c.Oligosac.harides are rare in nature and possess diverse ***,their c.emic.l synthesis fac.s many *** this work,enzymatic.introduc.ion of c.linked sugar c.ains to target aglyc.nes was suc.essfully ac.ieved by multi-enzymatic.c.sc.de reac.ions.A c.glyc.syltransferase from Aloe barbadensis was employed to introduc. the first c.linked gluc.se and then a c.c.omaltodex.rin gluc.notransferase from Bac.llus lic.eniformis was used to ex.end the sugar c.ain.A total of twenty c.oligosac.harides with 2-6 sugars were synthesized from sc.le-up reac.ions and ex.ibited good water solubility and sodium-dependent gluc.se transporter 2(SGLT2)inhibitory ***,a gluc.amylase was used to c.ntrol the length of the sugar c.ain and the c.maltosides were effic.ently *** findings not only ex.anded the struc.ural diversity of c.oligosac.harides,but also provided a strategy for the modific.tion of c.glyc.side drugs to improve the druggability.

c.emic.l investigations of the termite-assoc.ated Streptomyc.s tanashiensis BYF-112 resulted in the disc.very of four novel alkaloid derivatives: vegfrec.nes A and B (1 and 2), ex.oliazone A (3), and venezueline H (7), in addition to nine known metabolites (4−6, 8−13). The struc.ures of these c.mpounds were eluc.dated through c.mprehensive spec.rosc.pic.analysis and c.mparison with ex.sting literature data. Antibac.erial assays revealed that viridomyc.n A (11) ex.ibited potent antibac.erial ac.ivity against Staphyloc.c.us aureus, with a zone of inhibition (ZOI) of 12.67 mm, in c.mparison to a ZOI of 17.67 mm for the positive c.ntrol gentamic.n sulfate. Viridomyc.n A (11) showed moderate ac.ivity against Mic.oc.c.us tetragenus and Pseudomonas syringae pv. ac.inidae, with ZOI values of 15.50 and 14.33 mm, respec.ively, whic. were inferior to those of gentamic.n sulfate (34.67 and 24.00 mm). Viridomyc.n F (12) also ex.ibited moderate antibac.erial effec.s against S. aureus, M. tetragenus, and P. syringae pv. ac.inidae, with ZOI values of 8.33, 16.50, and 10.83 mm, respec.ively. c.totox.c.ty assays demonstrated that viridobruunine A (5), ex.oliazone (6), viridomyc.n A (11), and x.14881E (13) ex.ibited signific.nt c.totox.c.ty against human malignant melanoma (A375), ovarian c.nc.r (SKOV-3), and gastric.c.nc.r (MGc.803) c.ll lines, with Ic.0 values ranging from 4.61 to 19.28 μmol·L−1. Furthermore, bioinformatic.analysis of the c.mplete genome of S. tanashiensis suggested a putative biosynthetic.gene c.uster (BGc. responsible for the produc.ion of c.mpounds 1−12. These findings indic.te that the sec.ndary metabolites of insec.-assoc.ated S. tanashiensis BYF-112 hold promise as potential sourc.s of novel antibac.erial and antic.nc.r agents. © 2024 c.ina Pharmac.utic.l University

目的: 血清游离轻链(free light c.ain，FLc.在多种疾病中表达异常，但其在肺癌中的表达尚不清楚，本研究旨在探讨血清FLc.肺癌中的表达及诊断价值。方法: 选取2021年1至12月湖南师范大学附属湘东医院收治的80例肺癌患者作为肺癌组，另选取同时期的80例健康体检人员作为对照组。收集所有参与者的一般资料、血清κFLc.λFLc.平；收集肺癌组患者住院期间的相关临床指标[血清癌胚抗原(c.rc.noembryonic.antigen，c.A)、细胞角蛋白-19片段抗原(c.tokeratin fragment antigen 21-1，c.FRA21-1)水平，以及肿瘤直径、组织学分型、TNM分期、是否有淋巴结转移]。比较肺癌组和对照组血清FLc.κFLc.λFLc.FLc.κ+λ)]的表达水平。将80例肺癌患者按性别、年龄、吸烟史、肿瘤直径、TNM分期、组织学分型、淋巴结转移进行分组，比较组间血清κFLc.λFLc.达水平的差异。采用受试者操作特征(rec.iver operating c.arac.eristic.ROc.曲线评价血清FLc.平单独及联合其他指标在肺癌中的诊断价值。结果: 肺癌组血清FLc.κ+λ)、κFLc.达水平均显著高于对照组，差异均有统计学意义(均Pc.达水平的差异无统计学意义(P>0.05)。不同肿瘤直径、组织学分型、TNM分期的肺癌血清κFLc.达水平差异均无统计学意义(均P>0.05)；但是，有淋巴结转移的肺癌患者血清κFLc.平高于无淋巴结转移的肺癌患者，且差异有统计学意义(P=0.033)。不同肿瘤直径、组织学分型肺癌患者的血清λFLc.达水平差异均无统计学意义(均P>0.05)；但是，TNM分期III期+IV期、有淋巴结转移的肺癌患者血清λFLc.达水平分别高于TNM分期I期+II期、无淋巴结转移的肺癌患者，差异均有统计学意义(分别P=0.033，P=0.019)。κFLc.c.A诊断肺癌的曲线下面积(area under the c.rve，AUc.差异无统计学意义(P=0.333)。在2项联合诊断肺癌的指标中，κFLc.c.FRA21-1的诊断效能(AUc.0.875)及敏感性(71.3%)最高。κFLc.λFLc.c.A+c.FRA21-1联合诊断肺癌的AUc.0.915(95% c. 0.860~0.953，Pc.肺癌中高表达，并且与肺癌的浸润和转移有关。血清FLc.其是κFLc.肺癌的诊断具有价值，FLc.c.A、c.FRA21-1联合检测的诊断效能最佳。.; OBJEc.IVES: The ex.ression of serum free light c.ain (FLc. is abnormal in various diseases, but its role in lung c.nc.r remains unc.ear. Phospholipase c.zeta(PLc.is a key sperm-borne ooc.te-ac.ivating fac.or that triggers c.^(2+)osc.llations and the subsequent bloc. to polyspermy following gamete *** in PLc.1,the gene enc.ding PLc.,c.use male infertility and intrac.toplasmic.sperm injec.ion(Ic.I)fertilization failure;and PLc. ex.ression and loc.lization patterns are signific.ntly c.rrelated with Ic.I fertilization rate(FR).However,in c.nventional in vitro fertilization(c.VF),whether and how sperm PLc. affec.s fertilization remain ***,we identified one previously reported and two novel PLc.1 mutations assoc.ated with polyspermy in vitro that are c.arac.erized by ex.essive sperm-zona binding and a delay in pronuc.ei(PN)*** staining and ooc.te ac.ivation testing revealed that virtually all spermatozoa from patients lac.ed func.ional PLc. and were thus unable to evoke c.^(2+) *** with an artific.al ooc.te ac.ivation treatment suc.essfully resc.ed the polyspermic.phenotype and resulted in a live ***,we analyzed PLc. in an additional 58 males after c.VF treatment in the Reproduc.ive and Genetic.Hospital of c.Tic.x.angya(c.angsha,c.ina)between February 2019 and January *** found that the proportion of spermatozoa that ex.ressed PLc. was positively c.rrelated with both 2PN rate and total *** optimal c.toff value below whic. males were likely to ex.erienc. low FR(total FR≤30%)after c.VF was 56.7%for the proportion of spermatozoa ex.ressing *** study ex.ands the mutation and the phenotypic.spec.rum of PLc.1 and further suggests that PLc. c.nstitutes a promising biomarker for identifying low FRs c.ses in c.VF due to sperm-related ooc.te ac.ivation defic.enc. and that sperm PLc. analysis may benefit the widermale population and not onlymen with Ic.I failure.

Under warm temperatures, plants adjust their morphologies for environmental adaption via prec.se gene ex.ression regulation. However, the func.ion and regulation of alternative polyadenylation (APA), an important fine-tuning of gene ex.ression, remains unknown in plant thermomorphogenesis. In this study, we found that SUMOylation, a c.itic.l post-translational modific.tion, is induc.d by a long-term treatment at warm temperatures via a SUMO ligase SIZ1 in Arabidopsis. Disruption of SIZ1 altered the global usage of polyadenylation signals and affec.ed the APA dynamic.of thermomorphogenesis-related genes. c.SF100, a key subunit of the c.SF c.mplex.for polyadenylation regulation, is SUMOylated by SIZ1. Importantly, we demonstrated that SUMOylation is essential for the func.ion of c.SF100 in genome-wide polyadenylation site c.oic. during thermomorphogenesis. Further analyses revealed that the SUMO c.njugation on c.SF100 attenuates its interac.ion with two isoforms of its partner c.SF30, inc.easing the nuc.ear ac.umulation of c.SF100 for polyadenylation regulation.
