UV-induced senescence of human dermal fibroblasts restrained by low-stiffness matrix by inhibiting NF-κB activation

As a hallmark of skin aging,senescent human dermal fibroblasts(HDFs)are known to lose the ability to ***,they can still interact with their cellular environment and the surrounding *** the skin ages,the progressive slowing down of HDFs function decreases the skin’s structural integrity,which is more serious than if there is the dermal collagen matrix *** leads to matters of the unbalanced barrier under the skin,skin fragility,inadequate wound healing,as well as other cosmetic *** is also well documented that skin aging comes with significant stiffness ***,understanding the interactions between HDFs and the surrounding microenvironments during senescence may provide insights into skin *** we aim to inves-tigate matrix stiffness’effect on HDF senescence and elucidate possible mechanisms that make HDFs *** our experiments,HDFs were cultivated on Polydimethylsiloxane(PDMS)with various stiffnesses and exposed to UV light to trigger *** show that HDFs are significantly affected by senescence when cultured on a matrix with ***,the cells are not significantly affected when cultured on a low stiffness *** following characterization revealed cells cultured on stiffsubstrates under UV exposure had stimu-lated the nucleus factor kappa-B(NF-κB)*** contrast,cells on a matrix of softness only displayed low activation of NF-κ***-κB activity suppression with ammonium pyrrolidine dithiocarbamate(PDTC)decreases UV-induced HDFs senescence on *** together,we demonstrated that soft matrix defends HDFs against ultraviolet-induced senescence by inhibiting the activation of NF-κB.

变频驱动游梁有杆泵系统仿真与频率优化研究

针对电机匀速驱动游梁抽油机运行过程中周期性交变载荷变化大、系统能耗高及故障多的问题,分析了电机变频驱动对抽油机运动规律、悬点载荷、变速箱扭矩及电机功率的影响,建立了变频控制抽油系统动态能耗的仿真模型。以电机输出功率均方根最小、输出功率峰值最小和负发电最小为目标函数,综合考虑平均频率不变、上下冲程时间分配、频率范围等约束条件,建立了周期运行频率优化设计的数学模型,并采用粒子群优化算法求解周期频率分布函数的傅里叶系数,即周期运行频率分布。实例分析表明:优化实时频率可以显著改善抽油机的动力性能,悬点载荷、曲柄扭矩和输出功率的峰值均有所降低;电机平均输出功率降低16.2%,负发电减少14.77%,节能效果显著。所得结果可为变频驱动游梁有杆泵系统的设计及应用提供参考。

Genetic and antigenic characterization of recombinant nucleocapsid proteins derived from canine coronavirus and canine respiratory coronavirus in China

To characterize the antigenicity of nucleocapsid proteins(NP) derived from canine coronavirus(CCo V) and canine respiratory coronavirus(CRCo V) in China, the N genes of CCo V(CCo V-BJ70) and CRCo V(CRCo V-BJ202) were cloned from swabs obtained from diseased pet dogs in Beijing and then sequenced. The recombinant NPs(r NPs) were expressed in Escherichia coli and purified by nickel-affinity column and size exclusion chromatography. Sequencing data indicated that the N genes of CCo V-BJ70 and CRCo V-BJ202 belonging to two distinctly different groups were relatively conserved within each subgroup. Sodium dodecyl sulfate polyacrylamide gel electrophoresis(SDS-PAGE) results showed that r NPs of CCo V and CRCo V were expressed efficiently and isolated with a final purity of over 95%. Western blot analysis revealed the r NP from CRCo V could cross-react with mice antisera against human coronavirus(HCo V-229 E, NL63, OC43, HKU1), while r NP of CCo V had cross-reactivity with only anti-sera against viruses belonging to the same group(HCo V-229 E and NL63). In summary, CCo V and CRCo V r NPs were successfully expressed in E. coli and showed antigenic cross-reactivity with antisera raised against human coronaviruses. These findings indicate that further serologic studies on coronavirus infections at the animal-human interface are needed.

Strategies of behavior,energetic and thermogenesis of striped hamsters in response to food deprivation

The life history of many animals includes periods of food *** behavioral strategies are involved in small mammals in response to food shortage:an increase in activity behavior,representing the increased foraging or migratory behavior,and a decrease in activity level,serving as a mechanism for conserving ***,it is uncertain whether animals adopt both strategies in response to food shortage,and whether hormone and neuroendocrine mechanisms are involved in both *** the present study,changes in behavior and metabolic rate were examined in food-deprived striped hamsters(Cricetulus barabensis).The effects of leptin supplement on activity behavior,metabolic rate and hypothalamic neuropeptide gene expression were also *** behavior of food-deprived hamsters significantly changed with photoperiod phases:with increasing activity during the dark phase compared to those fed ad libitum,whereas decreasing activity and simultaneously increasing resting behavior during the light *** metabolic rate,body mass,and masses of fat depots and digestive tracts significantly decreased in food-deprived hamsters compared with ad libitum *** supplement tended to attenuate the increased activity in the dark *** expression of hypothalamic neuropeptide Y(NPY)was significantly upregulated in food-deprived hamsters,while was significantly attenuated by exogenous *** findings suggest that both behavior strategies are important behavioral adjustments in free-living animals to cope with food *** and hypothalamic NPY gene expression may be involved in the adjustments of physiology and behavior in animals demonstrating a hyperactivity strategy in response to food shortage.

Stem cell niche-inspired microcarriers with ADSCs encapsulation for diabetic wound treatment

Stem cell therapies have made great progress in the treatment of diabetic wounds during recent decades,while their short in vivo residence,alloimmune reactions,undesired behaviors,and dramatic losses of cell functions still hinder the translation of them into ***,inspired by the natural components of stem cell niches,we presented novel microfluidic hydrogel microcarriers with extracellular matrix(ECM)-like composition and adipose-derived stem cells(ADSCs)encapsulation for diabetic wound *** the hydrogel was synthesized by conjugating hyaluronic acid methacryloyl(HAMA)onto the Fibronectin(FN)molecule chain(FN-HAMA),the laden ADSCs in the microcarriers showed improved bioactivities and pro-regenerative *** on these features,we have demonstrated that these ADSCs microcarriers exhibited significant promotion of neovascularization,follicular rejuvenation,and collagen deposition in a mouse diabetic wound *** results indicated that the stem cell niche-inspired FN-HAMA microcarriers with ADSCs encapsulation have great clinical potential for diabetic wound treatment.

Developing a biocompatible and multifunctional adhesive hydrogel with injectability and self-healing ability for promoting wound healing is highly anticipated in various clinical *** this paper,we present a novel natural biopolymer-derived hydrogel based on the aldehyde-modified oxidized guar gum(OGG)and the carboxymethyl chitosan(CMCS)for efficiently improving wound healing with the encapsulation of vascular endothelial growth factor(VEGF).As the hydrogels are synthesized via the dynamically reversible Schiff base linkages,it is imparted with excellent self-healing ability and good shear thinning behavior,which make the hydrogel be easily and conveniently injected through a ***,the physiochemical properties,including porous structure,mechanical strength and swelling ratio of the hydrogel can be well controlled by regulating the concentrations of the ***,the hydrogel can attain strong adhesion to the tissues at physiological temperature based on the Schiff base between the aldehyde group on the hydrogel and the amino group on the *** on these features,we have demonstrated that the VEGF encapsulated hydrogel can adhere tightly to the defect tissue and improve wound repair in the rat model of defected skin by promoting cell proliferation,angiogenesis,and collagen *** results indicate that the multifunctional hydrogel is with great scientific significance and broad clinical application prospects.

Impact of hepatectomy and postoperative adjuvant transarterial chemoembolization on serum tumor markers and prognosis in intermediate-stage hepatocellular carcinoma

BACKGROUND Primary hepatocellular carcinoma(HCC)is a common malignant tumour,and its early symptoms are often not obvious,resulting in many patients experiencing middle-to late-stage disease at the time of *** optimal time for surgery is often missed for these patients,and those who do undergo surgery have unsatisfactory long-term outcomes and a high recurrence rate within five ***,postoperative follow-up treatments,such as transhepatic arterial chemoembolization(TACE),have become critical to improving survival and reducing recurrence *** To validate the prophylactic role of TACE after hepatic resection and to assess its impact on patient *** This study investigated the efficacy of TACE in patients with intermediate-stage HCC after *** the post-treatment results of the observation group and the control group were compared,it was found that the inclusion of TACE significantly improved the clinical efficacy,reduced the levels of tumour markers and did not aggravate the damage to liver ***,this may be an effective and comprehensive treatment strategy for patients with intermediate-stage HCC that helps to improve their quality of life and survival *** When the baseline data were analysed,no statistical differences were found between the two groups in terms of gender,age,hepatitis B virus,cirrhosis,Child-Pugh grading,number of tumours,maximum tumour diameter and degree of tumour *** assessment of clinical efficacy showed that the post-treatment overall remission rate of the observation group was significantly higher than that of the control *** terms of changes in tumour markers,the alpha-fetoprotein and carcinoembryonic antigen levels in the patients in the observation group decreased more significantly after treatment compared with those in the control *** post-treatment changes in liver function indicators were analysed,no statistical differences were found in the tot
