Inheritance of marsh spot disease resistance in cranberry common bean(Phaseolus vulgaris L.)

Common bean(Phaseolus vulgaris) is an annual legume crop that is grown worldwide for its edible dry seeds and tender pods. Marsh spot(MS) of the seeds is a physio-genic stress disease affecting seed quality in beans. Studies have suggested that this disease involves a nutritional disorder caused by manganese deficiency, but the inheritance of resistance to this disease has not been reported. A biparental genetic population composed of 138 recombinant inbred lines(RILs) was developed from a cross between an MS resistant cultivar ‘Cran09’ and an MS susceptible cultivar ‘Messina’. The 138 RILs and their two parents were evaluated for MS resistance during five consecutive years from 2015 to 2019 in sandy and heavy clay soils in Morden, Manitoba, Canada. The MS incidence(MSI) and the MS resistance index(MSRI) representing disease severity were shown to be both highly correlated heritable traits that had high broad-sense heritability values(h;) of 86.5% and 83.2%, respectively. No significant differences for MSI and MSRI were observed between the two soil types in all five-(MSI) or four-year(MSRI) data collection, but significant correlations among years were observed despite MS resistance was moderately affected by year. The MSIs and MSRIs displayed a right-skewed distribution, indicating a mixed genetic model involving a few major genes and polygenes. Using the joint segregation analysis method, the same four major genes with additive-epistasis effects showed the best fit for both traits, explaining 84.4% and85.3% of the phenotypic variance for MSI and MSRI, respectively. For both traits, the M1, M2, M3 and m4 acted as the favorable(resistant) alleles for the four genes where M and m represent two alleles of each gene. however, due to epistatic effects, only the individuals of the M1 M2 M3 M4 haplotype appeared to be highly resistant, whereas those of the m1 m2 m3 M4 haplotype were the most susceptible. Cu(II) Benzoylpyridine Thiosemicarbazone Complexes: Inhibition of human Topoisomerase IIα and Activity against Breast Cancer Cells

The focus of this research is on the study of a series of copper (II) benzoylpyridine thiosemicarbazone complexes. Of the six benzoylpyridine thiosemicarbazone ligands used in this study, two are reported for the first time;2-benzoylpyridine tert-butyl thiosemicarbazone (BZP-tBTSC), and 2-benzoylpyridine benzyl thiosemicarbazone (BZP-BzTSC). Once characterized by NMR, melting point, and MS, these mono-anionic tridentate ligands were then reacted with Cu2+ to form the new square planar metal complexes [Cu(BZP-tBTSC)Cl] and [Cu(BZP-BzTSC)Cl]. All of the copper complexes display marked inhibition of human topoisomerase IIα. The [Cu(BZP-tBTSC)Cl] complex shows marked activity against human breast cancer cell lines.

Therapeutic hypothermia is the one of the most effective measures against brain injury. Astrocytes are the brain’s most abundant cell type so it is essential to understand their stress response. One of the main mechanism by which hypothermia attenuates oedema formation, is preserving the brain’s water homeostasis. This water homeostasis is tightly regulated by a group of membrane proteins called aquaporins（AQPs）. In this study, we investigated the alterations in the expressional profile of all the cerebral AQPs with a particular interest of AQP4 membrane expression and calmodulin level in astrocytes cultured under hypoxic conditions for 6 h at mild hypothermia of 32 ℃. human aquaporin 4（AQP4） is the primary water channel protein in brain astrocytes. hypothermia is known to cause astrocyte swelling in culture, but the precise role of AQP4 in this swelling is unknown. Primary human cortical astrocytes were cultured under hypothermic（32 ℃） or normothermic（37 ℃） conditions. AQP4 transcript, total protein and surface localized protein were quantified using RT-q PCR, sandwich ELISA with whole cell lysates, or cell-surface biotinylation followed by ELISA analysis of the surface-localized protein, respectively. 4 h mild hypothermic incubation increased the surface localization of AQP4 in human astrocytes to 256 ± 2% of normothermic controls, despite no change in total protein expression. hypothermia-mediated increase in the AQP4 surface abundance in human astrocytes was blocked using a calmodulin antagonist（trifluoperazine;TFP） or the TRPV4 inhibitor hC-067047. hypothermia leads to an increase in surface localization of AQP4 in human astrocytes through a mechanism likely dependent on the TRPV4 calcium channel and calmodulin activation. hypothermia has a complex effect and there is no single factor that could explain its neuroprotection effect. Aphanomyces euteiches： A Threat to Canadian Field Pea Production

Field pea （Pisum sativum var. arvense L.） is an important legume crop around the world. It produces grains with high protein content and can improve the amount of available nitrogen in the soil. Aphanomyces root rot （ARR）, caused by the soil-borne oomycete Aphanomyces euteiches Drechs. （A. euteiches）, is a major threat to pea production in many pea-growing regions including Canada; it can cause severe root damage, wilting, and considerable yield losses under wet soil conditions. Traditional disease management strategies, such as crop rotations and seed treatments, cannot fully prevent ARR under conditions conducive for the disease, due to the longevity of the pathogen oospores, which can infect field pea plants at any growth stage. The development of pea cultivars with partial resistance or tolerance to ARR may be a promising approach to analyze the variability and physiologic specialization ofA. euteiches in field pea and to improve the management of this disease. As such, the detection of quantitative trait loci （QTL） for resistance is essential to field pea-breeding programs. In this paper, the pathogenic characteristics of A. euteiches are reviewed along with various ARR management strategies and the QTL associated with partial resistance to ARR.

禾谷孢囊线虫(cereal cyst nematode,CCN)是一类重要的土传小麦病原线虫,危害我国小麦的主要是燕麦孢囊线虫(heterodera avenae)和菲利普孢囊线虫(***)。我国对这些病原线虫的抗性资源十分缺乏,寻找新抗源是当前抗性育种的重要工作。本研究通过3年的田间病圃和温室接种鉴定,发现加拿大的硬粒小麦品种Waskana和Waskowa对***(河南许昌群体,hfc-1致病型)和***(河南荥阳群体,ha43致病型)都表现很强的抗性,单株孢囊数显著少于感病的普通小麦品种矮抗58、石4185和温麦19。显微观察可见,虽然2种线虫的幼虫都能够侵入Waskana和Waskowa的根组织内,但是根内的线虫数量显著少于感病对照普通小麦品种,最终在根系上形成的可见孢囊数量也较少。Waskana和Waskowa对2种病原线虫的抗性为我国抗CCN小麦品种选育提供了有较高利用价值的新抗源。根据南澳大利亚研究所的土传病害检测服务系统对土壤中病原线虫的分子检测结果,抗CCN品种Waskana和Waskowa根际土壤中的线虫虫卵量低于感病小麦品种,因此种植后可能降低土壤中禾谷孢囊线虫危害的风险。
