Screening and characterization of proteases produced by deep-sea cold seep bacteria

Fifty protease-producing strains were screened from sediment of deep-sea cold seep,and divided into four diff erent categories:Bacillus,Pseudoalteromonas,Vibrio,and Alteromonas according to the sequences of 16s rRNA.Their abilities to produce protease,amylase,and lipase were determined,and a Bacillus strain gcc-1 displayed very strong alkaline protease activity and stability under different thermal and acidic conditions.The purifi cation of the protease produced by strain gcc-1 was carried out by precipitation with ammonium sulfate,and sequentially chromatographed by anion exchange column and gel filtration.The purifi ed protease showed a single band at the molecular weight of 28 kDa by SDS-PAGE.The characterization results show that the purified protease exhibited a considerable activity and stability in a wide thermal range of 10-80℃and a wide acidic range of pH 6.5-11.5,and displayed highest activity at 40℃ and pH 8.5.Notably,the protease still maintained high activity even at low to 10℃.Furthermore,the protease exhibited good stability in presence of diff erent surfactants,organic solvents,oxidizing agent H_(2)O_(2),and commercial detergents.Therefore,the protease produced by gcc-1 is a cold active and high stable enzyme,and has a promising potential in laundry detergent as an additive.

The scientific evaluation of trends in China's future energy demands is highly important.Using provincial-level panel data from 1995 to 2015,we studied the relationships between the economic aggregate,the development of energy-intensive industries and energy demand from the perspective of changes in the proportion of energy-intensive industries in the national economy.We find that economic aggregate affects energy demand through energy-intensive industries and that changes in the economic structure are the key factor for change in energy demand.This means that China's future energy demand will be much lower than that contained in forecasts that did not consider this factor.Comprehensively promoting green-tech development and strengthening the regulation of energy-extensive industries will be one of the key options for realizing China's objective of controlling total energy consumption.

Reliable wave information is critical for marine engineering.Numerical wave models are useful tools to obtain wave information with continuous spatiotemporal distributions.However,the accuracy of model results highly depends on the quality of wind forcing.In this study,we utilize observations from five buoys deployed in the northern South China Sea from August to September 2017.Notably,these buoys successfully recorded wind field and wave information during the passage of five tropical cyclones of different intensities without sustaining any damage.Based on these unique observations,we evaluated the quality of four widely used wind products,namely CFSv2,ERA5,CCMP,and ERAI.Our analysis showed that in the northern South China Sea,ERA5 performed best compared to buoy observations,especially in terms of maximum wind speed values at 10 m height(U10),extreme U10 occurrence time,and overall statistical indicators.CFSv2 tended to overestimate non-extreme U10 values.CCMP showed favorable statistical performance at only three of the five buoys,but underestimated extreme U10 values at all buoys.ERAI had the worst performance under both normal and tropical cyclone conditions.In terms of wave hindcast accuracy,ERA5 outperformed the other reanalysis products,with CFSv2 and CCMP following closely.ERAI showed poor performance especially in the upper significant wave heights.Furthermore,we found that the wave hindcasts did not improve with increasing spatiotemporal resolution,with spatial resolution up to 0.5°.These findings would help in improving wave hindcasts under extreme conditions.

Ilmenite is an essential mineral for the extraction of titanium.Conventional physical separation methods have difficulty recovering fine ilmenite,and dressing plants have begun applying flotation to recover ilmenite.The interaction of reagent groups with Ti and Fe sites on the ilmenite surface dramatically influences the ilmenite flotation.However,the investigation on Fe sites has received more attention because the activity of Ti is lower than that of Fe.For the activators on ilmenite flotation,most are metal ions but typically lead ions.The metal ions of activators promote ilmenite flotation by increasing the active sites on the ilmenite surface.Combined reagents have a better selective separation of ilmenite than single reagents due to their synergistic effect.Combining the lead ion(Pb^(2+))and the benzyl hydroxamic acid(BHA)into a Pb-BHA complex has a marked effect on ilmenite flotation,which puts forward a new idea of developing combined reagents for ilmenite flotation.This review considers reagent types and action mechanisms in ilmenite flotation.On the basis of the analysis of previous research,a brief future outlook of reagent types and action mechanisms in ilmenite flotation is also proposed in this study.

目的探讨基于医护一体化理念的专项管理在输尿管镜碎石术患者中的应用价值。方法选择2019年1月—2022年12月上犹县中医院收治的80例输尿管镜碎石术患者,按随机数字表法分为2组,每组40例。对照组实施常规护理,观察组实施基于医护一体化理念的专项管理,2组均护理至患者出院。对比分析2组术后恢复情况、心理状态、并发症发生情况、护理满意度。结果观察组初次肛门排气时间(25.16±3.92)h、初次下床时间(30.78±4.21)h、拔除导尿管时间(62.85±5.47)h、住院时间(6.97±0.86)d均短于对照组[(32.59±4.87)h、(41.42±4.63)h、(79.71±6.02)h、(7.94±1.04)d,且出院前1 d焦虑自评量表(SAS)评分(42.39±3.12)分、抑郁自评量表(SDS)评分(43.79±2.64)分及并发症发生率(5.00%)低于对照组[(46.54±2.76)分、(48.34±2.15)分、(20.00%)],护理满意度(95.00%)高于对照组(67.50%),差异有统计学意义(P<0.05)。结论基于医护一体化理念的专项管理能够有效改善输尿管镜碎石术患者心理状态,加快术后恢复,且并发症较少,能够获得更高的护理满意度。

本文利用中国家庭能源消费调查(CRECS)数据考察了中国农村家庭的能源阶梯问题。研究发现家庭用能品种会随着收入提升呈现倒U形趋势,并且收入增长会助力于家庭的能源转型。其中,炊事用途随着收入增长能自发地进行能源升级,而"煤改电"政策则限制了收入效应在优化用能结构上的作用。考虑到中国的能源价格黏性,决策者应杜绝"一刀切"式的能源替代政策,将提高能源可支付性的重点放在收入增长上,加强效率瞄准,聚焦于贫困地区和低收入人群上。
