The propagation of small cracks contributes to the majority of the fatigue lifetime for structural *** significant interest,criteria for the growth of small cracks,in terms of the direction and speed of crack advancement,have not yet been *** this work,a new approach to identify the microstructurally small fatigue crack driving force is *** network and machine learning techniques are utilized to identify relevant micromechanical and microstructural variables that influence the direction and rate of the fatigue crack propagation.A multimodal dataset,combining results from a high-resolution 4D experiment of a small crack propagating in situ within a polycrystalline aggregate and crystal plasticity simulations,is used to provide training *** relevant variables form the basis for analytical expressions thus representing the small crack driving force in terms of a direction and a rate *** ability of the proposed expressions to capture the observed experimental behavior is quantified and compared to the results directly from the Bayesian network and from fatigue metrics that are common in the *** indicate that the direction of small crack propagation can be reliably predicted using the proposed analytical model and compares more favorably than other fatigue metrics.

AIM:To investigate a novel phacoemulsification system“EVA NEXUS”(D.O.R.C.,Dutch Opthalmic Research Center)in comparison to the existing system“EVA”in clinical *** to compare both phacoemulsification systems in terms of efficiency,safety and postoperative inflammatory ***:In this study standardized cataract surgery was performed on both eyes of the study participant,using the“EVA system”(control group,n=20)on one eye and the“EVA NEXUS system”(intervention group,n=20)on the other *** patients with cataract LOCS Grading 1-3 and no accompanying eye diseases were included in this study.A total of 20 patients were included in this study,with each treatment arm including 20 *** surgery a 0.1 mL aqueous humor sample was collected 1min after phacoemulsification to measure the total prostaglanin E2 concentrations using an enzyme-linked immunosorbent *** endothelial cell count,visual and refractive outcomes,and anterior chamber flare were evaluated preoperatively,and 1d,1wk,and 3mo ***:There were no statistically significant differences between both groups regarding intraoperative safety parameters including effective phacoemulsification time(P=0.904),balanced saline solution flow(P=0.701)and total surgery time(P=0.565).Postoperative prostaglandin E2 levels,anterior chamber flare as well as endothelial cell loss tended to be lower in the NEXUS-Group,however not being statistically significant(P=0.718;0.164;0.486).Both systems provided similar clinical outcomes,regarding best corrected visual acuity and refractive parameters,showing no statistically significant differences between both ***:Both systems show a high level of safety and efficency with similar results in terms of safety parameters including postoperative inflammatory activity and endothelial cell loss as well as visual and refractive *** statistically not significant,the EVA NEXUS system tends to cause less postoperative inflammatio

Cyanobacteria are promising oxygenic phototrophs for the production of various *** their(photo)biotechnological exploitation,molecular tools are required,such as,for the introduction and expression of heterologous genes,or the modulation of enzyme activities or entire *** and strategies for the development of photosynthetic biomanufacturing technologies based on cyanobacteria have been extensively reviewed,as well as certain specialized aspects of their genetic ***,options for metabolic engineering of specific cyanobacterial cells are still less developed than those for other bacteria of biotechnological *** addition to the standard genetic toolbox for“classical”metabolic engineering,we emphasize certain aspects,including recently developed vector systems for the extrachromosomal maintenance of genes and approaches based on clustered regularly interspaced short palindromic repeats(CRISPR)*** highlight the development of custom molecular tools for specific strains or products,discuss the emerging use of small regulatory proteins that appear promising for advanced metabolic engineering approaches to promote specific product formation,and provide an overview of suitable online ***,we discuss the current trends in this field and indicate their potential,such as using suitable product sensors that enable systematic screening,and optimization approaches.

Univariate and multivariate analysis of risk factors for severe clostridium difficile-associated diarrhoea: Importance of co-morbidity and serum C-reactive protein

AIM: To investigate risk factors for severe clostridium difficile associated diarrhoea (CDAD) in hospitalised patients. METHODS: We analysed risk factors for severe CDAD (associated with systemic signs of hypovolemia) in 124 hospitalised patients by retrospective chart review. RESULTS: Severe CDAD was present in 27 patients (22%). Statistical analysis showed a significant association with a higher 30-d mortality (33% vs 4%, P < 0.001) and a higher proportion of longer hospital stay exceeding 14 d (74% vs 52%, P = 0.048). Charlson co-morbidity score (OR 1.29 for 1 point increment, P < 0.05) and serum C-reactive protein at diagnosis (OR 1.15 for 10 mg/L increment, P < 0.001) were independent predictors of severe CDAD. CONCLUSION: Patients with a severe level of co- morbidity and high serum C-reactive protein levels at the time of diagnosis should receive particular attention.

Frequent concomitant manifestation of type 2 diabetesmellitus (T2DM) and Alzheimer's disease (AD) hasbeen recently demonstrated by epidemiological *** might be due to functional similarities between β-cells and neurons, such as secretion on demand ofhighly specific molecules in a tightly controlled *** additional similarity represents the age-relatedalteration of hyperphosphorylated tau in AD ***, alterations have been identified in β-cells of T2DM patients. The islet amyloid polypeptide has beenassociated with β-cell apoptosis. As a consequence ofincreasing age, the accumulation of highly modified proteins together with decreased regenerative potentialmight lead to increasing rates of apoptosis. Moreover, reduction of β-cell replication capabilities results in reduction of β-cell mass in mammals, simultaneously withimpaired glucose tolerance. The new challenge is tolearn much more about age-related protein modifications. This can lead to new treatment strategies forreducing the incidence of T2DM and AD.

Tissue-engineered constructs are promising to overcome shortage of organ donors and to reconstruct at least partsof injured or diseased tissues or organs. However, oxygen and nutrient supply are limiting factors in many tissues,especially after implantation into the host. Therefore, the development of a vascular system prior to implantationappears crucial. To develop a functional vascular system, different cell types that interact with each other need tobe co-cultured to simulate a physiological environment in vitro. This review provides an overview and a comparison ofthe current knowledge of co-cultures of human endothelial cells (ECs) with human adipose tissue-derived stem/stromal cells (ASCs) or bone marrow-mesenchymal stem cells (BMSCs) in three dimensional (3D) hydrogel *** stem cells (MSCs), BMSCs or ASCs, have been shown to enhance vascular tube formation of ECs and toprovide a stabilizing function in addition to growth factor delivery and permeability control for ECs. Althoughphenotypically similar, MSCs from different tissues promote tubulogenesis through distinct mechanisms. In this report,we describe differences and similarities regarding molecular interactions in order to investigate which of these two celltypes displays more favorable characteristics to be used in clinical applications. Our comparative study shows that ASCsas well as BMSCs are both promising cell types to induce vascularization with ECs in vitro and consequently arepromising candidates to support in vivo vascularization.

Syndesmotic Internal Brace^(TM) for anatomic distal tibiofibular ligament augmentation

Reconstruction of unstable syndesmotic injuries is not trivial, and there is no generally accepted treatment guidelines. Thus, there still remain considerable controversies regarding diagnosis, classification and treatment of syndesmotic injuries. Syndesmotic malreduction is the most common indication for early re-operation after ankle fracture surgery, and widening of the ankle mortise by only 1 mm decreases the contact area of the tibiotalar joint by 42%. Outcome of ankle fractures with syndesmosis injury is worse than without, even after surgical syndesmotic stabilization. This may be due to a high incidence of syndesmotic malreduction revealed by increasing postoperative computed tomography controls. Therefore, even open visualization of the syndesmosis during the reduction maneuver has been recommended. Thus, the most important clinical predictor of outcome is consistently reported as accuracy of anatomic reduction of the injured syndesmosis. In this context the Tight Rope~?system is reported to have advantages compared to classical syndesmotic screws. However, rotational instability of the distal fibula cannot be safely limited by use of 1 or even 2 Tight Ropes~?. Therefore, we developed a new syndesmotic Internal Brace^(TM) technique for improved anatomic distal tibiofibular ligament augmentation to protect healing of the injured native ligaments. The Internal Brace^(TM) technique was developed by Gordon Mackay from Scotland in 2012 using Swive Locks~? for knotless aperture fixation of a Fiber Tape~? at the anatomic footprints of the augmented ligaments, and augmentation of the anterior talofibular ligament, the deltoid ligament, the spring ligament and the medial collateral ligaments of the knee have been published so far.
