Different Aluminum Tolerance among Indica, Japonica and Hybrid Rice Varieties

Hydroponic cultures were conducted to compare the aluminum（Al） tolerance among different rice（Oryza sativa L.） varieties, including indica, japonica and their hybrids. The results showed that the root growth of rice plant was inhibited in different degrees among Al treated varieties. The Al tolerance observed through relative root elongation indicated that five japonica varieties including Longjing 9, Dharial, LGC 1, Ribenyou and Koshihikari were relatively more tolerant than indica varieties. Most indica varieties in this study, such as Aus 373 and 9311（awnless）, were sensitive to Al toxicity. The Al tolerance of most progenies from japonica × indica or indica × japonica crosses was constantly consistent with indica parents. The differences of Al tolerance among Longjing 9（japonica）, Yangdao 6（indica） and Wuyunjing 7（japonica） were studied. Biomass and the malondial-dehyde content of Yangdao 6 under Al exposure decreased and increased, respectively, while there was no significant effect on those of Longjing 9 and Wuyunjing 7. Remarkable reduction of root activities was observed in all these three rice varieties. Significantly higher Al content in roots was found in Yangdao 6 compared to Longjing 9 or Wuyunjing 7.

AIM： To explore the possible mechanisms of curcumin in rat colitis induced by trinitrobenzene sulfonic （TNBS） acid. METHODS： Rats with TNBS acid-induced colitis were treated with curcumin （30 mg/kg or 60 mg/kg per day ip）. changes of body weight and histological scores as well as survival rate were evaluated. Leukocyte infiltration was detected by myeloperoxidase （MPO） activity assay. The expression of cyclooxygenase-2 （COX-2） was detected by RT-PCR and Western blot. Inflammation cytokines were determined by RT-PCR. Local concentration of prostaglandin E2 （PGE2） in colon mucosa was determined by ELISA. RESULTS： Curcumin improved survival rate and histological image, decreased the macroscopic scores and MPO activity. Also curcumin reduced the expression of COX-2 and inflammation cytokines. In addition, treatment with curcumin increased the PGE2 level. CONCLUSION： Curcumin has therapeutic effects on TNBS acid-induced colitis, the mechanisms seem to be related to COX-2 inhibition and PGE2 improvement.

Background The role of tumor-infiltrating lymphocytes （TILs） in the immunopathogenesis of individual cancer is not clear and is a challenge for anti-tumor immunotherapy. This study aimed to investigate the effects of interleukin （IL）-18 and -12 on cytotoxic functions of TILs, Methods TILs from postoperative gastric cancer patients were costimulated with IL-18 and IL-12. SGC-7g01 tumor cells were pre-incubated with TILs and subcutaneously injected into BALB/C SCID mice. The function of TILs was evaluated by measuring tumor sizes in tumor-bearing mice, T helper （Th）l （tumor necrosis factor （TNF）-a, interferon （IFN）-y） and Th2 cytokine levels （IL-10 and IL-4） in serum and cytotoxicity of mouse natural killer （NK） and CD8＋ T cells. Results IL-18 and IL-12 synergistically inhibited the growth of SGC-7901 cells in vivo and significantly extended the survival rate of SGC-7901-bearing mice （66.7% vs. 13.7%, P 〈0.01）. Moreover, TILs could promote the secretion of TNF-a and IFN-y （（130.34±7.65） vs. （210.63±12.31） pg/ml, P 〈0.01; （14.23±1.97） vs. （30.52±2.12） pg/ml, P 〈0.01）, and downregulate IL-10 and IL-4 secretion （（103.72±11.21）vs. （61.36±5.41） pg/ml, P=0.021; （49.36±4.67）vs. （28.48±3.86） pg/ml, P=0.024）. Conclusion IL-18 and IL-12 can synergistically enhance cytotoxic functions of TILs from human gastric cancer.

Our previous studies have showed that C-C motif chemokine ligand 20(CCL20)advanced tumor progression and enhanced the chemoresistance of cancer cells by positively regulating breast cancer stem cell(BCSC)***,it is unclear whether CCL20 affects breast cancer progression by remodeling the tumor microenvironment(TME).Here,we observed that polymorphonuclear myeloid-derived suppressor cells(PMN-MDSCs)were remarkably enriched in TME of CCL20-overexpressing cancer cell orthotopic allograft ***,CCL20 activated the differentiation of granulocyte-monocyte progenitors(GMPs)via its receptor C-C motif chemokine receptor 6(CCR6)leading to the PMN-MDSC ***-MDSCs from CCL20-overexpressing cell orthotopic allograft tumors(CCL20-modulated PMN-MDSCs)secreted amounts of C-X-C motif chemokine ligand 2(CXCL2)and increased ALDH+BCSCs via activating CXCR2/NOTCH1/HEY1 signaling ***,C-X-C motif chemokine receptor 2(CXCR2)antagonist SB225002 enhanced the docetaxel(DTX)effects on tumor growth by decreasing BCSCs in CCL20high-expressing *** findings elucidated how CCL20 modulated the TME to promote cancer development,indicating a new therapeutic strategy by interfering with the interaction between PMN-MDSCs and BCSCs in breast cancer,especially in CCL20high-expressing breast cancer.

In vitro production of TNFα,IL-6 and sIL-2R in Chinese patients with ulcerative colitis

AIM To determine the tumor necrosis factor alpha (TNFα), interleukin 6 (IL 6) and soluble interleukin 2 receptor (sIL 2r) from peripheral blood mononuclear cells (PBMC) in 25 Chinese patients with ulcerative colitis and 20 healthy controls. METHODS PBMC were isolated by density gradient centrifugation of heparinized blood and cultures for 24 or 48 hours by stimulation with LPS or PHA. TNFα and sIL 2r were measured by ELISA method and IL 6 measured by bioassay. RESULTS TNFα production stimulated by LPS and sIL 2r production by PHA in ulcerative colitis were significantly lower than in healthy controls (TNFα 509(46-7244)ng/L vs 1995(117-18 950)ng/L, P 0 05; sIL 2r 264U/ml±115U/ml vs 236U/ml±139U/ml, P >0 05). IL 6 production by spontaneous release from PBMC in ulcerative colitis group was 109U/ml±94U/ml vs 44U/ml±39U/ml for those in healthy controls, P <0 01. IL 6 stimulated by LPS in ulcerative colitis group was (261U/ml±80U/ml) higher than in healthy controls (102U/ml±54U/ml, P <0 01). No correlation of TNFα, IL 6, sIL 2r production was found to disease activity, disease location and medication. CONCLUSION Cytokine production from PBMC was also disturbed in Chinese patients with ulcerative colitis.

The incidence of colorectal cancer(CRC)is increasing in China,with high ***,we aimed to evaluate the latest clinicopathological features and prognostic value of the KRAS/NRAS/BRAF mutation status in CRC patients in Central *** clinical data of 1549 CRC patients with stage I-IV disease diagnosed at Union Hospital,Tongji Medical College of Huazhong University of Science and Technology from 2015 to 2017 were collected and analyzed ***/NRAS/BRAF mutations were detected by real-time quantitative polymerase chain reaction(q-PCR)in 410 CRC patients,with mutation frequencies of KRAS,NRAS and BRAF of 47.56%,2.93%and 4.15%,*** gene mutation status and clinicopathological characteristics of 410 patients with CRC who underwent qPCR were *** KRAS and BRAF gene mutations were related to the pathological differentiation and number of metastatic lymph *** BRAF gene mutation was also associated with cancer thrombosis in blood *** regression analysis showed that there was no statistically significant difference in the overall survival(OS)between patients with KRAS,NRAS mutants and wild-type CRC patients,while the BRAF gene mutation was negatively correlated with the OS rate of CRC *** is suggested that the BRAF gene mutation may be an independent risk factor for the prognosis of CRC.

Background It is unclear whether catheter ablation(CA)for atrial fibrillation(AF)affects the long-term prognosis in the *** study aims to evaluate the relationship between CA and long-term outcomes in elderly patients with *** Patients more than 75 years old with non-valvular AF were prospectively enrolled between August 2011 and December 2017 in the Chinese Atrial Fibrillation Registry *** who underwent CA at baseline were propensity score matched(1:1)with those who did not receive *** outcome events included all-cause mortality,cardiovascular mortality,stroke/transient ischemic attack(TIA),and cardiovascular *** Overall,this cohort included 571 ablated patients and 571 non-ablated patients with similar characteristics on 18 *** a mean follow-up of 39.75±19.98 months(minimum six months),24 patients died in the ablation group,compared with 60 deaths in the non-ablation group[hazard ratio(HR)=0.49,95%confidence interval(CI):0.30-0.79,P=0.0024].Besides,6 ablated and 29 non-ablated subjects died of cardiovascular disease(HR=0.25,95%CI:0.11-0.61,P=0.0022).A total of 27 ablated and 40 non-ablated patients suffered stroke/TIA(HR=0.79,95%CI:0.48-1.28,P=0.3431).In addition,140 ablated and 194 non-ablated participants suffered cardiovascular hospitalization(HR=0.84,95%CI:0.67-1.04,P=0.1084).Subgroup analyses according to gender,type of AF,time since onset of AF,and anticoagulants exposure in initiation did not show significant *** In elderly patients with AF,CA may be associated with a lower incidence of all-cause and cardiovascular mortality.

采用异丙醇钛[(C4H9O)4Ti]为先驱体配制二氧化钛溶胶,以阳极氧化铝模板(AAO,孔径尺寸200nm,厚度约60μm)直接浸渍和负压辅助浸渍的方法制备二氧化钛纳米管或管棒混杂的阵列结构,所得产物在450℃下进行热处理后,化学刻蚀去除模板.利用FE-SEM,HR-TEM以及XRD对其微观形貌、结构等进行了观测和分析.结果表明,产物450℃热处理后为多晶锐钛矿相.模板法简单易行,制备出的管棒外径取决于模板孔道的尺寸,在负压辅助下可有效提高产物的连续性.
