Functional group richness increases multifunctionality in intensively managed grasslands

Background:Agricultural yields have increased continuously over the last few decades.However,a focus solely on production can harm the environment.Diversification of agriculture has been suggested to increase production and sustainability.Biodiversity experiments showed positive effects on ecosystems and productivity.However,application of these results to intensively managed grasslands has been questioned due to differences in plant species and management regimes.Research on whether diversity can benefit multifunctionality,that is,an integrated index of multiple ecosystem functions,under intensive management,is still scarce.Methods:To address this,we manipulated plant species richness from one to six species spanning three functional groups(legumes,herbs,and grasses)in intensively managed multispecies grassland leys and examined seven ecosystem functions.Results:we found that multifunctionality increased with functional group and species richness.Legume+herb mixtures showed high multifunctionality,while grass monocultures and mixtures with high proportions of grasses had low multifunctionality.Different plant species and plant communities drove different ecosystem functions.Legumes and herbs improved productivity and water availability,while grasses enhanced invasion resistance.These results indicate that multifunctionality and individual ecosystem functions can be promoted through targeted combinations of plants with complementary ecological traits.Conclusions:Plant diversity can improve multifunctionality also under intensive management,potentially benefitting agroeconomics and sustainability.

Background:Positive effects of plant species richness on community biomass in biodiversity experiments are often stronger than those from observational field studies.This may be because experiments are initiated with randomly assembled species compositions whereas field communities have experienced filtering.Methods:we compared aboveground biomass production of randomly assembled communities of 2–16 species(controls)with experimentally filtered communities from which subordinate species were removed,resulting in removal communities of 1–8 species.Results:Removal communities had(1)12.6%higher biomass than control communities from which they were derived,that is,with double species richness and(2)32.0%higher biomass than control communities of equal richness.These differences were maintained along the richness gradient.The increased productivity of removal communities was paralleled by increased species evenness and complementarity.Conclusions:Result(1)indicates that subordinate species can reduce community biomass production,suggesting a possible explanation for why the most diverse field communities sometimes do not have the highest productivity.Result(2)suggests that if a community of S species has been derived by filtering from a pool of 2S randomly chosen species it is more productive than a community derived from a pool of S randomly chosen species without filtering.

Background:Forests perform various important ecosystem functions that contribute to ecosystem services.In many parts of the world,forest management has shifted from a focus on timber production to multi-purpose forestry,combining timber production with the supply of other forest ecosystem services.However,it is unclear which forest types provide which ecosystem services and to what extent forests primarily managed for timber already supply multiple ecosystem services.Based on a comprehensive dataset collected across 150 forest plots in three regions differing in management intensity and species composition,we develop models to predict the potential supply of 13 ecosystem services.we use those models to assess the level of multifunctionality of managed forests at the national level using national forest inventory data.Results:Looking at the potential supply of ecosystem services,we found trade-offs(e.***.between both bark beetle control or dung decomposition and both productivity or soil carbon stocks)as well as synergies(e.***.for temperature regulation,carbon storage and culturally interesting plants)across the 53 most dominant forest types in Germany.No single forest type provided all ecosystem services equally.Some ecosystem services showed comparable levels across forest types(e.***.decomposition or richness of saprotrophs),while others varied strongly,depending on forest structural attributes(e.***.phosphorous availability or cover of edible plants)or tree species composition(e.***.potential nitrification activity).Variability in potential supply of ecosystem services was only to a lesser extent driven by environmental conditions.However,the geographic variation in ecosystem function supply across Germany was closely linked with the distribution of main tree species.Conclusions:Our results show that forest multifunctionality is limited to subsets of ecosystem services.The importance of tree species composition highlights that a lack of multifunctionality at the stand level can be compensated by managing forests at the

Trilineage Sequencing Reveals Complex TCRβTranscriptomes in Neutrophils and Monocytes Alongside T Cells

Recent findings indicate the presence of T cell receptor(TCR)-based combinatorial immune receptors beyond T cells in neutrophils and monocytes/macrophages.In this study,using a semiquantitative trilineage immune repertoire sequencing approach as well as under rigorous bioinformatic conditions,we identify highly complex TCRβtranscriptomes in human circulating monocytes and neutrophils that separately encode repertoire diversities one and two orders of magnitude smaller than that of T cells.Intraindividual transcriptomic analyses reveal that neutrophils,monocytes,and T cells express distinct TCRβrepertoires with less than 0.1%overall trilineage repertoire sharing.Interindividual comparison shows that in all three leukocyte lineages,the vast majority of the expressed TCRβvariants are private.we also find that differentiation of monocytes into macrophages induces dramatic individual-specific repertoire shifts,revealing a surprising degree of immune repertoire plasticity in the monocyte lineage.These results uncover the remarkable complexity of the two phagocyte-based flexible immune systems which until now has been hidden in the shadow of T cells.

Overlap of concurrent extrahepatic autoimmune diseases is associated with milder disease severity of newly diagnosed autoimmune hepatitis

Background:Concurrent extrahepatic autoimmune disorders(CEHAID)are frequently observed in autoimmune hepatitis(AIH).It is not clear whether there is any prognostic significance of CEHAID on AIH.The aim of this study was to examine the prognostic impact of CEHAID and the correlation with the disease severity of AIH.Methods:This study included 65 hospitalized subjects who fulfilled the accepted criteria for AIH during an 8-year period(2009–2016).All records were manually screened for presence of associated autoimmune diseases.Disease severity of AIH was assessed by liver laboratory tests including the ratio of aspartate aminotransferase to alanine aminotransferase(AST/ALT)and liver histology.Results:Among the enrolled patients,52(80%)were female(median age 61 years,IQR 45–75).Fifty-six(86.2%)were classified as type-1 AIH.In 26(40%)patients at least one additional extrahepatic autoimmune disease was diagnosed.Thirty-four subjects were referred to our hospital because of acute presentation of AIH(supposed by an acute elevation of hepatic enzymes)for subsequent liver biopsy resulting in initial diagnosis of AIH.This group was stratified into 3 subgroups:(A)AIH alone(n=14);(B)overlap with primary biliary cirrhosis(PBC)/primary sclerosing cholangitis(PSC)(n=11);and(C)with CEHAID(n=9).AST/ALT ratio was the lowest in subgroup C(median 0.64,IQR 0.51–0.94;P=0.023),compared to subgroup A(median 0.91,IQR 0.66–1.10)and subgroup B(median 1.10,IQR 0.89–1.36).Patients with AIH alone showed a trend to the highest grade of fibrosis(mean 2.3;95%CI:1.5–3.0)with no statistical significance compared to subjects with CEHAID(lowest grade of fibrosis;mean 1.5;95%CI:0.2–2.8;P=0.380)whereas the ongoing inflammation was comparable.Conclusions:AST/ALT ratio and extent of fibrosis were lower in subjects with AIH and CEHAID,compared to subjects with only AIH.Therefore,the occurrence of CEHAID might be a predictor for lower disease severity of newly diagnosed acute onset AIH,possibly caused by an earlier diagnosis or different modes of damage.

Chrysomelina beetlesstore 3-nitropropionic acid in form of a pretoxin,isoxazolin-5-one glucoside-conjugated ester,to protect themselves against predators.Here we identified a cytochrome P450 monooxygenase,CYP347w1,to be involved in the production of the 3-nitropropionic acid moiety of the isoxazolin-5-one glucoside ester.Knocking down CYP347w1 led to a significant depletion in the concentration of the isoxazolin-5-one glucoside ester and an increase in the concentration of the isoxazolin-5-one glucoside in the larval hemolymph.Enzyme assays with the heterologously expressed CYP347w1 showed freeβ-alanine was not the direct substrate.Homology modeling indicated thatβ-alanine-CoA ester can fit into CYP347w1’s active site.Furthermore,we proved that Phaedon cochleariae eggs are not able to de novo synthesize 3-NPA,although both isoxazolin-5-one glucoside and its 3-NPA-conjugated ester are present in the eggs.These results provide direct evidence for the involvement of CYP347w1 in the biosynthesis of a P.cochleariae chemical defense compound.

Comprehensive lifestyle intervention vs soy protein-based meal regimen in non-alcoholic steatohepatitis

BACKGROUND Non-alcoholic steatohepatitis(NASH) has become one of the leading causes of liver disease in the western world. In obese patients weight reduction is recommended. Up to now there are no specific guidelines for weight loss in order to reduce hepatic fat content.AIM To investigate the effects of a 24-wk guided lifestyle intervention program compared to a meal replacement regimen based on soy protein.METHODS Twenty-six subjects with NASH participated in a randomized single-center study. They were randomly assigned to either meal replacement group(MR-G)with soy-yogurt-honey preparation or to guided lifestyle change group(LC-G)with endurance activity and nutrition counselling. Serum alanine transaminase(ALT), aspartate transaminase(AST), lipid parameters, and adipokines were measured. Liver fat content and lipid composition were determined by magnetic resonance imaging and magnetic resonance spectroscopy. Body fat mass and lean body mass were assessed using Bod Pod? device. Pre-and post-intervention monitoring of parameters was performed. Statistical analyses were conducted with SPSS software, results were expressed as median(interquartile range).RESULTS Twenty-two subjects(MR-G, n = 11 and LC-G, n = 11) completed the study(9 women, 13 men; age 52.1(15.0) years, body mass index(BMI) 32.3(3.3) kg/m^2).In both groups a significant weight loss was achieved(MR-G:-6.4(3.6) kg, P were markedly reduced in both groups in comparable amount. There was a strong correlation between reduction in liver fat and decrease in ALT.Likewise, both groups showed an improvement in glycemic control and lipid profile. Transjugular intrahepatic portosystemic shunt in liver transplant recipients

AIM:To evaluate the efficacy of transjugular intrahepatic portosystemic shunts (TIPSs) after liver transplantation (LT). METHODS: Between November 1996 and December 2005, 10 patients with severe recurrent hepatitis C virus infection (n=4), ductopenic rejection (n=5) or portal vein thrombosis (n=1) were included in this analysis. Eleven TIPSs (one patient underwent two TIPS procedures) were placed for management of therapy-refractory ascites (n=7), hydrothorax (n=2) or bleeding from colonic varices (n=1). The median time interval between LT and TIPS placement was 15 (4-158) mo. RESULTS: TIPS placement was successful in all patients. The mean portosystemic pressure gradient was reduced from 12.5 to 8.7 mmHg. Complete and partial remission could be achieved in 43% and 29% of patients with ascites. Both patients with hydrothorax did not respond to TIPS. No recurrent bleeding was seen in the patient with colonic varices. Nine of 10 patients died during the study period. Only one of two patients, who underwent retransplantation after the TIPS procedure, survived. The median survival period after TIPS placement was 3.3 (range 0.4-20) mo. The majority of patients died from sepsis with multiorgan failure.CONCLUSION: Indications for TIPS and technical performance in LT patients correspond to those in non-transplanted patients. At least partial control of therapy-refractory ascites and variceal bleeding could be achieved in most patients. Nevertheless, survival rates were disappointing, most probably because of the advanced stages of liver disease at the time of TIPS placement and the high risk of sepsis as a consequence of immunosuppression.

Seasonal size-segregated PM10 and PAH concentrations in a rural area of sugarcane agriculture versus a coastal urban area in Southeastern Florida, USA

Airborne particulate matter （PM） is of health and environmental concern not only in highly urbanized areas, but also in rural areas that are used for intensive agricultural purposes, In this study, PM size- segregated samples were collected simultaneously for 12 months in a small town （Belle Glade, Florida）, which is the center of a vast sugarcane growing area and at Delray Beach, a coastal city in Palm Beach County, Florida. During the winter sampling period, when sugarcane foliage is burned just before harvest- ing to reduce the amount of plant matter to be handled, PM10 levels were 50% or higher than otherwise measured, indicating that sugarcane harvesting and processing is a major local source for PM10. For the rest of the year, PM10 levels at both sites are similar, suggesting that ambient PM levels at both sites are impacted by the major urban centers in Southern Florida. During late July and early August, the PM10 levels at both sites were substantially elevated and revealed the typical red-brownish color of Saharan dust. This has been reported to occur frequently with suitable meteorological conditions over the Atlantic Ocean coupled with a Sahara dust storm event. During the sugarcane harvesting season at Belle Glade, the concentrations of PAHs associated with PM10 were up to 15 times higher than those measured during the summer growing season, indicating a substantially higher exposure of the rural population to these often mutagenic and carcinogenic compounds.
