RNA interference(RNAi)is a powerful tool that post-transcriptionally silences target genes in eukaryotic ***,silencing efficacy varies greatly among different insect ***,we met with little success when attempting to knock down genes in the mirid bug Apolygus lucorum via dsRNA *** disappearance of double-stranded RNA(dsRNA)could be a potential factor that restricts RNAi ***,we found that dsRNA can be degraded in midgut fluids,and a dsRNase of ***(AldsRNase)was identified and *** alignment indicated that its 6 key amino acid residues and the Mg2+-binding site were similar to those of other insects’*** signal peptide and endonuclease non-specific domain shared high sequence identity with the brown-winged green stinkbug Plautia stali *** showed high salivary gland and midgut expression and was continuously expressed through the whole life cycle,with peaks at the 4th instar ecdysis in the whole *** purified AldsRNase protein obtained by heterologously expressed can rapidly degrade *** comparing the substrate specificity of AldsRNase,3 specific substrates(dsRNA,small interfering RNA,and dsDNA)were all degraded,and the most efficient degradation is ***,immunofluorescence revealed that AldsRNase was expressed in the cytoplasm of midgut *** cloning and functional study of AldsRNase,the enzyme activity and substrate specificity of the recombinant protein,as well as the subcellular localization of nuclease,the reason for the disappearance of dsRNA was explained,which was useful in improving RNAi efficiency in *** and related species.

Anti-inflammatory effects and mechanism of triterpene alcohols from camellia oil

Camellia oil， which contains a high content of triterpene alcohol， is known for a series of bioactivities including anti-inflammation. Amyrins are recognized as high bioactivity of anti-inflammation. However， no comparative study on triterpene alcohols from camellia oil. In this study， four high content triterpene alcohols from camellia oil， namely β-amyrin， ψ-taraxasterol， parkeol， and butyrospermol were evaluated through lipopolysaccharide induced RAW 264.7 cell inflammation. The results showed that butyrospermol exhibited the highest anti-inflammatory activity， surpassing that of β-amyrin. Characterization of signaling pathways showed that butyrospermol inhibited TLR4， NF-κB and MAPK pathways， suppressing the transcription of Tlr4， expression of p65， IκBα， ERK， JNK， and p38， and the phosphorylation of p65， IκBα， ERK， and p38. The anti-inflammatory effect of butyrospermol was further validated by phorbol 12-myristate 13-acetate induced mouse ear edema. The results in mouse showed that butyrospermol could inhibit the increase of TNF-α， IL-1β， p-JNK， p-p38， p-IκBα， and their corresponding mRNA levels. Our study provides new perspective on the anti-inflammatory role of different triterpene alcohols and explaining the bioactivity of camellia oil.

Cell-free RNA(cf RNA) allows assessment of health, status, and phenotype of a variety of human organs and is a potential biomarker to non-invasively diagnose numerous diseases. Nevertheless, there is a lack of highly efficient and bias-free cf RNA isolation technologies due to the low abundance and instability of cf ***, we developed a reproducible and high-efficiency isolation technology for different types of cell-free nucleic acids(containing cf RNA and viral RNA) in serum/plasma based on the inclusion of nucleic acids by metal-organic framework(MOF) materials, which greatly improved the isolation efficiency and was able to preserve RNA integrity compared with the most widely used research kit method. Importantly, the quality of cf RNA extracted by the MOF method is about 10-fold that of the kit method, and the MOF method isolates more than three times as many different RNA types as the kit method. The whole transcriptome mapping characteristics of cf RNA in serum from patients with liver cancer was described and a cf RNA signature with six cf RNAs was identified to diagnose liver cancer with high diagnostic efficiency(area under curve = 0.905 in the independent validation cohort) using this MOF method. Thus, this new MOF isolation technique will advance the field of liquid biopsy, with the potential to diagnose liver cancer.

Sesame is one of the eight major allergens that cause food allergies. Study of the epitopes of sesame allergens is important for understanding their sensitization mechanisms. Currently， less information is available on the epitope studies of sesame allergens. In this study， we analyzed the molecular characteristics， structure and homology of Ses i 3， one of the important sesame allergens. We predicted the B-cell linear epitopes of Ses i 3 using bioinformatics tools and characterized them by slot blot immuno-microarrays technology. Eight peptides as B-cell linear epitopes of Ses i 3 were identified， in addition， key amino acids in these epitopes were predicted and leucine 422 was identified as a key amino acid. The present work will contribute to further understanding of the sesame allergen and provide some help in the prevention and treatment of sesame allergy.

Methanotrophic bacteria are currently used industrially for the bioconversion of methane-rich natural gas and anaerobic digestion-derived biogas to valuable *** bacteria may also serve to mitigate the negative effects of climate change by capturing atmospheric greenhouse *** genetic tools have previously been developed for genetic and metabolic engineering of ***,the available tools for use in methanotrophs are significantly underdeveloped compared to many other industrially relevant bacteria,which hinders genetic and metabolic engineering of these *** such,expansion of the methanotroph genetic toolbox is needed to further our understanding of methanotrophy and develop biotechnologies that leverage these unique microbes for mitigation and conversion of methane to valuable ***,we determined the copy number of three broad-host-range plasmids in Methylococcus capsulatus Bath and Methylosinus trichosporium OB3b,representing phylogenetically diverse Gammaproteobacterial and Alphaproteobacterial methanotrophs,***,we show that the commonly used synthetic Anderson series promoters are functional and exhibit similar relative activity in *** and *** OB3b,but the synthetic series had limited ***,we mutagenized the native *** particulate methane monooxygenase promoter and identified variants with activity that expand the activity range of synthetic,constitutive promoters functional not only in ***,but also in Escherichia ***,the tools developed here advance the methanotroph genetic engineering toolbox and represent additional synthetic genetic parts that may have broad applicability in Pseudomonadota bacteria.

Silkworm pupa peptide ameliorates dextrose sodium sulfate-induced colitis by enhancing gut barriers and modifying gut flora

The anti-inflammatory properties of silkworm pupa peptide (SPP) have been recognized for their potential benefits in colitis treatment. However， the underlying mechanism of SPP in colitis animal models remains unclear. The objective of our work was to investigate the improving effect and the mechanism of action of SPP on a mouse model of dextrose sodium sulfate (DSS)-induced colitis. The findings suggested that SPP effectively eased ulcerative colitis (UC) symptoms in mice by decreasing disease activity index， ameliorating histopathological injury， and decreasing serum and colonic markers of inflammation. In addition， our research findings demonstrated that SPP restored intestinal barrier function by increasing the production of tight junction proteins such as zonula occludens-1 (ZO-1)， claudin-1， claudin-3， occludin， and adherens junction protein E-cadherin1. Further， SPP supplementation enhanced the concentration of short-chain fatty acids and positively altered the makeup of the gut bacteria in the mice’s gut. These findings underscore SPP’s ability to slow the progression of colitis and point to its possible use as a functional component in dietary supplements for the prevention of early-stage colitis.

Visible light communication(VLC)based on laser diodes demonstrates great potential for high data rate maritime,terrestrial,and aerial wireless data ***,we design and fabricate high-speed blue laser diodes(LDs)grown on c-plane gallium nitride(GaN)*** was achieved through active region design and miniaturization toward a narrow ridge waveguide,short cavity length,and single longitudinal mode Fabry–Perot laser *** fabricated mini-LD has a low threshold current of 31 mA and slope efficiency of 1.02 W/A.A record modulation bandwidth of 5.9 GHz(−3dB)was measured from the *** the developed mini-LD as a transmitter,the VLC link exhibits a high data transmission rate of 20.06 Gbps adopting the bit and power loading discrete multitone(DMT)modulation *** corresponding bit error rate is 0.003,satisfying the forward error correction *** demonstrated GaN-based mini-LD has significantly enhanced data transmission rates,paving the path for energy-efficient VLC systems and integrated photonics in the visible regime.

Arsenic， a known environmental carcinogen， disrupts intestinal homeostasis， posing a significant threat to human health. Mitigating its toxic effects is crucial， and this study explores the potential of swim bladder sulfated glycosaminoglycan (SBSG) in achieving this. Our previous in vitro studies have shown SBSG to ameliorate arsenic-induced damage in intestinal epithelial cells， but its in vivo effects remain elusive. The current investigation demonstrates that SBSG exhibits a beneficial prebiotic action in vivo， regulating gut microbiota， metabolites， and intestinal barrier function to counter arsenic's adverse effects. Specifically， SBSG regulates microbiota composition， suppressing pathogenic species like Alistipes and Candidatus＿Saccharimonas while promoting beneficial ones such as Ruminococcus and Akkermansia. In the colon， SBSG fermentation enhances the production of short-chain fatty acids (SCFAs)， leading to upregulation of GPR43， GPR109A， and Olfr78 receptors. Additionally， SBSG strengthens the intestinal barrier by increasing the expression of Claudin-1， Occludin， and ZO-1， and enhances mucin gene expression (MUC-1 and MUC-2) to address chemical barrier disruptions. Immunologically， SBSG modulates the RORγt/Foxp-3 pathway and the TLR4/MyD88/NF-κB signaling cascade， regulating the immune barrier. These findings suggest that SBSG could be a promising prebiotic candidate for maintaining intestinal health and may serve as a dietary supplement or adjunct in heavy metal detoxification therapies.

TheⅣ-Ⅵcompound GeTe is considered as a promising alternative to the toxic PbTe for high-efficiency mid-temperature thermoelectric ***,pristine GeTe suffers from a high concentration of Ge vacancies,resulting in an excessively high hole concentration(>1×10^(21)cm^(-3)),which greatly limits its thermoelectric *** address this issue,CuBiTe_(2)alloying is introduced to increase the formation energy of Ge vacancies in GeTe,thereby inhibiting the high carrier *** carrier scattering caused by the electronegativity difference between different elements is suppressed due to the similar electronegativity of Cu and Ge atoms.A relatively high hole mobility is obtained,which ultimately leads to a high power ***,by introducing Se as an alloying element at the anionic site in GeTe,dense point defects with mass/strainfield fluctuations are *** contributes to the strengthening of phonon scattering,thereby reducing the lattice thermal conductivity from 1.44 W·m^(-1)·K^(-1)for pristine GeTe to 0.28 W·m^(-1)·K^(-1)for Ge_(0.95)Cu_(0.05)Bi_(0.05)Te_(0.9)Se_(0.15)compound at 623 K.
