Neurotrophic keratopathy is a persistent defect of the corneal epithelium,with or without stromal ulceration,due to corneal nerve deficiency caused by a variety of *** treatment options for neurotrophic keratopathy are *** this study,an ophthalmic solution was constructed from a chitosan-based thermosensitive hydrogel with long-term release of murine nerve growth factor(CTH-mNGF).Its effectiveness was evaluated in corneal denervation(CD)mice and patients with neurotrophic *** the preclinical setting,CTH-mNGF was assessed in a murine corneal denervation ***-mNGF was transparent,thermosensitive,and ensured sustained release of mNGF for over 2. hours on the ocular surface,maintaining the local mNGF concentration around 1.1.0 pg/mL in *** denervation mice treated with CTH-mNGF for 1. days showed a significant increase in corneal nerve area and total corneal nerve length compared with non-treated and CTH treated mice.A subsequent clinical trial of CTH-mNGF was conducted in patients with stage 2.or {1.neurotrophic *** received topical CTH-mNGF twice daily for 8 *** sodium images,Schirmer’s test,intraocular pressure,Cochet-Bonnet corneal perception test,and best corrected visual acuity were *** total,six patients(total of seven eyes)diagnosed with neurotrophic keratopathy were *** 8 weeks of CTH-mNGF treatment,all participants showed a decreased area of corneal epithelial defect,as stained by ***,six out of seven eyes had fluorescence staining scores<***,best corrected visual acuity,intraocular pressure,Schirmer’s test and Cochet-Bonnet corneal perception test results showed no significant *** increase in corneal nerve density was observed by in vivo confocal microscopy after 8 weeks of CTH-mNGF treatment in three out of seven *** study demonstrates that CTH-mNGF is transparent,thermosensitive,and has sustained-release *** effecti

Human-specific insertions play important roles in human phenotypes and *** we reported a 446-bp insertion(Insert-446)in intron 1. of the TBC1.8B gene,located on chromosome X,and traced its origin to a portion of intron 6 of the EBF1.gene on chromosome ***,Insert-446 was present in the human Neanderthal and Denisovans genomes,and was fixed in humans after human-chimpanzee *** have demonstrated that Insert-446 acts as an enhancer through binding transcript factors that promotes a higher expression of human TBC1.8B gene as compared with orthologs in *** addition,over-expression TBC1.8B promoted cell proliferation and migration through“a dual finger”catalytic mechanism(Arg53. and Gln573.in the TBC domain in vitro and knockdown of TBC1.8B attenuated tumorigenesis in *** of Insert-446 prevented cell proliferation and migration in cancer and normal *** results reveal that the human-specific Insert-446 promotes cell proliferation and migration by upregulating the expression of TBC1.8B *** findings provide a significant insight into the effects of human-specific insertions on evolution.

This paper presents an efficient way to render large-scale vector maps on level-of-detail(LOD) digital elevation models(DEMs).By using the frame buffer and Voronoi diagram,we achieve a rapid simplification of large-scale vector maps while keeping their original topological *** the simplified maps,we establish level-of-detail vector map *** the detailed level,we use the stencil shadow volume approach to render the map accurately,and in the coarse level we use substitute techniques to render the map approximately yet more *** method proposed in this paper is universally applicable to rendering vector maps on various LOD terrain models,and the overlaying performance is independent of the complexity of underlying terrain *** use of the view frustum culling and sub-region index techniques,the impact of complexity of vector maps is reduced,and the rendering time is restricted to a certain ***,we can achieve seamless and rapid rendering of large-scale vector maps on LOD terrain surfaces.

The plants in genus Gentianella Moench (Gentianaceae) which comprised approximately 2.0 species, are mainly distributed in temperate regions of the world. Many Gentianella plants are intensely bitter and employed in traditional medicine to stimulate appetite, treat disorders of the gallbladder, and treat fever like the other bitter gentians in various regions of the world. Some species exhibit other remarkable therapeutic effects in the treatments of obesity, diabetes, and heart diseases. Eleven iridoids, twenty-eight xanthones, three C-glucoflavonoids, and eight other compounds have been isolated from the genus. Most of these compounds are associated with antimicrobial, anti-inflammatory, anti-oxidant, hypoglycemic, and antitumor activities, which provide an empirical base for the traditional utilization of the plants in genus Gentinella Moench.

Huangqi (Astragalus membranaceus), a traditional chinese medicine, has been used to ameliorate side effects of cancer chemotherapy in{1.. However, little is known about its molecular mechanisms. Here we show that induction of K562.or HEL cells with 1.5 mg/ml of Huangqi (Hex) (Components extracted from Huangqi) for 3.5 d results in the expression of β-globin gene in both cell lines and leads to terminal differentiation. Moreover, the apoptosis in HEL cells can be induced by increasing concentration of Huangqi (Hex) to 4.5 mg/ml for 3.5 d. Upregulation of Apaf-1. caspase-3.and acetylcholinesterase (AChE) in HEL cells may play a crucial role in the process of apoptosis. The prospect of inducing expression of adult (β) globin gene and apoptosis selectively in cancer cells is obviously attractive from a therapeutic point of view.

Magnetic holes are magnetic depression structures that exist widely in many plasma *** magnetic holes with durations of>1 s in the solar wind at Mercury’s orbit have drawn much attention,but the properties of the magnetic holes with shorter durations are still ***,we investigate the magnetic holes with durations of 0.1-100 s in the upstream region of Mercury’s bow shock based on observations by the MESSENGER(MErcury Surface,Space ENvironment,GEochemistry,and Ranging)*** can be divided into two groups according to the distribution of their duration:small-duration magnetic holes(SDMHs,0.6 s).The duration of each group approximately obeys a log-normal distribution with a median of~0.2. s and 3.s,*** 1.7%(3.0}6%)of the SDMHs(LDMHs)reduce the magnetic field strength by more than 50%.For both groups,some structures have a linear or quasi-linear polarization,whereas others have an elliptical *** magnetic hole events in both groups tend to have a higher rate of occurrence when the interplanetary magnetic field strength is *** occurrence rates are also affected by Mercury’s foreshock,which can increase(decrease)the occurrence rate of the SDMHs(LDMHs).This finding suggests that Mercury’s foreshock might be one source of the SDMHs and that the foreshock can destroy some *** observations suggest that a new group of magnetic holes with durations of<0.6 s exist in the upstream region of Mercury’s bow shock.

Immune checkpoint blockade(ICB)has emerged as a promising immunotherapeutic modality against cancer in the ***,only 1.-3.%of patients respond to ICB,primarily due to poor immunogenicity and insufficient T cell infiltration in solid ***,we presented an approach for high-performance cancer treatment using the programmed cell death protein-1.and programmed cell death ligand-1.PD-1.PD-L1.inhibitor(BMS-{1.{1.-loaded PEGylated graphene oxide(GPi).On the one hand,GPi dissociated tight junctions of vascular endothelial cells(VECs)in tumor,thus promoting the extravasation and intratumoral accumulation of liposomal doxorubicin(LipDox),which then effectively induced immunogenic cell death of tumor *** the other hand,GPi also stimulated VECs to upregulate the expression of cell-cell interaction molecules,such as intercellular cell adhesion molecule-1.and vascular cell adhesion molecule-1.which facilitated the infiltration of T cells in *** acting as a stimulator of VECs,GPi could exert responsive release of BMS-{1.{1.under the acidic tumor microenvironment and blockade PD-1.PD-L1.axis in ***,the alternating administration of GPi and LipDox effectively inhibited tumor growth in a 4T1.tumor model,providing a novel treatment mode for chemo-immunotherapy.

Two-proton emissions from the excited states of 1.,1.Ne, 2.P and 2.,2.S were investigated experimentally by the radioactive beams bombarding on the 1.7Au target via the Coulomb excitation. The complete-kinematics measurements were actualized by the stacks of silicon-strip detectors and CsI+PIN array detectors. The invariant mass of final systems as well as the relative momentum, opening angle, and relative energy of the two emitted protons was reconstructed under the framework of relativ istic kinematics. Visible proton-proton correlations were observed in these systems. The mechanisms of two-proton emission were analyzed in a simple schematic model, in which the extreme decay modes like He cluster emission, three-body 2.phase-space decay, and two-body sequential decay were taken into account. With the help of Monte-Carlo simulations, the two protons emitted from the 6.1. MeV excited state of Ne and the excited states in the energy region of 9.6–1..4 MeV of S, 1. 2. respectively, exhibited prominent features of2.He cluster decay while for the other cases, no obvious diproton emissions were observed.
