire passages(OR:2.258),white blood cells(WBC)≤6.0×10^(9)/L(OR:1.689),platelet(PLT)≤250×10^(9)/L(OR:2.505),serumγ-glutamyl transferase(γ-GT)≤35 U/L(OR:2.190),and albumin≥40 g/L(OR:1.762).The PEP group had later pain onset,higher pain frequency and longer hospital stay than those of the non-PEP pain group(P0.05).Conclusions:This study indicated that age≤65 years,primary ERCP,dilated extrahepatic bile duct,no papilla opening,pancreatic guidewire passages,lower WBC,lower PLT,normalγ-GT and elevated albumin were independent risk factors for post-ERCP abdominal pain without PEP.The pain occurred earlier in non-PEP patients than in PEP patients.

To investigate the influence of electric parameters and slag system on the solidification quality of electroslag ingot during electroslag remelting,different power supply modes,current strengths and remelting slag systems were used to conduct electroslag remelting experiments on 304L austenitic stainless steel,and the macrostructure of electroslag ingots was analyzed.The results indicate that the depth of the metal pool decreases with the reduction of remelting frequency in the low frequency power supply mode.The effects of different power supply modes,such as low-frequency,direct current straight polarity(DCSP),and direct current reverse polarity(DCRP),on reducing the depth of the metal pool increase in that order.By reducing the remelting current strength in the same power supply mode,the depth of metal pool is reduced.When compared to the binary slag system of 70%CaF2+30%Al2O3,the ternary slag system of 60%CaF2+20%Al2O3+20%CaO is more effective in reducing the depth of the metal pool during remelting.Utilizing the 60%CaF2+20%Al2O3+20%CaO ternary slag system results in a shallower and flatter metal pool,with columnar crystal growth occurring closer to the axial crystal.This effect is observed for both low frequency and direct current(DC)power supply modes.

Objective This study prospectively investigates the association between immunoglobulin G(IgG)N-glycan traits and ischemic stroke(IS) risk.Methods A nested case-control study was conducted in the China suboptimal health cohort study,which recruited 4,313 individuals in 2013–2014. Cases were identified as patients diagnosed with IS, and controls were 1:1 matched by age and sex with cases. Ig G N-glycans in baseline plasma samples were analyzed.Results A total of 99 IS cases and 99 controls were included, and 24 directly measured glycan peaks(GPs) were separated from Ig G N-glycans. In directly measured GPs, GP4, GP9, GP21, GP22, GP23, and GP24 were associated with the risk of IS in men after adjusting for age, waist and hip circumference,obesity, diabetes, hypertension, and dyslipidemia. Derived glycan traits representing decreased galactosylation and sialylation were associated with IS in men(FBG2S2/(FBG2 + FBG2S1 + FBG2S2): odds ratio(OR) = 0.92, 95% confidence interval(CI): 0.87–0.97;G1n: OR = 0.74, 95% CI: 0.63–0.87;G0n: OR =1.12, 95% CI: 1.03–1.22). However, these associations were not found among women.Conclusion This study validated that altered Ig G N-glycan traits were associated with incident IS in men, suggesting that sex discrepancies might exist in these associations.

The Tibet ASγexperiment just reported their measurement of sub-PeV diffuse gamma-ray emission from the Galactic disk,with the highest energy up to 957 TeV.These diffuse gamma rays are most likely the hadronic origin by cosmic ray(CR)interaction with interstellar gas in the galaxy.This measurement provides direct evidence to the hypothesis that the Galactic Cosmic Rays(GCRs)can be accelerated beyond PeV energies.In this work,we try to explain the sub-PeV diffuse gamma-ray spectrum with different CR propagation models.We find that there is a tension between the sub-PeV diffuse gamma-ray and the local CR spectrum.To describe the sub-PeV diffuse gamma-ray flux,it generally requires larger local CR flux than measurement in the knee region.We further calculate the PeV neutrino flux from the CR propagation model.Even all of these sub-PeV diffuse gamma rays originate from the propagation,the Galactic Neutrinos(GNs)only account for less than∼15%of observed flux,most of which are still from extragalactic sources.

Herein,an iron(II)-catalyzed decarboxylative and oxidative decarbonylative cross-coupling ofα-fluoro cinnamic acids with aliphatic aldehydes is presented.This methodology provided a novel and practical strategy for the construction of monofluoroalkenes from abundant and readily available aliphatic aldehydes,which are key functional groups in used in synthetic transformations in the pharmaceutical and agrochemical sciences.

The reduction behavior of iron ore powder by high-volatile coal was investigated,and its kinetic mechanism was clarified.The effect of volatiles in coal on the reduction reaction of iron ore was compared by utilizing a Xinjiang lignite with a high volatile content and its pyrolysis carbon produced by high-temperature pyrolysis to remove volatiles,serving as a reductant.The mass loss and gas composition of the samples during the reduction process were detected using thermo-gravimetric analysis and gas chromatography,and the morphological changes of iron ore powder were observed through scanning electron microscopy.The kinetic parameters of the iron oxide reduction reaction were calculated by the Flynn-Ozawa-Wall method,and the kinetic mechanism of volatile participation in the iron oxide reduction reaction was determined through the Coats-Redfern method.The results indicate that the coupling effect between the high-volatile coal pyrolysis and reduction reactions occurs during the second stage of the entire coupling process,which corresponds to the late stage of coal pyrolysis with a substantial release of H_(2)and CO.The volatiles in coal actively participated in the reduction reaction,reducing the initiation temperature of the reaction by around 200℃.The reduction of iron oxides by high-volatile coal was jointly promoted by the"hydrogen cycle"and"carbon cycle",resulting in a higher reduction extent and metallization rate at the end of the reaction.When high-volatile coal was used as the reductant,the average activation energy for the entire process was 76.5 kJ/mol,a significant decrease compared to the employment of pyrolysis carbon without volatiles as the reductant(1167 kJ/mol).
