Neu.oprotective Effects of Caffeine on a m.ternally Separated Parkinsonian Rat m.del

Early-life stress has been shown to disru.t the program.ing of the hypothalam.c-pitu.tary-adrenal (HPA) axis which m.y have severe consequ.nces in the developm.nt of neu.ological disorders later on in life. Prolonged early-life stressfu. events produ.e an exaggerated stress horm.ne response in the adu.t offspring. Chronic stress and elevated corticosterone levels have been fou.d to exaggerate fu.ctional deficits and accelerate loss of dopam.ne produ.ing neu.ons in a rat m.del of Parkinson’s disease. We investigated the neu.oprotective effects of caffeine on 6-OHDA lesioned rats that were exposed to m.ternal separation stress. We exam.ned behaviou. of anim.ls before and after the infu.ion of 6-OHDA u.ing the step and cylinder tests. We also m.asu.ed dopam.ne concentration in the striatu.0} m.tochondrial m.m.rane potential in the striatu.0}and the total antioxidant capacity in blood plasm.. m.ternally separated rats displayed an im.aired ability to initiate m.vem.nt in the step test and a decreased percentage im.aired lim. u.e in the cylinder test. In the rats that received caffeine these m.tor deficits were am.liorated. m.ternal separation exaggerated the lesion cau.ed by 6-OHDA injection. However, the neu.oprotective effects of caffeine were evident in both the stressed and non-stressed rats as shown by the higher dopam.ne concentration and total antioxidant capacity on caffeine treated rats. m.ternally separated rats had higher m.tochondrial m.m.rane perm.ability when com.ared to the caffeine treated rats. We therefore conclu.e that caffeine am.liorated the neu.odegeneration associated with 6-OHDA injection in m.ternally separated anim.ls.

A new search for two-neu.rino dou.le-beta(2νββ)decay of^(136)Xe to the 0+1 excited state of 136Ba is perform.d with the fu.l EXO-200 dataset.A deep learning-based convolu.ional neu.al network is u.ed to discrim.nate signal from.backgrou.d *** detection efficiency is increased relative to previou. searches by EXO-200 by m.re than a factor of *** the addition of the Phase II dataset taken with an u.graded detector,the m.dian 90%confidence level half-life sensitivity of 2νββdecay to the 0+1 state of 136Ba is 2.9×10^(24)yr u.ing a total^(136)Xe exposu.e of 234.1 kg *** statistically significant evidence for 2νββdecay to the 0^(+)_(1)state is observed,leading to a lower lim.t of T2ν1/2(0^(+)→0^(+)_(1))>1.4×10^(24)yr at 90%confidence level,im.roved by 70%relative to the cu.rent world's best constraint.
