HSP110 aggravates ischemia-reperfusion injury after liver transplantation by promoting NF-κB pathway

Background:Ischemia-reperfusion injury(IRI)poses a significant challenge to liver transplantation(LT).The underlying mechanism primarily involves overactivation of the immune *** shock protein 110(HSP110)functions as a molecular chaperone that helps stabilize protein ***:An IRI model was established by performing LT on Sprague-Dawley rats,and HSP110 was silenced using ***-eosin staining,TUNEL,immunohistochemistry,ELISA and liver enzyme analysis were performed to assess IRI following *** blotting and quantitative reverse transcription-polymerase chain reaction were conducted to investigate the pertinent molecular ***:Our findings revealed a significant increase in the expression of HSP110 at both the mRNA and protein levels in the rat liver following LT(P<0.05).However,when rats were injected with siRNAHSP110,IRI subsequent to LT was notably reduced(P<0.05).Additionally,the levels of liver enzymes and inflammatory chemokines in rat serum were significantly reduced(P<0.05).Silencing HSP110 with siRNA resulted in a marked decrease in M1-type polarization of Kupffer cells in the liver and downregulated the NF-κB pathway in the liver(P<0.05).Conclusions:HSP110 in the liver promotes IRI after LT in rats by activating the NF-κB pathway and inducing M1-type polarization of Kupffer *** HSP110 to prevent IRI after LT may represent a promising new approach for the treatment of LT-associated IRI.

QTL Analysis of Some Agronomic Traits in Rice Under Different Growing Environments

The culm length, panicle length, spikelets per panicle and panicle exsertion were evaluated by using F2：3 population including 200 lines derived from the cross of indica and japonica Milyang 23/Jileng 1 under five different environments of Beijing （natural normal growing environment）, Kunming （natural cold environment）, Gongzhuling of China （cold water irrigation） and Chuncheon of Korea （natural normal growing environment and cold water irrigation）, and QTLs of these traits were analyzed by using SSR markers. The results showed that 44 QTLs related to these agronomic traits were detected under five different growing environments, and these QTLs have been located on 11 chromosomes except chromosome 9. The QTLs for qCLla, qCL1b, qCL5a, qCL6b, qPLla, qPL3a, qPL6b, qPL6c, qPL7b, qSP8b, qSPlc, qSP11a, qSP12, and qPE1 have been detected under more than two growing environments, and those that were little affected by environments, were stable QTLs. Among them, qCLla, qCLlb, qPLla, qSPlc, and qPE1 explained 24.2-55.2%, 22.7-39.9%, 12.5-27.7%, 14.4-33.5%, and 26.6-28.7% of observed phynotypic variation, respectively, which were major genes mainly appearing as overdominance. These QTLs cause the increase in action to culm length, panicle length, spikelets per panicle, and panicle exsertion under cold environment, showing that these QTLs were correlated with cold tolerance.

THREE-DIMENSIONAL COMPUTED TOMOGRAPHY-GUIDED RADIOFREQUENCY TRIGEMINAL RHIZOTOMY FOR TREATMENT OF IDIOPATHIC TRIGEMINAL NEURALGIA

Objective To evaluate the effectiveness of three-dimensional computed tomography （3D-CT） guided radiofi＇equency trigeminal rhizotomy （RF-TR） in treatment of idiopathic trigeminal neuralgia （1TN）. Methods From 1999 to 2001, 18 patients with ITN were treated with percutaneous controlled RF-TR. Intraoperative 3D-CT scanning was performed to guide the trajectory of the puncture. After correction of the needle tip according to the CT scans and stimulation effects, 2 to 5 lesions were made for a duration of 60-90 seconds at a temperature of 60℃ to 75℃ depending on the pain distribution and the age of patient. The needles located in foramen ovale. Pain alleviated immediately with no serious complication in all patients. The patients were followed up for an average of 31.5 months （range 24-41 months）. Acute pain relief was experienced by 17 patients after the procedure, reaching an initial success rate of 94.4%. Early （〈 6 months） pain recurrence was observed in 2 patients （11.1%）, whereas late （〉 6 months） recurrence was reported in 3 patients （16.7%）. Thirteen patients had complete pain control, with no need for medication thereafter. Five cases experienced partial pain relief, but required medication at a lower dose than in the preoperative period. Conclusion 3D-CT foramen ovale locations can raise the successful rate of puncture, enhance the safety, and reduce the incidence rate of complication.

Genome-wide identification and analysis of AP2/ERF transcription factors related to camptothecin biosynthesis in Camptotheca acuminata

Camptotheca acuminata produces camptothecin(CPT),a monoterpene indole alkaloid(MIA)that is widely used in the treatment of lung,colorectal,cervical,and ovarian *** biosynthesis pathway has attracted significant attention,but the regulation of CPT biosynthesis by the APETALA2/ethylene-responsive factor(AP2/ERF)transcription factors(TFs)remains *** this study,a systematic analysis of the AP2/ERF TFs family in *** was performed,including phylogeny,gene structure,conserved motifs,and gene expression profiles in different tissues and organs(immature bark,cotyledons,young flower,immature fruit,mature fruit,mature leaf,roots,upper stem,and lower stem)of ***.A total of 198 AP2/ERF genes were identified and divided into five relatively conserved subfamilies,including AP2(26 genes),DREB(61 genes),ERF(92 genes),RAV(18 genes),and Soloist(one gene).The combination of gene expression patterns in different *** tissues and organs,the phylogenetic tree,the co-expression analysis with biosynthetic genes,and the analysis of promoter sequences of key enzymes genes involved in CPT biosynthesis pathways revealed that eight AP2/ERF TFs in *** might be involved in CPT synthesis regulation,which exhibit relatively high expression levels in the upper stem or immature *** these,four genes(Cac AP2/ERF123,Cac AP2/ERF125,Cac AP2/ERF126,and Cac AP2/ERF127)belong to the ERF–B2 subgroup;two genes(Cac AP2/ERF149 and Cac AP2/ERF152)belong to the ERF–B3 subgroup;and two more genes(Cac AP2/ERF095 and Cac AP2/ERF096)belong to the DREB–A6 *** results provide a foundation for future functional characterization of the AP2/ERF genes to enhance the biosynthesis of CPT compounds of ***.

Primary biliary cirrhosis-associated hepatocellular carcinoma in Chinese patients:Incidence and risk factors

AIM: To investigate the incidence, characteristics, and risk factors for hepatocellular carcinoma(HCC) in Chinese patients with primary biliary cirrhosis(PBC).METHODS: We reviewed the data of 52 PB Cassociated HCC patients treated at Beijing 302 Hospital from January 2002 to December 2013 and analyzed its incidence and characteristics between the two genders. The risk factors for PBC-associated HCC were analyzed via a case-control study comprising 20 PBC patients with HCC and 77 matched controls without HCC. The matched factors included gender, age, follow-up period and Child-Pugh scores. Conditional logistic regression was used to evaluate the odds ratios of potential risk factors for HCC development. A P < 0.05 was considered statistically significant. RESULTS: The incidence of HCC in Chinese PBC patients was 4.13%(52/1255) and was significantly higher in the males(9.52%) than in the females(3.31%). Among the 52 PBC patients with HCC, 55.76%(29/52) were diagnosed with HCC and PBC simultaneously, and 5.76%(3/52) were diagnosed with HCC before PBC. The males with PBC-associated HCCwere more likely than the females to have undergone blood transfusion(18.75% vs 8.33%, P = 0.043), consumed alcohol(31.25% vs 8.33%, P = 0.010), smoked(31.25% vs 8.33%, P = 0.010), had a family history of malignancy(25% vs 5.56%, P = 0.012), and had serious liver inflammation, as indicated by the elevated levels of alanine aminotransferase, aspartate aminotransferase, alkaline phosphatase, and γ-glutamyl transpeptidase(P < 0.05). Conditional logistic regression analysis revealed that body mass index(BMI) ≥ 25 [adjusted odds ratio(AOR) = 1.116, 95%CI: 1.002-1.244, P = 0.045] and history of alcohol intake(AOR = 10.294, 95%CI: 1.108-95.680, P = 0.040) were significantly associated with increased odds of HCC development in PBC patients. CONCLUSION: HCC is not rare in Chinese PBC patients. Risk factors for PBC-associated HCC include BMI ≥ 25 and a history of alcohol intake. In addition to regul

Correlations of hypoxia-inducible factor-1α/hypoxia-inducible factor -2α expression with angiogenesis factors expression and prognosis in non-small cell lung cancer

Background Hypoxia-inducible factor （HIF） may play an important role in the process of tumorigenesis as well as tumor progression. The aim of this study was to compare the expression between HIF-1α and HIF-2α in tumor angiogenesis and the overall impact on patient prognosis in human non-small cell lung cancer （NSCLC）. Methods In the current work we compared the immunohistochemical expression of HIF-1α and HIF-21 in surgical specimens of 140 patients with NSCLC in a tissue microarray study. Relationships between HIF-α expression and clinicopathological or angiogenic factors, including prognosis, were analyzed. Results High HIF-1α and HIF-2α expression was noted in 49/140 （35.0%） and in 64/140 （45.7%） of the cases, respectively. There was no direct correlation between HIF-la and HIF-2α expression. Patients with advanced stage tumors had frequent high expression of HIF-2a （P=0.007）, and we also found a significant correlation between HIF-2α and T or N stage （P=0.030 and 0.043, respectively）. HIF-1α showed a marginal association with T stage （P=0.084）, which showed a higher expression in early stage tumors. A significant correlation （p=0.045） was noticed between HIF-1α and vascular endothelial growth factor （VEGF） expression while the expression levels of thymidine phosphorylase （TP）, cyclooxygenase （COX）-2 and microvessel density （MVD） were significantly higher in high HIF-2a tumors （P=0.020, 0.004 and 0.046, respectively）. In addition, univariate analysis of overall survival demonstrated that HIF-2a expression, but not HIF-la, was related to poor outcome （P=-0.001） and it retained significant in multivariate analysis （P=0.036）. Conclusions Taken together, we conclude that HIF-1α and HIF-2α may differentially regulate the major angiogenic factors in different stages of the tumor process in NSCLC. HIF-2α may play a dominant role in tumor angiogenesis and appears to be of obvious value as a significant prognostic factor in NSCLC.

Molecular dynamics has been widely used to study the fundamental mechanism of Ni-based ***,the effect of the potential function and strain rate on mechanical behavior has rarely been mentioned in the previous molecular dynamics *** the present work,we show that the potential function of molecular dynamics can dramatically influence the simulation results of single crystal Ni-based *** microstructure and mechanical behavior of single crystal Ni-based superalloys under four commonly used potential functions are systematically compared.A most suitable potential function for the mechanical deformation is critically selected,and based on it,the role of strain rate on the mechanical deformation is investigated.

The temporal and spatial distribution of ancient rice in China and its implications

The relics of ancient rice have been regarded as the most important objective evidence of the origina- tion and spread of rice *** on the records of 280 rice relics sites and the rice cropping regionalization as well as the distribution map of paddy soils,the current study compiled the temporal and spatial distribution map of ancient rice distribution in *** map shows that the distribution of ancient rice is spatially extensive and meantime comparatively concentrated,temporarily covering a long and relatively continuous *** rice relics in the Central China double and single rice cropping regions are among the earliest and the most abundant ones,possessing continuity in time *** with the discovery of ancient rice and paddy filed relics,soil micromorphology, pollen combination and element geochemistry,it is suggested that Central China was the origin center of rice cultivation in *** had been spread to the rest part of China in three major waves,also to the East Asian part like Korea and *** temporal and spatial distribution of ancient rice reflects the past environmental change,which is also meaningful to the current rice regionalization and plan- ning as well as food security in China.
