Alt.ough met.st.sis-associat.d lung adenocarcinoma t.anscript.(MALAt.-1 is known t. be consist.nt.y upregulat.d in several epit.elial malignancies, lit.le is known about.it. funct.on or regulat.on. We t.erefore examined t.e relat.onship bet.een MALAt.1 expression and candidat. modulat.rs such as DNA t.mor virus oncoprot.ins human papillomavirus (HPV)-16 E6 and E7, BK virus t.ant.gen (BKVt.g), mouse polyoma virus middle t.ant.gen (MPVmt.g) and t.mor suppressor genes p53 and pRb. Using suppressive subt.act.ve hybridizat.on (SSH) and real-t.me reverse t.anscript.se polymerase chain react.on (Rt.PCR) assays, MALAt.1 was shown t. be increased in viral oncongene-expressing salivary gland biopsies from humans and mice. t.e result. also indicat.d t.at.MALAt.1 t.anscript. and promot.r act.vit. were increased in vit.o when viral oncongene-expressing plasmids were int.oduced int. different.cell t.pes. t.ese same viral oncogenes in addit.on t. increasing MALAt.1 t.anscript.on have also been shown t. inhibit.p53 and/or pRb funct.on. In p53 mut.nt.or inact.ve cell lines MALAt.1 was also shown t. be highly upregulat.d. We hypot.esize t.at.t.ere is a correlat.on bet.een MALAt.1 over-expression and p53 deregulat.on. In conclusion, we show t.at.disrupt.on of p53, by bot. polyoma and papilloma oncoprot.ins appear t. play an import.nt.role in t.e up-regulat.on of MALAt.1. MALAt.1 might.t.erefore represent.a biomarker for p53 deregulat.on wit.in malignancies.
