Fabrication of all-transparent polymer-based and encapsulated nanofluidic devices using nano-indentation lithography

In this paper,we describe a novel and simple process for the fabrication of all-transparent and encapsulated polymeric nanofluidic devices using nano-indentation ***,a nanomechanical probe is used to‘scratch’nanoscale channels on polymethylmethacrylate(PMMA)substrates with sufficiently high ***,polydimethylsiloxane(PDMS)is used twice to duplicate the nanochannels onto PDMS substrates from the‘nano-scratched’PMMA substrates.A number of experiments are conducted to explore the relationships between the nano-indentation parameters and the nanochannel dimensions and to control the aspect ratio of the fabricated *** addition,traditional photolithography combined with soft lithography is employed to fabricate microchannels on another PDMS‘cap’*** manually aligning the substrates,all uncovered channels on two separate PDMS substrates are bonded to achieve a sealed and transparent nanofluidic device,which makes the dimensional transition from microscale to nanoscale *** smallest dimensions of the achievable nanochannels that we have demonstrated thus far are of~20 nm depth and~800 nm width,with lengths extendable beyond 100μ*** flow experiments are performed to verify the reliability of the *** types of colloidal solution are used to visualize the fluid flow through the nanochannels,that is,ethanol is mixed with gold colloid or fluorescent dye(fluorescein isothiocyanate),and the flow rate and filling time of liquid in the nanochannels are estimated based on time-lapsed image *** simplicity of the fabrication process,bio-compatibility of the polymer substrates,and optical transparency of the nanochannels for flow visualization are key characteristics of this approach that will be very useful for nanofluidic and biomolecular research applications in the future.

Inhibition of mycobacterial membrane protein large 3(MmpL3)thereby affecting the mycolic acid biosynthetic pathway has been proven to be an effective strategy for developing antitubercular *** on the X-ray crystal structure of MmpL3 inhibitor complexes,a series of novel 1,2,4-triazole derivatives were designed,synthesized and evaluated antitubercular activity against Mtb strain *** structure–activity relationship exploration resulted in the identification of compounds 21 and 28,which possess potent antitubercular activity against Mtb strain H37Rv[minimum inhibitory concentration(MIc)=0.03–0.13μg/mL]and the clinical isolates of multidrug resistance(MDR)and extensive drug resistance(XDR)tuberculosis(MIc=0.06–1.0μg/mL).Moreover,compounds 21 and 28 showed neglectable cytotoxicity(Ic_(50)≥32μg/mL)to the mammalian Vero cells and favorable physicochemical and pharmacokinetic properties according to the in silico absorption,distribution,metabolism and excretion(ADME)***,the potential target of representative 1,2,4-triazole 28 was identified to be MmpL3 using a microscale thermophoresis(MST)assay.

Isolating higher yielding and more stable rice genotypes in stress environments: fine-tuning a selection method using production and resilience score indices

In Asia, the rice crop sustains millions of people. However, growing demand for this crop needs to be met while simultaneously reducing its water consumption to cope with the effects of climate change. Lowland cropping systems are the most common and productive but have particularly high water requirements. High-yielding rice genotypes adapted to drier environments(such as rainfed or aerobic rice ecosystems) are needed to increase the water use efficiency of cropping. Identifying these genotypes requires fast and more accurate selection methods. It is hypothesized that applying a new quantitative selection method(the score index selection method), can usefully compare rice yield responses over different years and stress intensities to select genotypes more rapidly and efficiently. Applying the score index to previously published rice yield data for 39 genotypes grown in no-stress and two stress environments, identified three genotypes(ARB 8, IR55419-04 and ARB 7) with higher and stable yield under moderate to severe stress conditions. These genotypes are postulated to be better adapted to stress environment such as upland and aerobic environments. Importantly, the score index selection method offers improved precision than the conventional breeding selection method in identifying genotypes that are well-suited to a range of stress levels within the target environment.

cDc42 controls intestinal epithelial(IEc)stem cell(IESc)*** aberrant cDc42 initiates intestinal inflammation or neoplasia is *** utilized models of inflam-matory bowel diseases(IBD),colorectal cancer,aging,and IESc injury to determine the loss of intestinal cdc42 upon inflammation and *** specimens were collected to determine the levels of cDc42 in IBD or colorectal ***42 floxed mice were crossed with Villin-cre,Villin-creERT2 and/or Lgr5-eGFP-IRES-creERT2,or Bmi1-creERT2 mice to generate cdc42 deficient ***,colitis,aging,and intestinal organoid were used to evaluate cDc42 upon mucosal inflammation,IESc/progenitor regenerative capacity,and IEc *** studies revealed that increased cDc42 in colorectal cancer correlated with lower survival;in contrast,lower levels of cDc42 were found in the inflamed IBD *** cdc42 depletion significantly reduced Lgr5+IEscs,increased progenitors'hyperplasia,and induced mucosal inflammation,which led to crypt *** cdc42 depletion markedly enhanced irra-diation-or chemical-induced *** or inhibition of cdc42 reduced colonic Lgr5+IESc *** conclusion,depletion of cdc42 reduces the IESc regeneration and IEc repair,leading to prolonged mucosal *** monogenic loss of cdc42 in-duces mucosal inflammation,which could result in intestinal neoplasia in the context of aging.

Developing a new agenda for increased food and climate security

In many countries, political and environmental pressures are currently combining to generate a perfect storm of circumstances that is reducing food availability, increasing food costs and thereby reducing the availability of food to many. The UK is currently considering new national food and land management policies, and attention is also being given to legislation to address diet-related health issues. Many now argue for a revolution in UK farming practices to reduce their impact on the natural environment. The UK is not alone in facing these and other challenges. Both the contribution of agriculture to greenhouse gas(GHG) emissions and the effects of climate change on food production are issues receiving worldwide *** agricultural practices can result in greater c capture, reduced GHG emissions, enhanced soil quality and enhanced biodiversity. However, it is questioned if such farming systems will be productive enough to feed a growing population with the food required for social and health benefits. To fully exploit the impact of new plant science in farmer fields, it is imperative to effectively link science to farming practices and conduct a broader conversation around the food revolution with social scientists and with the general public.

Macroalgae are multicellular,aquatic autotrophs that play vital roles in global climate maintenance and have diverse applications in biotechnology and eco-engineering,which are directly linked to their multicellularity ***,their genomic diversity and the evolutionary mechanisms underlying multicellularity in these organisms remain *** this study,we sequenced 110 macroalgal genomes from diverse climates and phyla,and identified key genomic features that distinguish them from their microalgal *** for cell adhesion,extracellular matrix formation,cell polarity,transport,and cell differentiation distinguish macroalgae from microalgae across all three major phyla,constituting conserved and unique gene sets supporting multicellular *** genes show phylum-and climate-specific expansions that may facilitate niche ***,our study reveals genetic determinants of convergent and divergent evolutionary trajectories that have shaped morphological diversity in macroalgae and provides genome-wide frameworks to understand photosynthetic multicellular evolution in aquatic environments.
