Serum-circulating His-tRNA synthetase inhibits organ-targeted immune responses

His-tRNA synthetase (HARS) is targeted by autoantibodies in chronic and acute inflammatory anti-Jo-1-positive antisynthetase syndrome. The extensive activation and migration of immune cells into lung and muscle are associated with interstitial lung disease, myositis, and morbidity. It is unknown whether the sequestration of HARS is an epiphenomenon or plays a causal role in the disease. Here, we show that HARS circulates in healthy individuals, but it is largely undetectable in the serum of anti-Jo-1-positive antisynthetase syndrome patients. In cultured primary human skeletal muscle myoblasts (HSkMC), HARS is released in increasing amounts during their differentiation into myotubes. We further show that HARS regulates immune cell engagement and inhibits CD4+ and CD8+ T-cell activation. In mouse and rodent models of acute inflammatory diseases, HARS administration downregulates immune activation. In contrast, neutralization of extracellular HARS by high-titer antibody responses during tissue injury increases susceptibility to immune attack, similar to what is seen in humans with anti-Jo-1-positive disease. Collectively, these data suggest that extracellular HARS is homeostatic in normal subjects, and its sequestration contributes to the morbidity of the anti-Jo-1-positive antisynthetase syndrome.

Minqin County in northwestern China is highly affected by *** have been initiated in recent decades to combat desertification in *** assess the effectiveness of these campaigns,this study used a dense Landsat time series from 1987 to 2017 to investigate the interannual dynamics of vegetation coverage and greenness over the past 31 ***,this study applied an advanced technology to reconstruct a high-quality Landsat annual time ***,one image in the vegetation-peak season was selected as the base image in each year,and then problematic pixels were interpolated by the neighborhood similar pixel interpolator using ancillary images in the same ***,the land cover map and the enhanced vegetation index(EVI)were derived from all reconstructed ***,the change of vegetation coverage and EVI values over the 31 years were *** results show that the total vegetation coverage and greenness increased during the 31 *** this change trend to other factors suggests that vegetation in Minqin County is highly affected by agriculture and groundwater supply rather than by *** mitigate desertification in a sustainable way,agriculture should be well managed to avoid overconsumption of natural resources such as underground water.

Luminescent cyclometalated iridium(Ⅲ) dipyridoquinoxaline indole complexes as biological probes

Four luminescent cyclometalated iridium(Ⅲ) dipyridoquinoxaline complexes appended with an indole moiety [Ir(N^C)2(N^N)] (PF6) (HN^C = 2-phenylpyridine, Hppy; N^N = 2-(N-(2-(indole-3-acetamido)ethyl)aminocarbonyl)dipyrido[3,2-f:2′,3′-h]quinox- aline, dpqC2indole (1a), N^N = 2-(N-(6-(indole-3-acetamido)hexyl)aminocarbonyl)dipyrido[3,2-f:2′,3′-h]quinoxaline, dpqC6indole (1b); HN^C = 7,8-benzoquinoline, Hbzq, N^N = dpqC2indole (2a), N^N = dpqC6indole (2b)) have been synthesized and characterized. Upon irradiation, all the complexes displayed moderately intense and long-lived luminescence under ambient condi-tions and in 77 K glass. On the basis of the photophysical data, the emission of the complexes has been assigned to an excited state of triplet metal-to-ligand charge-transfer (3MLCT) ((dπ(Ir)→π*(N^N)) character. Cyclic voltammetric studies revealed indole-based and iridium-based oxidations at ca. +1.10 V and +1.24 V vs. SCE, respectively, and ligand-based reductions at ca. -1.07 to-2.29 V vs. SCE. The interactions of the complexes with an indole-binding protein, bovine serum albumin (BSA), have been examined by emission titrations.

A Dynamic Forecasting System with Applications in Production logistics

Production logistics involve the co-ordination of ac tivities such as production and materials control (PMC), inventory management, p roduct life cycle management, etc. Those activities demand for an accurate forec asting model. However, the conventional methods of making sell and buy decision based on human forecast or conventional moving average and exponential smoothing methods is no longer be sufficient to meet the future need. Furthermore, the un derlying statistics of the market information change from time to time due to a number of reasons such as change of global economic environment, government poli cies and business risks. This demands for highly adaptive forecasting model which is robust enough to response and adapt well to the fast changes in the dat a characteristics, in other words, the trajectory of the "dynamic characteristic s" of the data. In this paper, an adaptive time-series modelling method was proposed for short -term dynamic forecasting. The method employs an autoregressive (AR) time-seri es model to carry out the forecasting process. A modified least mean square (MLM S) adaptive filter algorithm was established for adjusting the AR model coeffici ents so as to minimise the sum of squared of forecasting errors. A prototype dyn amic forecasting system was built based on the adaptive time-series modelling m ethod. Basically, the dynamic forecasting system can be divided into two phases, i.e. the Learning Phase and the Application Phase. The learning procedures star t with the determination of upper limit of the adaptation gain based on the conv ergence in the mean square criterion. Hence, the optimum ELMS filter parameters are determined using an iteration algorithm which changes each filter parameter i.e. the order, the adaptation gain andthe values initial coefficient vector on e by one inside a predetermined iteration range. The Rice RMR1 Associates with a Distinct Prevacuolar Compartment for the Protein Storage Vacuole Pathway

Transport of vacuolar proteins from Golgi apparatus or trans-Golgi network （TGN） to vacuoles is a receptor-mediated process via an intermediate membrane-bound prevacuolar compartment （PVC） in plant cells. Both vacuolar sorting receptor （VSR） and receptor homology region-transmembrane domain-RING-H2 （RMR） proteins have been shown to function in transporting storage proteins to protein storage vacuole （PSV）, but little is known about the nature of the PVC for the PSV pathway. Here, we use the rice RMR1 （OsRMR1） as a probe to study the PSV pathway in plants. Immunogold electron microscopy （EM） with specific OsRMR1 antibodies showed that OsRMR1 proteins were found in the Golgi apparatus, TGN, and a distinct organelle with characteristics of PVC in both rice culture cells and developing rice seeds, as well as the protein body type II （PBII） or PSV in developing rice seeds. This organelle, also found in both tobacco BY-2 and Arabidopsis suspension cultured cells, is morphologically distinct from the VSR-positive multivesicular lytic PVC or multivesicular body （MVB） and thus represent a PVC for the PSV pathway that we name storage PVC （sPVC）. Further in vivo and in vitro interaction studies using truncated OsRMR1 proteins secreted into the culture media of transgenic BY-2 suspension cells demonstrated that OsRMR1 functions as a sorting receptor in transporting vicilin-like storage proteins.

Prevacuolar compartments （PVCs） and endosomal compartments are membrane-bound organelles mediating protein traffic to vacuoles in the secretory and endocytic pathways of plant cells. Over the years, great progress has been made towards our understanding in these two compartments in plant cells. In this review, we will summarize our contributions toward the identification and characterization of plant prevacuolar and endosomal compartments. Our studies will serve as important steps in future molecular characterization of PVC biogenesis and PVC-mediated protein traffickinq in plant cells.

Background:Data of living-donor liver transplantation(LDLT)suggested that donor ductal anomaly may contribute to postoperative biliary complications in recipients and in *** retrospective study aimed to determine if the occurrence of postoperative biliary stricture in donors or recipients in rightlobe LDLT(RLDLT)is related to donor biliary anatomy ***:We analyzed our RLDLT recipients’clinical data and those of their graft *** recipients were divided into 2 groups:with and without postoperative biliary *** 2 groups were *** primary endpoints were donor biliary anatomy type and postoperative biliary complication incidence;the secondary endpoints were 1-,3-and 5-year graft and patient survival ***:Totally 127 patients were included in the study;25(19.7%)of them developed biliary anastomotic *** these 25 patients,16 had type A biliary anatomy,3 had type B,2 had type C,3 had type D,and 1 had type *** the 127 donors,96(75.6%)had type A biliary anatomy,13(10.2%)had type B,6(4.7%)had type C,10(7.9%)had type D,and 2(1.6%)had type *** stricture was seen in 2 donors,who had type A biliary *** of the recipients or donors developed bile *** association between the occurrence of postoperative biliary stricture and donor biliary anatomy type was found(P=0.527).Conclusions:The incidence of biliary stricture in donors or recipients after RLDLT was not related to donor biliary anatomy *** postoperative complications were similar in whatever type of donor bile duct anatomy,donor ductal anomaly should not be considered a contraindication to donation of right liver lobe.
