Background:Keloid scarring is a fibroproliferative disease caused by aberrant genetic activation with an unclear underlying *** predisposition,aberrant cellular responses to environmental factors,increased inflammatory cytokines and epithelial-mesenchymal transition(EMT)phenomena are known as major *** this study,we aimed to identify the molecular drivers that initiate keloid ***:Bulk tissue RNA sequencing analyses of keloid and normal tissues along with ex vivo and in vitro tests were performed to identify the contributing genes to keloid *** animal model of inflammatory keloid scarring was reproduced by replication of a skin fibrosis model with intradermal bleomycin injection in C57BL/6 ***:Gene set enrichment analysis revealed upregulation of Wnt family member 5A(WNT5A)expression and genes associated with EMT in keloid ***,human keloid tissues and the bleomycin-induced skin fibrosis animal model showed significantly increased expression ofWNT5A and EMT *** activation of the interleukin(IL)-6/Janus kinase(JAK)/signal transducer and activator of transcription(STAT)pathway and subsequent elevation of EMT markerswas also observed in keratinocytes co-cultured withWNT5A-activated fibroblasts or keloid ***,WNT5A silencing and the blockage of IL-6 secretion via neutralizing IL-6 antibody reversed hyperactivation of the STAT pathway and EMT markers in ***,STAT3 silencing significantly reduced the EMT-like phenotypes in both keratinocytes and IL-6-stimulated ***:Intercellular communication via the WNT5A and STAT pathways possibly underlies a partial mechanism of EMT-like phenomena in keloid ***-6 secreted from WNT5A-activated fibroblasts or keloid fibroblasts activates the JAK/STAT signaling pathway in adjacent keratinocytes which in turn express EMT markers.A better understanding of keloid development and the

Body-Shaping Membrane to Regenerate Breast Fat by Elastic Structural Holding

Tissue regeneration requires structural holding and movement support using tissue-type-specific aids such as bone casts,skin bandages,and joint ***,an unmet need exists in aiding breast fat regeneration as the breast moves following continuous body motion by exposing the breast fat to dynamic ***,the concept of elastic structural holding is applied to develop a shape-fitting moldable membrane for breast fat regeneration(“adipoconductive”)after surgical defects are *** membrane has the following key characteristics:(a)It contains a panel of honeycomb structures,thereby efficiently handling motion stress through the entire membrane;(b)a strut is added into each honeycomb in a direction perpendicular to gravity,thereby suppressing the deformation and stress concentration upon lying and standing;and(c)thermo-responsive moldable elastomers are used to support structural holding by suppressing large deviations of movement that occur *** elastomer became moldable upon a temperature shift above T_(m).The structure can then be fixed as the temperature *** a result,the membrane promotes adipogenesis by activating mechanotransduction in a fat miniature model with pre-adipocyte spheroids under continuous shaking in vitro and in a subcutaneous implant placed on the motion-prone back areas of rodents in vivo.

Small interfering RNA （siRNA） is an attractive therapeutic candidate for sequence- specific gene silencing to treat incurable diseases using small molecule drugs. However, its efficient intracellular delivery has remained a challenge. Here, we have developed a highly biocompatible fluorescent carbon dot （CD）, and demonstrate a functional siRNA delivery system that induces efficient gene knockdown in vitro and in vivo. We found that CD nanoparticles （NPs） enhance the cellular uptake of siRNA, via endocytosis in tumor cells, with low cytotoxicity and unexpected immune responses. Real-time study of fluorescence imaging in live cells shows that CD NPs favorably localize in cytoplasm and successfully release siRNA within 12 h. Moreover, we demonstrate that CD NP-mediated siRNA delivery significantly silences green fluorescence protein （GFP） expression and inhibits tumor growth in a breast cancer cell xenograft mouse model of tumor-specific therapy. We have developed a multifunctional siRNA delivery vehicle enabling simultaneous bioimaging and efficient downregulation of gene expression, that shows excellent potential for gene therapy.

Enhancing ammonia production rates from electrochemical nitrogen reduction by engineering three-phase boundary with phosphorus-activated Cu catalysts

Electrochemical N_(2) reduction reaction(eNRR) over Cu-based catalysts suffers from an intrinsically low activity of Cu for activation of stable N_(2) molecules and the limited supply of N_(2) to the catalyst due to its low solubility in aqueous ***,we propose phosphorus-activated Cu electrocatalysts to generate electron-deficient Cu sites on the catalyst surface to promote the adsorption of N_(2) *** eNRR system is further modified using a gas diffusion electrode(GDE) coated with polytetrafluoroethylene(PTFE) to form an effective three-phase boundary of liquid water-gas N_(2)-solid catalyst to facilitate easy access of N_(2) to the catalytic *** a result,the new catalyst in the flow-type cell records a Faradaic efficiency of 13.15% and an NH_(3) production rate of 7.69 μg h^(-1) cm^(-2) at-0.2 V_(RHE),which represent 3.56 and 59.2 times increases from those obtained with a pristine Cu electrode in a typical electrolytic *** work represents a successful demonstration of dual modification strategies;catalyst modification and N_(2) supplying system engineering,and the results would provide a useful platform for further developments of electrocatalysts and reaction systems.

Background Community water fluoridation is an effective public health strategy for preventing dental caries,*** exist about potential health *** study explores associations between tap water fluoride levels and pediatric disease burden,as well as neurodevelopmental outcomes at 6 years of *** This nationwide population-based cohort study included children born in Korean cities with and without tap water fluoridation projects,between 2006 and 2012,aiming for a fluoride concentration of 0.8±0.2 mg/L in treated tap *** from the National Health Insurance Service were used,spanning from birth to *** relationship between exposure to fluoridated tap water and incidence of 16 childhood diseases that were previously identified as potentially linked to fluoride exposure were ***,we evaluated the neurodevelopmental outcomes across various domains,including gross motor,fine motor,cognition,language,social skills,and self-help *** assessments were performed using data from a comprehensive national health screening program for children aged six *** A fluoride-unexposed group included 22,881 children,whereas a fluoride-exposed group comprised 29,991 children(52%males).Children in the fluoride-exposed group had a decreased risk of dental caries and bone fractures[hazard ratio(95%confidence interval,CI),0.76(0.63–0.93)and 0.89(0.82–0.93),respectively]and increased risk of hepatic failures[1.85,(1.14–2.98)]compared to those in the unexposed ***,the risk ratio of abnormal neurodevelopmental screening outcomes increased by 9%,but this was statistically uncertain(95%CI,0.95–1.26).Conclusions Fluoridated tap water was associated with an increased risk of hepatic failure but a decreased risk of bone fractures in *** association between fluoridated tap water and neurodevelopmental screening outcomes at 6 years remains unclear,highlighting the need for further studies to clarify this associ

^(27)Al MAS-NMR study of inorganic polymer formation at ambient temperature

Inorganic polymers are a novel class of materials formed by the polymerization of silicon, aluminium and oxygen species to form an amorphous three-dimensional framework structure. The basis of this process is the alkaline solutions to induce a certain amount of Si and AI atoms to dissolve from a feedstock such as aluminosilicate. A study of 27A1 MAS-NMR was carried out in an attempt to understand the reaction mechanism of the inorganic polymerization at ambient temperature. Scanning electron microscopy （SEM） and X-ray diflYactometry （XRD） were also employed to establish the composition and microstructure of the inorganic polymerization. Specimens were prepared with different A1/Si mole ratios from the starting materials. The higher the AI content, the more sufficient the AI atoms that can combine with SiO4, and the longer the reaction time, the more the bonded Si--O--A1--O polymer structure, and then the higher the A1 content, the fewer the octahedral A1 with a uniform Si--O--A1--O structure in four directions, because four AI atoms are combined with SiO4, resulting in a uniform Si--O--AI--O structure in four directions. The results show that they have an amorphous microstructure.

Difference in extrusion temperature dependences of microstructure and mechanical properties between extruded AZ61 and AZ91 alloys

The effects of extrusion temperature on the microstructure and tensile properties of extruded AZ61 and AZ91 alloys are investigated by subjecting them to hot extrusion at 300 and 400℃.Although the average grain size of the extruded AZ61 alloy slightly increases from 9.5 to 12.6μm with increasing extrusion temperature,its resultant microstructural variation is *** contrast,the average grain size of the extruded AZ91 alloy significantly increases from 5.7 to 22.5μm with increasing extrusion temperature,and the type of Mg17Al12 precipitates formed in it changes from fine dynamic precipitates with a spherical shape to coarse static precipitates with a lamellar *** the extrusion temperature increases,the tensile yield strength of the extruded AZ61 alloy increases from 183 to 197 MPa while that of the extruded AZ91 alloy decreases from 232 to 224 *** tensile elongations of the extruded AZ61 and AZ91 alloys decrease with increasing extrusion temperature,but the degree of decrease is significant in the latter *** different extrusion temperature dependences of the tensile properties of the extruded AZ61 and AZ91 alloys are discussed in terms of their microstructural characteristics,strengthening mechanisms,and crack initiation sites.

Utility of a deep learning model and a clinical model for predicting bleeding after endoscopic submucosal dissection in patients with early gastric cancer

BACKGROUND Bleeding is one of the major complications after endoscopic submucosal dissection(ESD)in early gastric cancer(EGC)*** are limited studies on estimating the bleeding risk after ESD using an artificial intelligence *** To derivate and verify the performance of the deep learning model and the clinical model for predicting bleeding risk after ESD in EGC *** Patients with EGC who underwent ESD between January 2010 and June 2020 at the Samsung Medical Center were enrolled,and post-ESD bleeding(PEB)was investigated *** split the entire cohort into a development set(80%)and a validation set(20%).The deep learning and clinical model were built on the development set and tested in the validation *** performance of the deep learning model and the clinical model were compared using the area under the curve and the stratification of bleeding risk after *** A total of 5629 patients were included,and PEB occurred in 325 *** area under the curve for predicting PEB was 0.71(95%confidence interval:0.63-0.78)in the deep learning model and 0.70(95%confidence interval:0.62-0.77)in the clinical model,without significant difference(P=0.730).The patients expected to the low-(leeding rate of 2.2%,3.9%,and 11.6%,respectively,in the deep learning model;4.0%,8.8%,and 18.2%,respectively,in the clinical *** A deep learning model can predict and stratify the bleeding risk after ESD in patients with EGC.

Discharge Characteristics of Large-Area High-Power RF Ion Source for Positive and Negative Neutral Beam Injectors

A large-area high-power radio-frequency（RF） driven ion source was developed for positive and negative neutral beam injectors at the Korea Atomic Energy Research Institute（KAERI）. The RF ion source consists of a driver region, including a helical antenna and a discharge chamber, and an expansion region. RF power can be transferred at up to 10 kW with a fixed frequency of 2 MHz through an optimized RF matching system. An actively water-cooled Faraday shield is located inside the driver region of the ion source for the stable and steady-state operations of high-power RF discharge. Plasma ignition of the ion source is initiated by the injection of argongas without a starter-filament heating, and the argon-gas is then slowly exchanged by the injection of hydrogen-gas to produce pure hydrogen plasmas. The uniformities of the plasma parameter,such as a plasma density and an electron temperature, are measured at the lowest area of the driver region using two RF-compensated electrostatic probes along the direction of the shortand long-dimensions of the driver region. The plasma parameters will be compared with those obtained at the lowest area of the expansion bucket to analyze the plasma expansion properties from the driver region to the expansion region.
