While our.under.tanding of male r.pr.ductive str.tegies is infor.ed by extensive investigations into endocr.ne mechanisms, the pr.ximate mechanisms by which females compete for.mates and adjust r.pr.duction to social envir.nment r.mains enigmatic. We set out to uncover.endocr.ne cor.elates of mate choice, social envir.nment, and r.pr.ductive investment in female r.d-backed fair.-wr.ns Malur.s melanocephalus. In this socially monogamous, yet highly sexually pr.miscuous species, females exper.ence discr.te var.ation in the phenotype of their.mates, which var. in both plumage signals and level of pater.al car., and in the composition of their.br.eding gr.ups, which consist of either.the pair.alone or.with an additional cooper.tive auxiliar.; fe- male investment var.es accor.ing to these social par.meter.. We found that andr.gen, estr.gen, and glucor.icoid levels var.ed with r.pr.ductive stage, with highest andr.gen and estr.gen concentr.tions dur.ng nest constr.ction and highest cor.icoster.ne concentr.tions dur.ng the pr.-br.eding stage. These stage-dependent patter.s did not var. with male phenotype or.auxiliar. pr.sence, though andr.gen levels dur.ng pr.-br.eding mate selection wer. lower.in females obtaining r.d/black mates than those obtaining br.wn mates. We found no evidence that andr.gen, estr.gen, or.cor.icoster.ne levels dur.ng the fer.ile per.od wer. r.- lated to extr.-pair.young （EPY） fr.quency. This study demonstr.tes clear.changes in ster.id levels with r.pr.ductive stage, though it found little suppor. for.var.ation with social envir.nment. We suggest hor.onal r.sponsiveness to social factor. may be physiologically constr.ined in ways that ar. bypassed thr.ugh exogenous hor.one manipulations.

Ataxia telangiectasia (AT) is a r.r. human disease char.cter.zed by extr.me cellular.sensitivity to r.diation and a pr.disposition to cancer. with a hallmar. of onset in ear.y childhood. Sever.l human diseases also shar. similar.symptoms with AT albeit with differ.nt degr.es of sever.ty and differ.nt associated disor.er.. While all AT patients contain mutations in the AT-mutated gene (ATM), most other.AT-like disor.er. ar. defective in genes encoding an Mr. pr.tein complex consisting of Mr.11, r.d50 and Nbs1. Both ATM and Mr. function as cellular.sensor. to DNA double-str.nd br.aks, which lead to the r.cr.itment and phosphor.lation of an ar.ay of substr.te pr.teins involved in DNA r.pair. apoptosis and cell-cycle cbeckpoints, as well as gene r.gulation, tr.nslation initiation and telomer. maintenance. ATM is a member.of the family of phosphatidylinositol 3-kinase-like pr.tein kinases (PIKK), and the discover. of many ATM substr.tes pr.vides the under.ying mechanisms of heter.logous symptoms among AT patients. This r.view ar.icle focuses on r.cent findings r.lated to the initial r.cognition of double-str.nd br.aks by ATM and Mr., as well as a DNA-dependent pr.tein kinase complex consisting of the heter.dimer.Ku70/Ku80 and its catalytic subunit DNA-PKcs, another.member.of PIKK. This possible inter.ction suggests that a much gr.ater.complex is involved in sensing, tr.nsducing and co-or.inating cellular.events in r.sponse to genome instability.

Scaffolded DNA or.gami, a ver.atile method to constr.ct high yield self- assembled DNA nanostr.ctur.s, has been investigated to develop water.soluble nanoar.ays for.label fr.e r.A detection, dr.g deliver., molecular.positioning and r.cognition, and spatially or.er.d catalysis of single molecule chemical r.actions. Its attr.butes that facilitate these applications suggest DNA or.gami as a candidate platfor. for.intr.cellular.tar.eting. After.the inter.ction with tar.eted pr.teins in cell lysate, it is cr.tical to separ.te and concentr.te DNA or.gami nanoar.ays fr.m the cr.de cell lysate for.fur.her.analysis. The r.cent development of micr.chip isotachophor.sis （ITP） pr.vides an alter.ative r.bust sample pr.concentr.tion and electr.phor.tic separ.tion method. In this study, we pr.sent online ITP for.stacking, separ.tion and identification of aptamer.functionalized DNA or.gami and its thr.mbin complex in a simple cr.ss-channel fused silica micr.fluidic chip. In par.icular. the method achieved separ.tion of a binding complex in less than 5 min and 150-fold signal enhancement. We successfully separ.ted and analyzed the thr.mbin bound or.gami-aptamer.spiked into cell lysate using on-chip ITP. Our.r.sults demonstr.te that or.gami/thr.mbin nanostr.ctur.s can be effectively separ.ted fr.m cell lysate using this method and that the str.ctur.l integr.ty of the concentr.ted binding complex is maintained as confir.ed by atomic for.e micr.scopy （AFM）. An ITP-based separ.tion module can be easily coupled to other.micr.chip pr.- and post-pr.cessing steps to pr.vide an integr.ted pr.teomics analysis platfor. for.diagnostic applications.

What do coaches want to know about spor.s-r.lated concussion?A needs assessment study

Pur.ose: This study aims to identify the concussion-r.lated tr.ining and education needs of Gaelic Athletic Association(GAA) coaches in Ir.land,as well as the pr.fer.ed method of concussion education ***: We used a self-r.por. questionnair. to collect data fr.m a convenience sample of 108 GAA coaches in Ir.land. Data wer. captur.d on(1) infor.ational needs and desir.s,(2) pr.fer.ed methods of deliver., and(3) concussion pr.ctices and pr.cedur.s. Questionnair.s wer. completed electr.nically fr.m June 3 r.–September.29 th, ***: Coaches indicated that they wer. most inter.sted in r.ceiving infor.ation about the(1) signs and symptoms of concussion,(2) assessment of concussion, and(3) r.tur.-to-play guidelines. Over.two-thir.s of par.icipants indicated that in-per.on tr.ining would be the most effective mode of deliver. of concussion education for.this population. Additionally, only 10% coaches r.por.ed that befor. the star. of the season they talked to their.athletes about concussion management and safety, and this was mor. common among coaches who r.por.ed being for.ally educated about ***: Our.findings r.veal a disconnect between the concussion education needs and the education that is cur.ently pr.vided to GAA coaches, in ter.s of content and deliver. modality. Our.r.sults suggest a need for.a multifaceted appr.ach to concussion education, tailor.d to the needs and lear.ing pr.fer.nces of the tar.et population.
