During the development of diet-induced obesity， the change of energy matebolism is closely related to the function of the circadian clock in mammals. Luteolin (LU)， one of the most common natural flavonoids riched in many edible plants， can ameliorate obesity by activating adipose tissue browning， but its effect on circadian clock in this process remains poorly understood. Here we found that dietary LU improved circadian misalignment of energy expenditure in high-fat diet (HFD)-fed wild-type (WT) mice. Moreover， dietary LU efficiently elevated UCP1 levels in adipose tissue during the dark period， which was similar to the LU-increased hepatic PER2 expressions. Hepatic PPARα/RxRα/FGF21 pathway was rhythmically elevated by dietary LU in HFD-fed WT mice， whereas the promotion was inhibited in Per2-/-mice. Meanwhile， Per2 deletion abolished the effects of dietary LU on adipose tissue browning in HFD-fed mice. Further， LU treatment directly activated PPARα/RxRα/FGF21 signaling in primary cultured hepatocytes from WT mice rather than Per2-/-mice. Taken together， the deletion of the core clock component Per2 impedes LU-induced adipose tissue browning through weakening PPARα/RxRα/FGF21 pathway in mice， providing a new insight into the interplay of energy metabolism and circadian clock for the anti-obesity activity of LU.

Biomolecular condensates or membraneless organelles(MLOs) formed by liquid-liquid phase separation(LLPS) divide intracellular spaces into discrete compartments for specific functions. Dysregulation of LLPS or aberrant phase transition that disturbs the formation or material states of MLOs is closely correlated with neurodegeneration, tumorigenesis, and many other pathological processes. Herein, we summarize the recent progress in development of methods to monitor phase separation and we discuss the biogenesis and function of MLOs formed through phase separation. We then present emerging proof-of-concept examples regarding the disruption of phase separation homeostasis in a diverse array of clinical conditions including neurodegenerative disorders, hearing loss, cancers, and immunological diseases. Finally, we describe the emerging discovery of chemical modulators of phase separation.

目的:通过观察祛湿化瘀方对非酒精性脂肪性肝病(NAFLD)小鼠回肠肠道血管屏障功能的影响,揭示祛湿化瘀方治疗NAFLD的机制。方法:(1)采用高脂高糖(HFHC)饮食模型,C57BL/6j小鼠随机分为正常组,HFHC饮食模型组,祛湿化瘀方低、高剂量组,12周末开始给药,直至18周末;(2)检测小鼠肝组织甘油三酯(TG),血清生化指标;HE染色观察肝组织、回肠病理;油红O染色观察肝组织脂质;免疫荧光观察回肠质膜相关蛋白1(PV-1)的表达;Western Blot检测肝脏LBP和回肠Wnt3a、p-β-catenin、VEGF的表达。结果:与正常组比较,模型组小鼠肝组织TG,血清ALT、AST活性,肝组织NAS评分,血清TG、TC、LDL-C、空腹血糖、空腹胰岛素、HOMA-IR显著升高(P<0.01),祛湿化瘀方干预后各项指标均显著降低(P<0.05,P<0.01)。模型组小鼠肝组织HE染色显示肝细胞轮廓肿胀、形态不规则,脂肪变性、气球样变及炎性细胞浸润多见;回肠HE染色显示回肠组织结构受到破坏,可见绒毛顶部少量细胞脱落;肝组织油红O染色显示肝细胞轮廓变大变圆,细胞质中可见大量脂滴;回肠PV1免疫荧光结果显示PV1表达显著升高(P<0.01);Western Blot结果显示肝脏LBP、回肠Wnt3a、p-β-catenin/β-catenin、VEGF表达显著升高(P<0.05,P<0.01);祛湿化瘀方干预后肝组织、回肠病理均改善,肝细胞中脂滴明显变小,回肠PV1表达显著降低(P<0.05),肝脏LBP、回肠Wnt3a、p-β-catenin/β-catenin、VEGF表达显著降低(P<0.05,P<0.01)。结论:祛湿化瘀方可通过抑制回肠中Wnt3a/β-catenin/VEGF信号通路,下调PV1的表达,保护肠道血管屏障的功能,改善NAFLD。

The herd immunity against SARS-CoV-2 is continuously consolidated across the world during the ongoing ***,the potential function of the nonconserved epitopes in the reverse preexisting cross-reactivity induced by SARS-CoV-2 to other human coronaviruses is not well *** our research,we assessed T cell responses to both conserved and nonconserved peptides shared by SARS-CoV-2 and SARS-CoV,identifying cross-reactive CD8^(+)T cell epitopes using enzyme-linked immunospot and intracellular cytokine staining ***,in vitro refolding and circular dichroism were performed to evaluate the thermal stability of the HLA/peptide ***,single-cell T cell receptor reservoir was analyzed based on tetramer ***,we discovered that cross-reactive T cells targeting SARS-CoV were present in individuals who had recovered from COVID-19,and identified SARS-CoV-2 CD8^(+)T cell epitopes spanning the major structural antigens.T cell responses induced by the nonconserved peptides between SARS-CoV-2 and SARS-CoV were higher and played a dominant role in the cross-reactivity in COVID-19 ***-T cell reactivity was also observed within the identified series of CD8^(+)T cell *** representative immunodominant peptide pairs,although the HLA binding capacities for peptides from SARS-CoV-2 and SARS-CoV were similar,the TCR repertoires recognizing these peptides were *** results could provide beneficial information for the development of peptide-based universal vaccines against coronaviruses.

Obesity is associated with skeletal muscle mass loss and physical dysfunction. Krill oil (KO) has been shown to be beneficial in human health. However， the effect of KO on obesity-induced skeletal muscle atrophy is still unclear. In this study， the male C57BL/6j mice were fed a high-fat diet (HFD) for 12 weeks to induce obesity， and then were intragastric administration with 400 mg/kg bw KO for an additional 6 weeks. The results showed that KO treatment reduced body weight， fat accumulation and serum pro-inflammatory cytokines in HFD-induced obese mice. Importantly， KO treatment attenuated skeletal muscle atrophy in HFD-fed mice， as evidenced by preserving skeletal muscle mass， average myofiber cross-sectional area and grip strength. KO administration also mitigated obesity-induced ectopic lipid deposition and inflammatory response in skeletal muscle. Additionally， KO treatment inhibited the transcriptional activities of nuclear factor-kappa B (NF-κB) p65 and forkhead box O 3a (FoxO3a)， and then down-regulated muscle atrophy F-box (MAFbx) and muscle-specific RING finger protein 1 (MuRF1) protein levels in skeletal muscle from HFD-fed mice. KO administration also improved obesity-induced impaired muscle protein synthesis via activating PI3K/Akt pathway. Furthermore， KO treatment enhanced muscle mitochondrial biogenesis in HFD-induced obese mice via activating PGC-1α pathway. Collectively， KO might be developed as a potential nutritional supplement for the prevention and treatment of obesity-induced skeletal muscle atrophy.
