Objective Many studies have revealed the crucial roles of miRNA in multiple human cancers,including lung adenocarcinoma(LUAD).In this study,we sought to explore new miRNA-mRNA pairs that are associated with LUAD *** A novel miRNA-mRNA regulatory network associated with prognosis in LUAD was identified and validated using the bioinformatic tools including OncomiR database,StarBase,miRnet,GEPIA2,*** Twenty key miRNAs were compiled after the analysis of the expression and prognostic value in OncomiR and *** mRNAs of these key miRNAs were predicted in miRnet,and the resulting mRNAs were also analyzed for their prognostic values and expression patterns in GEPIA2 and UALCAN,*** expression correlation analysis was performed in ***,a new miRNA-mRNA network was built,of which each RNA pair showed negative expression correlation,opposite expression pattern,and prognostic ***-protein interaction network was under construction for the mRNAs,and 19 hub genes were *** analysis showed that“Cell Cycle,Mitotic”was the most significantly enriched ***,a miRNA-hub gene sub-network was *** selected and validated the regulatory relationship of some miRNA-hub pairs,including hsa-miR-1976/RFC2,hsa-let-7c-5p/RFC2,hsa-let-7c-5p/ESPL1,hsa-let-7c-5p/CDC25A,and hsa-miR-101-3p/***,over-expression of hsa-miR-1976 and hsa-let-7c-5p resulted in significant cell cycle *** Our results determined new prognosis-associated miRNA-mRNA pairs and might shed further light on the mechanism via which miRNA-mRNA network influences prognosis in LUAD.

目的:探讨在高氧诱导的新生大鼠支气管肺发育不良(BPD)中自噬和核苷酸结合寡聚化结构域样受体蛋白3(NLRP3)炎症小体之间的关系及其对肺发育结局的影响。方法:用85%O 2制造BPD新生大鼠模型,与常氧(21%O 2)新生大鼠对比,选择生后第3、7、14天的新生大鼠肺组织做病理切片,使用苏木精-伊红染色观察肺组织形态变化(RAC、MLI),Western blot测肺组织LC3、P62蛋白,确定自噬流变化。雷帕霉素组新生大鼠与高氧组在同一高氧环境中,于生后第2、4、6天予以腹腔注射雷帕霉素。高氧组与常氧组注射同等量生理盐水。选择生后第7天,Western blot测肺组织MTOR、LC3、P62、NLRP3、ASC、cleaved-caspase-1、cleaved-IL-1β,PCR测LC3、NLRP3、cleaved-caspase-1的mRNA,确定其自噬与炎症小体活性之间的关系。结果:肺组织HE染色示生后第7、14天RAC(P<0.05)常氧组均高于高氧组;MLI(P<0.05)高氧组均高于常氧组。Western blot示生后第3、7天LC3-Ⅱ/LC3-Ⅰ(P<0.01),P62(P<0.05)高氧组均高于常氧组。生后第7天肺组织HE染色示高氧组RAC低于常氧组和雷帕霉素组(P<0.05),MLI高于常氧组和雷帕霉素组(P<0.05),生后第7天Western blot示高氧组的MTOR、LC3-Ⅱ/LC3-Ⅰ、P62、NLRP3、ASC、cleaved-caspase-1、cleaved-IL-1β均高于正常组和雷帕霉素组(P<0.05)。qPCR示高氧组的LC3mRNA低于正常组和雷帕霉素组(P<0.05),而NLRP3、caspase-1的mRNA均高于正常组和雷帕霉素组(P<0.05)。结论:自噬流与炎症小体之间的相互作用是BPD发生发展的重要因素,通过增加自噬流可以减少炎症小体的活性,减少cleaved-IL-1β的生成,进而改善BPD,为进一步治疗BPD提供依据。

A novel organocatalytic cascade process initiated by 1,6-conjugated addition has been successfully developed. A range of pharmaceutically active 2-amino-4-aryl-4H-chromenes were readily obtained in high yields（88%-99%） and excellent enantiopurities（86%-99% ee）. Twenty-seven novel pyrrolidine-2,4-dione derivatives containing N-substituted phenylhydrazine moiety were synthesized. Their structures were confirmed by IH NMR, 13C NMR and MS. The half effective concentration （ECs0） values of the title compounds against the phytopathogenic fungi Rhizoctonia cerealis were evaluated. Com- pounds 61 and 6q displayed good bioactivity with EC50 values of 1.626 and 2.043 μg/mL, respectively. The 3D quantitative structure activity relationship（3D-QSAR） model of CoMFA was established with reliable cross-validated correlation coefficient q2 value of 0.585 and Noncross-validated correlation coefficient r2 value of 0.971. This model provided a tool for guiding further design and synthesis of novel pyrrolidine-2,4-dione derivatives with high fungicidal activity.

Study on the Therapeutic Effects and Mechanisms of Human Mesenchymal Stem Cell-Derived Exosomes Carrying NGF Gene in Treating Ischemic Stroke in Rats

Objective:To investigate the therapeutic effects and mechanisms of human mesenchymal stem cell-derived exosomes(hMSCs-Exo)carrying the NGF gene in treating ischemic stroke in rats,aiming to provide new insights and treatment methods for ischemic stroke ***:After successful construction of the cerebral ischemia model in 40 male SPF-grade SD rats aged 6-8 weeks,the model rats were randomly divided into 4 groups:Sham group,PBS group,hMSCs-Exo group,and NGF-hMSCs-Exo group,with 10 rats in each *** rat MCAO model was prepared using the classic filament method,and NGF-hMSCs-Exo were injected via the tail vein into the MCAO model *** expression of the NGF gene in brain ischemic tissues,neuronal regeneration,and rat neurological function recovery were observed using TTC staining,memory function evaluation,Western blot,qRT-PCR,and other ***:Compared with the Sham group,neurological deficits were significant in the PBS group(P<0.01).Compared with the PBS group,neurological scores improved in the hMSCs-Exo group and NGF-hMSCs-Exo group(P<0.05).Compared with the hMSCs-Exo group,the improvement in neurological deficits was more significant in the NGF-hMSCs-Exo group(P<0.05).The infarct area after NGF-hMSCs-Exo intervention was significantly reduced(P<0.05)compared with the Sham *** with the PBS group,relative expression levels of NGF mRNA and protein decreased,while Caspase-3 mRNA and protein expression significantly increased in the PBS group(P<0.01).Compared with the PBS group and hMSCs-Exo group,there were differences in NGF and Caspase-3 mRNA and protein expression in the NGF-hMSCs-Exo group rat brain tissues(P<0.05).Conclusion:Treatment with human mesenchymal stem cell-derived exosomes carrying the NGF gene improves cognitive function and exerts protective effects on SD rats while inhibiting apoptotic levels in cells.
