Single molecule protein sequencing would tremendously impact in proteomics and human biology and it would promote the development of novel diagnostic and therapeutic ***,its technological realization can only be envisioned,and huge challenges need to be *** difficulties are inherent to the structure of proteins,which are composed by several different *** long standing efforts,only few complex techniques,such as Edman degradation,liquid chromatography and mass spectroscopy,make protein sequencing ***,these techniques present significant limitations in terms of amount of sample required and dynamic range of *** is known that proteins can distinguish closely similar ***,several proteins can work as biological nanopores in order to perform single molecule detection and ***,while DNA sequencing by means of nanopores is demonstrated,very few examples of nanopores able to perform reliable protein-sequencing have been reported ***,we investigate,by means of molecular dynamics simulations,how a re-engineered protein,acting as biological nanopore,can be used to recognize the sequence of a translocating peptide by sensing the MshapeH of individual *** our simulations we demonstrate that it is possible to discriminate with high fidelity,9 different amino-acids in a short peptide translocating through the engineered *** method,here shown for fluorescence-based sequencing,does not require any labelling of the peptidic *** results can pave the way for a new and highly sensitive method of sequencing.

We present a 940 nm quasi-continuous wave semiconductor laser designed as a building block for high-power fiber coupled pump *** laser comprises a 400μm narrow-stripe array mounted on an aluminum nitride substrate using hard *** chip has been optimized for high optical power and low lateral far-field *** vertical and six lateral structure variations have been investigated to determine the best achievable *** at 1 ms pulse width and a repetition rate of 10 Hz,the laser device reaches a maximum pulse power of 86 W from a 400μm aperture and more than 62%maximum conversion *** lateral far-field angles(95%power enclosed)of11.5 and 13.5,depending on the epitaxial design,enable efficient multimode fiber *** potential for highly reliable applications has been demonstrated.

Alternatives for large-scale production of cultured beef: A review

Cultured beef is a method where stem cells from skeletal muscle of cows are cultured in vitro to gain edible muscle tissue. For large-scale production of cultured beef, the culture technique needs to become more efficient than today＇s 2-dimensional（2D） standard technique that was used to make the first cultured hamburger. Options for efficient large-scale production of stem cells are to culture cells on microcarriers, either in suspension or in a packed bed bioreactor, or to culture aggregated cells in suspension. We discuss the pros and cons of these systems as well as the possibilities to use the systems for tissue culture. Either of the production systems needs to be optimized to achieve an efficient production of cultured beef. It is anticipated that the optimization of large-scale cell culture as performed for other stem cells can be translated into successful protocols for bovine satellite cells resulting in resource and cost efficient cultured beef.

Influence of solvent mixtures on HPMCAS-celecoxib microparticles prepared by electrospraying

Hypromellose acetate succinate(HPMCAS) microparticles containing the poorly-water soluble drug celecoxib(CEL) were prepared by electrospraying intended for oral drug delivery. Various solvent mixtures with different solubility for CEL and HPMCAS were used to induce changes in the polymer structural conformation of the microparticles. The performance of the prepared microparticles was evaluated by studying the solid state from, particle size and morphology, radial drug distribution and drug release. CEL was amorphous in all electrosprayed HPMCAS microparticles. The particle size and morphology was dependent on the solubility of HPMCAS in the solvent mixture used with poorer solvents resulting in smaller microparticles with rougher appearance. The CEL distribution on the particles surface was relatively homogeneous and similar for all microparticles. Drug release from the microparticles was observed at a higher rate depending on the solubility of HPMCAS in the solvent used for electrospraying, and in all cases an at least 4-fold higher rate was observed compared with the crystalline drug. Drug precipitation from the supersaturated solution was inhibited by HPMCAS for all microparticles based on its parachute effect while crystalline CEL did not reach supersaturation. This study demonstrated that electrospraying can be used to produce microparticles with tailored properties for pharmaceutical application by adjusting solvent selection.

The scientific interest of exercise medicine for the treatment of cancer is ever *** published and updated guidelines for exercise training in cancer patients by the American College of Sports Medicine(ACSM),the Clinical Oncol-ogy Society of Australia(COSA)or the Exercise and Sports Science Australia(ESSA)are leading the way towards an indi-vidualized approach for exercise *** guidelines provide physicians and therapists with a comprehensive and detailed overview about the beneficial effects of exercise training and,more so,summarize the evidence on potential dose-response mechanisms,including pathways of exercise-induced stimuli to counteract tumour microenvironmental ***,the most optimal types and doses of exercise training across the cancer disease and treatment continuum are yet to be ***,the purpose of this narrative review was to illustrate the current implications but also limitations of exercise training during the different stages of cancer therapy,as well as to discuss necessary future *** a second purpose,special attention will be given to the current role of exercise in the treatment of cancer in Germany.

Natural killer(NK)cells participate in early immune defenses against pathogens and tumors and play a major role as immune effector and regulatory *** NK cell-mediated elimination of an infected or cancerous cell is a highly regulated process that requires the formation of a cell contact,the establishment of an immunological synapse and the polarization and release of lytic ***,the detachment of NK cells from target cells is important for NK cells to bind and kill other cells in a process called serial ***,very little is known about this detachment ***,we show that NK detachment is directly connected to the successful killing of a target *** inhibition of killing due to reduced NK cell cytotoxicity or increased target cell resistance results in defective detachment and prolonged contact *** effect leads to sustained Ca^(2+) flux in NK cells and the hypersecretion of proinflammatory *** defective cytotoxicity with enhanced cytokine secretion via reduced detachment may explain inflammatory pathologies in several diseases.

Support for the predictions of the pollinator-mediated stabilizing selection hypothesis

Aims Floral traits are frequently used in traditional plant systematics because of their assumed *** potential reason for the apparent constancy of flower size is that effective pollen transfer between flowers depends on the accuracy of the physical fit between the flower and ***,flowers are likely to be under stronger stabilizing selection for uniform size than vegetative plant ***,as predicted by the pollinator-mediated stabilizing selection(PMSS)hypothesis,an accurate fit between flowers and their pollinators is likely to be more important for specialized pollination systems as found in many species with bilaterally symmetric(zygomorphic)flowers than for species with radially symmetric(actinomorphic)*** In a comparative study of 15 zygomorphic and 13 actinomorphic species in Switzerland,we tested whether variation in flower size,among and within individuals,is smaller than variation in leaf size and whether variation in flower size is smaller in zygomorphic compared to actinomorphic *** findings Indeed,variation in leaf length was significantly larger than variation in flower length and ***-individual variation in flower and leaf sizes did not differ significantly between zygomorphic and actinomorphic *** line with the predictions of the PMSS,among-individual variation in flower length and flower width was significantly smaller for zygomorphic species than for actinomorphic species,while the two groups did not differ in leaf length *** suggests that plants with zygomorphic flowers have undergone stronger selection for uniform flowers than plants with actinomorphic *** supports that the relative uniformity of flowers compared to vegetative structures within species,as already observed in traditional plant systematics,is,at least in part,a consequence of the requirement for effective pollination.

A Population Balance Model for Disperse Systems:Particle Size Distribution in Suspension Polymerization

On the basis of population balance a mathematical model is developed to describe the formation of polymer particle in styrene suspension polymerization. The characteristics of coalescence and breakage of droplets and the gel effect are analyzed in particular. Parameters of the models are estimated by experimental data on reaction conversion and particle size distribution. The results show that the model is suitable for predicting polymerization processes.
