Successful embryo implantation requires highly coordinated maternal-embryo *** failure is a major factor contributing to ***,the mechanism underlying implantation failure remains *** improved understanding of the early implantation process not only improves the success rate of assisted reproductive treatments but also helps in studying the pathophysiology of reproductive *** to ethical concerns,in vivo studies of human embryo implantation are not ***,the results obtained from animal models cannot be directly applied to *** the years,in vitro implantation models have been developed to investigate implantation *** this review,we discuss the use of different models for generating embryo-like surrogates to study early embryo development and implantation in vitro,with a specific focus on stem cell-derived blastocyst-like embryo *** is no definitive evidence that the recently established embryo-like models re-capitulate all developmental events of human embryos during the peri-implantation ***,stem cell-derived embryo surrogates are the most valuable tools for studying the mechanisms of early cell lineage differentiation and developmental failures during implantation.

This review summarizes the drugs and chemicals that may modulate embryo ***-hormonal molecules,including aspirin,improved endometrial blood flow,while low molecular weight heparin,vitamin E,sildenafil,and atosiban modulated the expression of endometrial *** factors,including human chorionic gonadotropin and growth hormones,can regulate the expression of endometrial receptivity *** immunomodulatory molecules,including granulocyte colony-stimulating factor,peripheral blood mononuclear cells,autologous platelet-rich plasma,and intralipid and intravenous immunoglobulins,may improve implantation rate by modulating endometrial immune *** extracts of the Chinese herbs Paeonia lactiflora and Perilla frutescens increased the expression of leukemia inhibitory factors in endometrial epithelial ***,the use of the commercially available Library of Pharmacologically Active Compounds with a high-throughput screening method has provided an approach to screen for compounds that may potentially enhance or suppress embryo *** these biomedical findings translate into clinical effects that enhance or suppress embryo implantation requires further investigation.

The developmental origins of health and diseases(DOHaD)is a concept stating that adverse intrauterine environments contribute to the health risks of *** the theory emerged more than 30 years ago,many epidemiological and animal studies have confirmed that in utero exposure to environmental insults,including hyperglycemia and chemicals,increased the risk of developing noncommunicable diseases(NCDs).These NCDs include metabolic syndrome,type 2 diabetes,and complications such as diabetic *** the effects of different environmental insults on early embryo development would aid in understanding the underlying mechanisms by which these insults promote NCD *** stem cells(ESCs)have also been utilized by researchers to study the *** have pluripotent characteristics and can be differentiated into almost every cell lineage;therefore,they are excellent in vitro models for studying early developmental *** importantly,human ESCs(hESCs)are the best alternative to human embryos for research because of ethical *** this review,we will discuss different maternal conditions associated with DOHaD,focusing on the complications of maternal ***,we will review the differentiation protocols developed to generate different cell lineages from ***,we will review how hESCs are utilized as a model for research into the *** effects of environmental insults on hESC differentiation and the possible involvement of epigenetic regulation will be discussed.

Objective To report the experience in sperm extraction from testicular biopsies (TESE) performed from March 1996 to July 1998 in men with non obstructive azoospermia (NOA) Methods Comparisons of age, volume of both testes, serum FSH and testosterone in men, and histology of testicular samples in the first cycles between cycles with spermatozoa found and those without spermatozoa found were performed Comparisons of fertilization, cleavage and pregnancy rates between cycles with spermatozoa injected and those with spermatids injected were performed Results Spermatozoa were found in only 12 out of 26 first TESE cycles (46 2%) and other cycles had spermatids (round cells) only Age of men, history of mumps orchitis/oligozoospermia, volume of both testes and serum FSH/testosterone levels in men were not significantly different between cycles with and without spermatozoa The fertilization rate was significantly higher in cycles with spermatozoa injected than those with round cell injections (63 3% vs 23 2%, P <0 0001, Chi squared test) The pregnancy rate was 14 3% per cycle when spermatozoa were injected Conclusions TESE followed by Intracytoplasmic sperm injection (ICSI) is an effective treatment in patients with NOA Less than half of the patients undergoing TESE had spermatozoa recovered Age of men, volume of both testes and serum FSH/ testosterone levels in men were not useful in predicting successful recovery Compared to using ejaculated and epididymal spermatozoa, fertilization and pregnancy rates were achieved when testicular spermatozoa were used for ICSI

Glycodelin is a glycoprotein with four published isoforms, namely glycodelin-A (amniotic fluid isoform), glycodelin-S (seminal plasma isoform) glycodelin-F (follicular fluid isoform) and glycodelin-C (cumulus isoform) (Seppala et al., 2002;Chiu et al., 2003b, 2007a). They have the same protein structure but have different glycosylation (Seppala et al., 2002;yeung et al., 2006).

目的探讨人输卵管上皮细胞分泌的胚胎营养因子(ETF)对着床前鼠胚胎基因表达的影响。方法采用液相色谱技术从体外培养的人输卵管上皮细胞系 OE-E6/E7的培养液中分离出ETF,加入培养液后培养鼠胚胎。采用差异显示 RT-PCR 技术比较 ETF 培养的鼠胚胎(培养组)与普通培养液(未加入 ETF)培养的鼠胚胎(对照组)间的基因表达差异。将差异表达片段分离后,进行扩增、克隆、测序和分析。结果培养组胚胎与对照组胚胎共分离克隆了8个差异表达基因,即:RABgeranylgeranyl 转移酶、DEAD-box 蛋白50、RNA 结合结构域蛋白27、二磷酸甲羟戊酸盐脱羧酶、丝氨酸/精氨酸重复基质1、肿瘤差异表达基因2(TDE2)、T 细胞受体α/δ、胚胎发育因子1。结论人输卵管上皮细胞分泌的 ETF 对着床前鼠胚胎的基因表达有影响。
