Clinical characteristics and outcome of a cohort of 101 patients with hepatocellular carcinoma

AIM: To conduct a cohort study of 101 patients with hepatocellular carcinoma (hCC) presenting to a tertiary care medical referral center in Germany between 1997 and 1999. METhODS AND RESULTS: Data were retrospectively analyzed by chart review. In 95 cases (72 males and 23 females) sufficient data were available for analysis. Twenty five (29%) of 85 patients were hBsAg or anti hBc positive, 21/85 (25%) were anti hCV positive, and 6/85 (7%) were positive for both hBV and hCV-markers. Age was significantly lower in hBV positive patients than in the other two groups. Thirty one (34%) of 90 patients had histories of alcohol abuse. In 79/94 (84%) patients, cirrhosis was diagnosed. Of these cirrhotic patients, 29/79 (37%) belonged to Child Pugh's group (ChILD) A, 32/79 (40%) to ChILD B, and 18/79 (23%) to ChILD C. AFP was elevated in 61/91 (67%) patients. A single tumor nodule was found in 38/94 (40%), more than one nodule in 31/94 (34%), and 25/94 (26%) had a diffusely infiltrating tumor, i.e. the tumor margins could not be seen on imaging procedures. Portal vein thrombosis was present in 19/94 (20%). Imaging data consistent with lymph node metastases were found in 10/92 (11%), while distant metastases were found in 8/93 (9%). According to Okuda 28/94 (30%) were grouped to stage I, 53/94 (56%) were grouped to stage II, and 13/94 (14%) were grouped to stage II. Survival data were available for 83 patients. The Kaplan-Meier estimate for median survival was 8 4 months. Factors influencing survival were the Okuda score, the presence of portal vein thrombosis, and the presence of ascites. The presence of non complicated liver cirrhosis by itself, distant metastases, or infection with hepatitis viruses did not influence survival. AFP positivity by itself did not influence survival, though patients with an AFP value greater than 100 microg/L did experience shortened survival. Treatment of metastatic colorectal carcinomas by systemic inhibition of vascular endothelial growth factor signaling in mice

AIM： Tumor angiogenesis has been shown to be promoted by vascular endothelial growth factor （VEGF） via stimulating endothelial cell proliferation, migration, and survival. Blockade of VEGF signaling by different means has been demonstrated to result in reduced tumor growth and suppression of tumor angiogenesis in distinct tumor entities. here, we tested a recombinant adenovirus, AdsFItl-3, that encodes an antagonistically acting fragment of the VEGF receptor 1 （Fit-l）, for systemic antitumor effects in pre-established subcutaneous CRC tumors in mice.METhODS： Murine colorectal carcinoma cells （CT26） were inoculated subcutaneously into Balb/c mice for in vivo studies. Tumor size and survival were determined. 293 cell line was used for propagation of the adenoviral vectors. human lung cancer line A549 and human umbilical vein endothelial cells were transfected for in vitro experiments.RESULTS： Infection of tumor cells with AdsFlt1-3 resulted in protein secretion into cell supernatant, demonstrating correct vector function. As expected, the secreted sFlt1-3 protein had no direct effect on CT26 tumor cell proliferation in vitro, but endothelial cell function was inhibited by about 46% as compared to the AdLacZ control in a tube formation assay. When AdsFlt1-3 （5×10^9 PFU/animal） was applied to tumor bearing mice, we found a tumor inhibition by 72% at d 12 after treatment initiation, in spite of these antitumoral effects, the survival time was not improved. According to reduced intratumoral microvessel density in AdsFlt1-3-treated mice, the antitumor mechanism can be attributed to angiostatic vector effects. We did not detect increased systemic VEGF levels after AdsFlt1-3 treatment and liver toxicity was low as judged by serum alanine aminotransferase determination.CONCLUSION： In this study we confirmed the value of a systemic administration of AdsFItl-3 to block VEGF signaling as antitumor therapy in an experimental metastatic colorectal carcinoma model in mice.

Integrated Debris Flow Disaster Mitigation—A Comprehensive Method for Debris Flow Disaster Mitigation

Integrated disaster mitigation needs interpreting torrent catchment areas as complex landscape systems. The history of valley-evolution shows the influence of climate and vegetation on the valley-evolution. The energy-concept (energy dissipation concept including the idea of the energy-line) is used for a simple explanation of debris flow. Examples of heavy debris flow disasters in controlled torrents in the Alps and Pyrenees give hints, which expanding the time scale can show that side-effects restrict or counteract the mitigation measures. A pallet of different mitigation measures to avoid or to reduce some of the side-effects is shown. The comprehensive method of disaster mitigation also includes the effect of vegetation. The pallet includes: avoiding hazards (hazard mapping, warning and alarming), appropriate land use and avoiding disaster-enhancing measures in the landscape and technical measures, which take into account their side-effects. The energy line is used as simple design theory. The Jiu-Jitsu Principle is explained too. With this comprehensive method a more sustainable reduction of disasters seems possible.

Simulation Modeling in Plant Breeding: Principles and Applications

Conventional plant breeding largely depends on phenotypic selection and breeder＇s experience, therefore the breeding efficiency is low and the predictions are inaccurate. Along with the fast development in molecular biology and biotechnology, a large amount of biological data is available for genetic studies of important breeding traits in plants, which in turn allows the conduction of genotypic selection in the breeding process. however, gene information has not been effectively used in crop improvement because of the lack of appropriate tools. The simulation approach can utilize the vast and diverse genetic information, predict the cross performance, and compare different selection methods. Thus, the best performing crosses and effective breeding strategies can be identified. QuLine is a computer tool capable of defining a range, from simple to complex genetic models, and simulating breeding processes for developing final advanced lines. On the basis of the results from simulation experiments, breeders can optimize their breeding methodology and greatly improve the breeding efficiency. In this article, the underlying principles of simulation modeling in crop enhancement is initially introduced, following which several applications of QuLine are summarized, by comparing the different selection strategies, the precision parental selection, using known gene information, and the design approach in breeding. Breeding simulation allows the definition of complicated genetic models consisting of multiple alleles, pleiotropy, epistasis, and genes, by environment interaction, and provides a useful tool for breeders, to efficiently use the wide spectrum of genetic data and information available.

The Galactic halo magnetic field revisited

Recently, Sun et al. published new Galactic 3D-models of magnetic fields in the disk and halo of the Milky Way and the distribution of cosmic-ray electron density by taking into account the thermal electron density model NE2001 by Cordes ＆ Lazio. The models successfully reproduce observed continuum and polarization all-sky maps and the distribution of rotation measures of extragalactic sources across the sky. however, the model parameters obtained for the Galactic halo, although reproducing the observations, seem physically unreasonable： the magnetic field needs to be significantly stronger in the Galactic halo than in the plane and the cosmic-ray distribution must be truncated at about 1 kpc to avoid excessive synchrotron emission from the halo. The reason for these unrealistic parameters was the low scale-height of the warm thermal gas of about 1 kpc adopted in the NE2001 model. however, this scale- height seemed reasonable and was well studied by numerous investigations. Recently, the scale-height of the warm gas in the Galaxy was revised by Gaensler et al. to about 1.8 kpc, by showing that the 1 kpc scale-height results from a systematic bias in the analysis of pulsar data. This implies a higher thermal electron density in the Galactic halo, which in turn reduces the halo magnetic field strength to account for the observed rotation measures of extragalactic sources. We slightly modified the NE2001 model according to the new scale-height and revised the Sun et al. model parameters accordingly： the strength of the regular halo magnetic field is now 2 μG or lower, and the physically unrealistic cutoff in z for the cosmic-ray electron density is removed. The simulations based on the revised 3D-models reproduce all-sky observations as before.

Significance of the carbon sink produced by h_2O–carbonate–CO_2–aquatic phototroph interaction on land

One of the most important questions in the science of global change is how to balance the atmospheric CO2 budget. There is a large terrestrial missing carbon sink amounting to about one billion tonnes of carbon per annum. The locations, magnitudes, variations, and mechanisms responsible for this terrestrial missing carbon sink are uncertain and the focus of much continuing debate. Although the positive feedback between global change and silicate chemical weathering is used in geochemical models of atmospheric CO2, this feedback is believed to operate over a long timescale and is therefore generally left out of the current discussion of human impact upon the carbon budget. here, we show, by synthesizing recent findings in rock weathering research and studies into biological carbon pump effects in surface aquatic ecosystems, that the carbon sink produced by carbonate weathering based on the h2O- carbonate-CO2-aquatic phototroph interaction on land not only totals half a billion tonnes per annum, but also displays a significant increasing trend under the influence of global warming and land use change; thus, it needs to be included in the global carbon budget.

自旋翻转和自旋转移是实现基于内嵌富勒体系自旋逻辑功能器件设计的先决条件.本文以双磁性中心内嵌富勒烯Y_2C_2@C8_2-C_2(1)体系为例,采用第一性原理计算方法,结合Λ进程理论模型和自编的遗传算法程序,在该内嵌富勒烯体系中分别实现了亚皮秒时间尺度内的自旋翻转和自旋转移过程.计算结果表明,优化后的内嵌Y_2C_2团簇结构和实验得到的各项数据基本吻合,并且会对外部的C8_2-C_2(1)笼结构产生一定的排斥力,但由于富勒烯笼状结构具有很强的稳定性,所以整个体系仍然保持碳笼结构的完整性.通过对自旋密度分布与激光脉冲作用下自旋期望值演化的具体分析,经由Λ进程的自旋翻转是基于两个Y元素的整体自旋翻转;自旋转移则源自两个磁性中心以及碳笼之间在激光脉冲作用下的自旋密度重新分布.本文结果揭示了Y_2C_2@C8_2-C_2(1)体系中的超快自旋动力学机理,可望为基于实际内嵌富勒烯分子的自旋逻辑功能器件设计提供理论依据.

Modeling and simulation of recurrent phenotypic and genomic selections in plant breeding under the presence of epistasis

Recurrent selection is an important breeding method for population improvement and selecting elite inbreds or fixed lines from the improved ***,a computer simulation tool called QuMARS has been developed,which allows the simulation and optimization of various recurrent selection *** major objective in this study was to use the QuMARS tool to compare phenotypic recurrent,marker-assisted recurrent,and genomic selections(abbreviated respectively as PS,MARS and GS)for both short-and long-termbreeding ***,twomarker selection models were considered,i.e.,stepwise(Rstep)and forward regressions(Forward).For GS,three prediction models were considered,i.e.,genomic best linear unbiased predictors(GBLUP),ridge regression(Ridge),and regression by Moore-Penrose general inverse(InverseMP).To generate genotypes and phenotypes for a given individual during simulation,one additive and two epistasis genetic models were considered with three levels of *** demonstrated that selection responses from GBLUP-based GS and MARS(Forward)were consistently greater than those from PS under the additive model,particularly in early selection *** contrast,selection response from PS was consistently superior over MARS and GS under epistatic *** the two epistasis models,total genetic variance and the additive variance component were increased in some cases after *** simulation,we concluded that GS and PS were effective recurrent selection methods for improved breeding of targeted traits controlled by additive and epistatic quantitative trait loci(QTL).QuMARS provides an opportunity for breeders to compare,optimize and integrate new technology into their conventional breeding programs.
