Targeting the Warburg effect:A revisited perspective from molecular mechanisms to traditional and innovative therapeutic strategies in cancer

Cancer reprogramming is an important facilitator of cancer development and survival,with tumor cells exhibiting a preference for aerobic glycolysis beyond oxidative phosphorylation,even under sufficient oxygen supply *** metabolic alteration,known as the Warburg effect,serves as a significant indicator of malignant tumor *** Warburg effect primarily impacts cancer occurrence by influencing the aerobic glycolysis pathway in cancer *** enzymes involved in this process include glucose transporters(GLUTs),HKs,PFKs,LDHs,and ***,the expression of transcriptional regulatory factors and proteins,such as FOXM1,p53,NF-κB,HIF1a,and c-Myc,can also influence cancer ***,lncRNAs,miRNAs,and circular RNAs play a vital role in directly regulating the Warburg ***,gene mutations,tumor microenvironment remodeling,and immune system interactions are closely associated with the Warburg ***,the development of drugs targeting the Warburg effect has exhibited promising potential in tumor *** comprehensive review presents novel directions and approaches for the early diagnosis and treatment of cancer patients by conducting in-depth research and summarizing the bright prospects of targeting the Warburg effect in cancer.

The RUBY reporter for visual selection in soybean genome editing

Current systems to screen for transgenic soybeans(Glycine max)involve laborious molecular assays or the expression of fluorescent markers that are difficult to see in soybean ***,a visual system for early screening of transgenic plants would increase the efficiency of crop improvement by genome *** RUBY reporter system,which consists of three genes encoding betalain biosynthetic enzymes,leading to the accumulation of purple pigment in transgenic tissue,has been employed in some plants and dikaryon ***,we assessed the RUBY reporter for visual verification during soybean *** show that RUBY can be expressed in soybean,allowing for visual confirmation of transgenic events without the need for specialized *** with visible accumulation of purple pigment in any tissue were successfully transformed,confirming the accuracy of the RUBY system as a visual *** also assessed the genetic stability of the transgene across generations,which can be performed very early,using the cotyledons of the ***-free seedlings have a distinct green color,facilitating the selection of genome-edited but transgene-free soybean seedlings for *** the RUBY system,we quickly identified a transgene-free Gmwaxy mutant in the T1 *** system thus provides an efficient and convenient tool for soybean genome editing.

Dielectric nanocomposites with superb high-temperature capacitive performance based on high intrinsic dielectric constant polymer

Advancements in power electronics necessitate dielectric polymer films capable of operating at high temperatures and possessing high energy *** significant strides have been achieved by integrating inorganic fillers into high-temperature polymer matrices,the inherently low dielectric constants of these matrices have tempered the magnitude of *** this work,we report an innovative nanocomposite based on sulfonylated polyimide(SPI),distinguished by the incorporation of sulfonyl groups within the SPI backbone and the inclusion of wide bandgap hafnium dioxide(HfO_(2))*** nanocomposite has demonstrated notable enhancements in thermal stability,dielectric properties,and capacitive performance at elevated *** simulations at both molecular and mesoscopic levels have elucidated the mechanisms behind these improvements,which could be attributed to confined segmental motion,an optimized electronic band structure,and a diminished incidence of dielectric breakdown ascribed to the presence of sulfonyl ***,the SPI-HfO_(2)nanocomposite demonstrates a high charge-discharge efficiency of 95.7%at an elevated temperature of 150℃and an applied electric field of 200 MV/***,it achieves a maximum discharged energy density of 2.71 J/cm^(3),signalling its substantial potential for energy storage applications under extreme conditions.

Clinical Observation on Treatment of Knee Osteoarthritis by Acupuncture and Tuina Therapy

Objective： To observe the effect of acupuncture and chinese tuina manipulation therapy in treating knee osteoarthritis, and analyze the alteration of musculus quadriceps femoris strength. Methods： A total of 80 patients were randomly assigned to a treatment group receiving acupuncture combined chinese tuina manipulation therapy （n=40） and a control group receiving Fenbid treatment （n=40）, curative effective score, Lequene ＆ Mery assessment tests, and isokinetic evaluation were recorded before and after the treatment course. Results： The total effective rate was 90.0 % in the treatment group and 87.5% in the control group, without a significant difference between the two groups. Lequene ＆ Mery assessment tests had no significant change. Musculus quadriceps femoris isokinetic evaluation improved significantly after the treatment course in the two groups （/9〈0.05）, and there was a significant difference between the two groups （P〈0.05）. Conclusion： Acupuncture combined with tuina manipulation therapy and Fenbid treatment both can reduce knee pain and joint disability in the short term, though no improvement was shown in the severity of the disease. Acupuncture combined with tuina manipulation therapy is better than Fenbid in improving the strength, total work and explosive force of the musculus quadriceps femoris.

学术背景:近年来有关中医药治疗膝骨性关节炎的临床报道很多,而有关其治疗机制的实验研究相对滞后。目的:总结膝骨性关节炎时膝关节组织形态及细胞因子的变化以及近10年中医药干预对其的影响。检索策略:应用计算机检索PubMed数据库1985-01/2007-05有关膝骨性关节炎发病机制的文章,检索词"Osteoarthritis",限定文章语言种类为English。同时计算机检索中国期刊全文数据库1997-01/2007-05有关中医药治疗膝骨性关节炎的文章,检索词"膝骨性关节炎",限定文章语言种类为中文。对相关资料汇总进行初审,选取所有与膝骨性关节炎相关的文献,并查找原文。纳入标准:①关于中医药治疗膝骨性关节炎的作用机制。②膝骨性关节炎发病机制的研究进展。排除标准:重复性研究。文献评价:共收集到符合上述要求的文献39篇,其中31篇关于中医药治疗膝骨性关节炎的作用机制,8篇关于膝骨性关节炎发病机制的研究进展。资料综合:中医药治疗膝骨性关节炎有着丰富的临床经验与独特的优势。随着中医药治疗膝骨性关节炎实验研究的不断深入,对于中医药治疗膝关节炎作用机制的认识,已经从简单的组织形态学分析发展到免疫组织化学、分子生物学等多学科、多层次、多环节的认识。近年来,各种实验研究发现中医药对膝骨性关节炎的影响主要表现在膝骨性关节炎的组织形态学、细胞因子、骨内压、氧自由基等方面。结论:中医药治疗膝骨性关节炎的临床方法是多样的,其治疗作用机制也是复杂的,应加强有关中医药防治关节软骨退变的前瞻性研究,更深入地探讨临床中医药治疗本病的作用机制。

HMGA2,a pivotal transcription factor,functions as a versatile regulator implicated in the progression of diverse aggressive *** this study,mass spectrometry was employed to identify ubiquitin-specific proteases that potentially interact with HMGA2,and USP48 was identified as a deubiquitinating enzyme of *** enforced expression of USP48 significantly increased HMGA2 protein levels by inhibiting its degradation,while the deprivation of USP48 promoted HMGA2 degradation,thereby suppressing tumor invasion and *** discovered that USP48 undergoes SUMOylation at lysine 258,which enhances its binding affinity to *** subsequent phenotypic screening of small molecules,we identified DUB-IN-2 as a remarkably potent pharmacological inhibitor of ***,the small-molecule inhibitor targeting USP48 induces destabilization of ***,upregulation of USP48 or HMGA2 in cancerous tissues is indicative of poor prognosis for patients with colorectal cancer(CRC).Collectively,our study not only elucidates the regulatory mechanism of DUBs involved in HMGA2 stability and validates USP48 as a potential therapeutic target for CRC,but also identifies DUB-IN-2 as a potent inhibitor of USP48 and a promising candidate for CRC treatment.

Metal-organic frameworks(MOFs) are highly promising porous materials known for their exceptional porosity, extensive surface area, and customizable pore structures, making them an ideal solution for hydrogen storage. However, most MOFs research remains confined to the laboratory, lacking practical applications. To address this, the author proposes a shift towards practical applications, the creation of a comprehensive MOFs database, alignment of synthesis with practical considerations, and diversification of MOFs applications. These steps are crucial for harnessing the full potential of MOFs in real-world energy challenges.

Regulation of the quantum barrier and carrier transport toward high-efficiency quasi-2D Dion-Jacobson tin perovskite solar cells

Quasi-2D Dion-Jacobson(DJ)tin halide perovskite has attracted much attention due to its elimination of Van der Waals gap and enhanced environmental ***,the bulky organic spacers usually form a natural quantum well structure,which brings a large quantum barrier and poor film quality,further limiting the carrier transport and device ***,we designed three organic spacers with different chain lengths(ethylenediamine(EDA),1,3-propanediamine(PDA),and 1,4-butanediamine(BDA))to investigate the quantum barrier *** and experimental characterizations indicate that EDA with short chain can reduce the lattice distortion and dielectric confinement effect,which is beneficial to the effective dissociation of excitons and the inhibition of trap-free non-radiative *** addition,EDA cation shows strong interaction with the inorganic octahedron,realizing large aggregates in precursor solution and high-quality films with improved structural ***,femtosecond transient absorption proves that EDA cations can also weaken the formation of small n-phases with large quantum barrier to achieve effective carrier transport between different ***,the quasi-2D DJ(EDA)FA_(9)Sn_(10)I_(31)solar cells achieves a 7.07%power conversion efficiency with good environment ***,this work sheds light on the regulation of the quantum barrier and carrier transport through the chain length of organic spacer for qua si-2D DJ lead-free perovskites.
