leptin Modulates the Differentiation of Keratinocytes via Autophagy in Psoriasis Patients with Metabolic Syndrome

Objective: Psoriasis is associated with a high prevalence of metabolic syndrome (MS), and patients with concomitant psoriasis and MS are more severely affected and less responsive to treatment. However, the molecular mechanisms behind these effects are unknown. Recent studies have shown that leptin may serve as a molecular link between psoriasis and MS, suggesting that high leptin concentrations may ex.cerbate psoriasis. However, the molecular mechanism of this effect is still unclear. We aimed to investigate the effect of leptin on autophagy in patients with psoriasis. Methods: From January 2021 to June 2022 in PlA General Hospital, we enrolled 51 patients with psoriasis, including 21 patients with MS and 30 without MS, and 30 healthy controls who had undergone nevus surgery. We measured the epidermal leptin, P62, and lC3B concentrations of patients by immunohistochemistry, and measured the serum leptin concentration by enzyme-linked immunosorbent assay. We then performed correlation analyses to compare these proteins' concentrations between patients with concomitant psoriasis and MS, patients with psoriasis alone, and healthy control groups. Additionally, we performed western blotting after in vitro culture of HaCaT cells with different concentrations of leptin and measured the ex.ression levels of the autophagy markers Beclin1, lC3B, and P62; the differentiation markers K10, K16, and K17; and PI3K/AKT/mTOR signaling pathway-related proteins of HaCat cells. Nex., we transfected ATG5 into HaCaT cells to revert autophagy and used the specific PI3K inhibitor lY294002 to block PI3K/AKT/mTOR signaling. The ex.ression levels of K10, K16, and K17 of HaCat cells were again measured. One-way analysis of variance was used for the comparison of means of multiple samples, and lSD-t post hoc test was used for comparison between the 2 groups. Serum response factor（SRF） is an ancient and evolutionarily conserved member of the MADS box.family of transcriptional regulators that regulates a wide range of genes involved in cell growth,migration,cytoskeletal organization,and ***,we isolated the homolog of the vertebrate SRF in silkworm,Bombyx.mori,referred to as *** of ex.ression profiles showed that BmSRF was widely ex.ressed in silkworm tissues,including the ovary,testis,head,midgut,fat body,wing disc,middle（MSG）,and posterior（PSG） silk glands,but we didn’t detected its ex.ression in anterior（ASG）,epiderm,malpighian tubule and *** the wing development stage from the 5th instar larva to the first day adult,the BmSRF gene ex.ressed persistently and ex.ibited the ex.ression peak at the onset of the *** ex.mine the function of BmSRF in the wing development of silkworm,we performed gene silencing by injected the BmSRF dsRNAs into wing disc on the first day of 5th instar（V1）,and then the treated silkworms were reared under standard conditions and developed to *** of the adults treated by the BmSRF dsRNA showed abnormal phenotype which ex.ibit additional veins and blister on the ***,more sockets and scales were present on the wings in the abnormal *** phenomenon may be resulted from the increased *** is well known that the tracheae which are scabbarded in the veins can transport the ox.gen,and the ox.gen has been shown to play a key role in the formation of *** conclusion,our results demonstrated that BmSRF plays an important role during wing development in *** silencing of BmSRF ex.ression in silkworm produced additional veins and blistered wings.

Virtually coupled train set control subject to space-time separation: A distributed economic MPC approach with emergency braking configuration

The emerging virtual coupling technology aims to operate multiple train units in a Virtually Coupled Train Set (VCTS) at a minimal but safe distance. To guarantee collision avoidance, the safety distance should be calculated using the state-of-the-art space-time separation principle that separates the Emergency Braking (EB) trajectories of two successive units during the whole EB process. In this case, the minimal safety distance is usually numerically calculated without an analytic formulation. Thus, the constrained VCTS control problem is hard to address with space-time separation, which is still a gap in the ex.sting literature. To solve this problem, we propose a Distributed Economic Model Predictive Control (DEMPC) approach with computation efficiency and theoretical guarantee. Specifically, to alleviate the computation burden, we transform implicit safety constraints into ex.licitly linear ones, such that the optimal control problem in DEMPC is a quadratic programming problem that can be solved efficiently. For theoretical analysis, sufficient conditions are derived to guarantee the recursive feasibility and stability of DEMPC, employing compatibility constraints, tube techniques and terminal ingredient tuning. Moreover, we ex.end our approach with globally optimal and distributed online EB configuration methods to shorten the minimal distance among VCTS. Finally, ex.erimental results demonstrate the performance and advantages of the proposed approaches. © 2024 Beijing Jiaotong University

The poor corrosion and wear resistances of Mg alloys seriously limit their potential applications in various *** conventional epox. coating easily forms many intrinsic defects during the solidification process,which cannot provide sufficient *** the current study,we design a double-layer epox. composite coating on Mg alloy with enhanced anti-corrosion/wear properties,via the spin-assisted assembly *** outer layer is functionalized graphene(FG)in waterborne epox. resin(WEP)and the inner layer is Ce-based conversion(Ce)*** FG sheets can be homogeneously dispersed within the epox. matrix.to fill the intrinsic defects and improve the barrier *** Ce film connects the outer layer with the substrate,showing the transition *** corrosion rate of Ce/WEP/FG composite coating is 2131 times lower than that of bare Mg alloy,and the wear rate is decreased by~90%.The improved corrosion resistance is attributed to the labyrinth effect(hindering the penetration of corrosive medium)and the obstruction of galvanic coupling *** synergistic effect derived from the FG sheet and blocking layer ex.ibits great potential in realizing the improvement of multi-functional integration,which will open up a new avenue for the development of novel composite protection coatings of Mg alloys.

This paper describes the development and optimization plans for the China Railway Ex.ress (CR Ex.ress). As a new type of international land transport organization, CR Ex.ress has emerged with the continuous ex.ansion of China toward European investment and trade, and in particular, has ex.anded with the continuous progress of the One Belt and One Road (OBOR) initiative. In addition to improving the service quality of CR Ex.ress, the operating costs must be reduced for developing “smart railways” that serve “smart cities”. We propose a dual-objective-based function mathematical optimization model; the satisfaction of the cargo owner is considered, and the timeliness, transportation capacity, and goods category constraints of CR Ex.ress transportation are designed. Moreover, we present the normalized equivalent method of the two-objective function of the model. Finally, a case study is conducted against the background of certain trains in the western corridor of CR Ex.ress to validate the effectiveness of the model and research methods proposed in this study. © 2024 Beijing Jiaotong University

Food poisoning due to money tree seeds: A case report of tox.c encephalopathy

Background The money tree is a favored indoor potted plant in China. Its seeds resemble chestnuts in both shape and flavor when cooked, which are generally considered non-Tox.c and safe for consumption. Case presentation This report presents the first published case of food poisoning due to the ingestion of large amounts of money tree seeds. A 67-year-old man ex.ibited tox.c encephalopathy symptoms and dyspnea after consuming the seeds for approx.mately 6 months. After empirical mechanical ventilation, therapeutic plasma ex.hange, and other medical interventions, the patient's clinical status improved significantly. He was subsequently discharged from the hospital. Conclusion Consuming a substantial quantity of money tree seeds within a limited timeframe can lead to poisoning, resulting in adverse outcomes, particularly tox.c encephalopathy. Moreover, plasma ex.hange may have some therapeutic effect. © Wolters Kluwer Health, Inc. All rights reserved.

DeepRice: A deep learning and deep feature based classification of Rice leaf disease subtypes

Rice stands as a crucial staple food globally, with its enduring sustainability hinging on the prompt detection of rice leaf diseases. Hence, efficiently detecting diseases when they have already occurred holds paramount importance for solving the cost of manual visual identification and chemical testing. In the recent past, the identification of leaf pathologies in crops predominantly relies on manual methods using specialized equipment, which proves to be time-consuming and inefficient. This study offers a remedy by harnessing Deep learning (Dl) and transfer learning techniques to accurately identify and classify rice leaf diseases. A comprehensive dataset comprising 5932 self-generated images of rice leaves was assembled along with the benchmark datasets, categorized into 9 classes irrespective of the ex.ent of disease spread across the leaves. These classes encompass diverse states including healthy leaves, mild and severe blight, mild and severe tungro, mild and severe blast, as well as mild and severe brown spot. Following meticulous manual labelling and dataset segmentation, which was validated by horticulture ex.erts, data augmentation strategies were implemented to amplify the number of images. The datasets were subjected to evaluation using the proposed tailored Convolutional Neural Networks models. Their performance are scrutinized in conjunction with alternative transfer learning approaches like VGG16, x.eption, ResNet50, DenseNet121, Inception ResnetV2, and Inception V3. The effectiveness of the proposed custom VGG16 model was gauged by its capacity to generalize to unseen images, yielding an ex.eptional accuracy of 99.94%, surpassing the benchmarks set by ex.sting state-of-the-art models.
