Sporopollenin in the pollen cell wall protects male gametophytes from stresses.Phenylpropanoid derivatives,including guaiacyl(G)lignin units,are known to be structural components of sporopollenin,but the exact composition of sporopollenin remains to be fully resolved.We analyzed the phenylpropanoid derivatives in sporopollenin from maize and Arabidopsis by thioacidolysis coupled with nuclear magnetic resonance(NMR)and gas chromatography–mass spectrometry(GC–MS).The NMR and GC–MS results confirmed the presence of p-hydroxyphenyl(H),G,and syringyl(S)lignin units in sporopollenin from maize and Arabidopsis.Strikingly,H units account for the majority of lignin monomers in sporopollenin from these species.We next performed a genome-wide association study to explore the genetic basis of maize sporopollenin composition and identified a vesicle-associated membrane protein(ZmVAMP726)that is strongly associated with lignin monomer composition of maize sporopollenin.Genetic manipulation of VAMP726 affected not only lignin monomer composition in sporopollenin but also pollen resistance to heat and UV radiation in maize and Arabidopsis,indicating that VAMP726 is functionally conserved in monocot and dicot plants.Our work provides new insight into the lignin monomers that serve as structural components of sporopollenin and characterizes VAMP726,which affects sporopollenin composition and stress resistance in pollen.

Two-dimensional(2D) rare-earth oxides(REOs) are a large family of materials with various intriguing applications and precise facet control is essential for investigating new properties in the 2D limit.However,a bottleneck remains with regard to obtaining their 2D single crystals with specific facets because of the intrinsic non-layered structure and disparate thermodynamic stability of different facets.Herein,for the first time,we achieve the synthesis of a wide variety of high-quality 2D REO single crystals with tailorable facets via designing a hard-soft-acid-base couple for controlling the 2D nucleation of the predetermined facets and adjusting the growth mode and direction of crystals.Also,the facet-related magnetic properties of 2D REO single crystals were revealed.Our approach provides a foundation for further exploring other facet-dependent properties and various applications of 2D REO,as well as inspiration for the precise growth of other non-layered 2D materials.

SARS-CoV-2 ORF10 suppresses the antiviral innate immune response by degrading MAVS through mitophagy

The global coronavirus disease 2019(COVID-19)pandemic caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)has caused severe morbidity and mortality in humans.It is urgent to understand the function of viral genes.However,the function of open reading frame 10(ORF10),which is uniquely expressed by SARS-CoV-2,remains unclear.In this study,we showed that overexpression of ORF10 markedly suppressed the expression of type I interferon(IFN-I)genes and IFN-stimulated genes.Then,mitochondrial antiviral signaling protein(MAVS)was identified as the target via which ORF10 suppresses the IFN-I signaling pathway,and MAVS was found to be degraded through the ORF10-induced autophagy pathway.Furthermore,overexpression of ORF10 promoted the accumulation of LC3 in mitochondria and induced mitophagy.Mechanistically,ORF10 was translocated to mitochondria by interacting with the mitophagy receptor Nip3-like protein X(NIX)and induced mitophagy through its interaction with both NIX and LC3B.Moreover,knockdown of NIX expression blocked mitophagy activation,MAVS degradation,and IFN-I signaling pathway inhibition by ORF10.Consistent with our observations,in the context of SARS-CoV-2 infection,ORF10 inhibited MAVS expression and facilitated viral replication.In brief,our results reveal a novel mechanism by which SARS-CoV-2 inhibits the innate immune response;that is,ORF10 induces mitophagy-mediated MAVS degradation by binding to NIX.

为了了解内蒙古地区腹泻犊牛中大肠杆菌的污染状况、耐药情况及蒲公英水煎液对多重耐药大肠杆菌耐药性的消除作用。本试验采用革兰染色、生化试验和PCR方法对采集到的107份犊牛源病料进行大肠杆菌的分离和鉴定,采用K-B法检测分离菌株对18种抗菌药物的敏感性,采用PCR方法进行8种耐药基因的检测,采用琼脂稀释法测定蒲公英水煎液对分离菌株的最小抑菌浓度(MIC),通过将菌株与蒲公英水煎液作用后进行耐药消除试验。结果显示,从107份病料中分离鉴定出44株犊牛源大肠杆菌,分离率为41%;大肠杆菌分离株对氨苄西林、头孢噻吩、头孢噻肟、环丙沙星和左氧氟沙星5种抗菌药物的耐药率均达到50%以上;β-内酰胺类耐药基因中bla CTX-M基因携带率为86%,喹诺酮类耐药基因中qnr(S)基因携带率为32%;蒲公英水煎液对筛选出的耐药菌株的MIC为200 mg/mL;经蒲公英水煎液处理后的大肠杆菌对抗菌药物的敏感性增强,而且更不容易出现耐药。结果表明,内蒙古地区腹泻犊牛中大肠杆菌的耐药情况严重,而蒲公英水煎液对大肠杆菌的多重耐药有一定的消除作用,可降低临床应用抗菌药物的耐药性,增加临床疗效,本试验结果为临床合理用药、有效预防和治疗犊牛腹泻提供了参考依据。

Zika virus(ZIKV),a positive-sense single-stranded RNA virus,causes congenital ZIKV syndrome in children and Guillain-Barre Syndrome(GBS)in adults.ZIKV expresses nonstructural protein 5(NS5),a large protein that is essential for viral replication.ZIKV NS5 confers the ability to evade interferon(IFN)signalling;however,the exact mechanism remains unclear.In this study,we employed affinity pull-down and liquid chromatography-tandem mass spectrometry(LC-MS/MS)analyses and found that splicing factor 3b subunit 3(SF3B3)is associated with the NS5-Flag pull-down complex through interaction with NS5.Functional assays showed that SF3B3 overexpression inhibited ZIKV replication by promoting IFN-stimulated gene(ISG)expression whereas silencing of SF3B3 inhibited expression of ISGs to promote ZIKV replication.GTP cyclohydrolase I(GCH1)is the first and ratelimiting enzyme in tetrahydrobiopterin(BH4)biosynthesis.NS5 upregulates the expression of GCH1 during ZIKV infection.And GCH1 marginally promoted ZIKV replication via the IFN pathway.Additionally,GCH1 expression is related to the regulation of SF3B3.Overexpression of the SF3B3 protein effectively reduced GCH1 protein levels,whereas SF3B3 knockdown increased its levels.These findings indicated that ZIKV NS5 binding protein SF3B3 contributed to the host immune response against ZIKV replication by modulating the expression of GCH1.

背景2016年全面推行家庭医生签约服务以来,居民基层首诊意愿逐步提升,但签约居民基层首诊效果评价尚需进一步探讨。目的了解签约居民的首诊性评价现状,探讨其影响因素并提出改善策略。方法采用多阶段分层随机抽样法在山东省三市抽取54所社区卫生服务机构/乡镇卫生院,共4000名签约居民。采用汉化版基本医疗质量评估量表(PCAT)的首诊性维度问卷进行调查,比较不同特征的签约居民首诊性维度得分,采用多元线性回归分析影响签约居民首诊性评价的因素。结果共回收有效问卷3859份,问卷有效回收率为96.48%。其中,乡镇卫生院签约居民为2086名(54.1%),社区卫生服务中心为1773名(45.9%);三个地区的首诊性维度总得分为3.49分,得分最高的条目是“您的签约机构周六、周日开门吗”(3.74分);得分最低的条目是“在非营业期间,如果您在晚上生病了,您的签约机构会有医生或护士出诊吗?”(3.05分)。多元线性回归分析结果显示,签约机构、个人月收入、文化程度、婚姻状况、职业、身体健康、是否确诊慢性疾病是签约居民首诊性维度得分的影响因素(P<0.05)。结论签约居民的首诊性评价较好,但还需加强家庭医生签约服务政策宣传,引导签约居民形成基层首诊的就医观念,通过明确非工作日出诊流程、提升基层医疗卫生机构服务供给能力、加强医疗卫生机构共享平台建设、发挥医疗保障制度杠杆作用等措施推动居民基层首诊,进而提升签约居民首诊性评价。

Genotype analysis of rotaviruses isolated from children during a phase III clinical trial with the hexavalent rotavirus vaccine in China

The oral hexavalent live human-bovine reassortant rotavirus vaccine(RV6)developed by Wuhan Institute of Biological Products Co.,Ltd(WIBP)has finished a randomized,placebo-controlled phase III clinical trial in four provinces of China in 2021.The trail demonstrated that RV6 has a high vaccine efficacy against the prevalent strains and is safe for use in infants.During the phase III clinical trial(2019–2021),200 rotavirus-positive fecal samples from children with RV gastroenteritis(RVGE)were further studied.Using reverse transcription-polymerase chain reaction and high-throughput sequencing,VP7 and VP4 sequences were obtained and their genetic characteristics,as well as the differences in antigenic epitopes of VP7,were analyzed in detail.Seven rotavirus genotypes were identified.The predominant rotavirus genotype was G9P[8](77.0%),followed by prevalent strains G8P[8](8.0%),G3P[8](3.5%),G3P[9](1.5%),G1P[8](1.0%),G2P[4](1.0%),and G4P[6](1.0%).The amino acid sequence identities of G1,G2,G3,G4,G8,and G9 genotypes of isolates compared to the vaccine strains were 98.8%,98.2%–99.7%,88.4%–99.4%,98.2%,94.2%–100%,and 93.9%–100%,respectively.Notably,the vaccine strains exhibited high similarity in amino acid sequence,with only minor differences in antigenic epitopes compared to the Chinese endemic strains.This supports the potential application of the vaccine in preventing diseases caused by rotaviruses.

Experimental-computational study of fibrous particle transport and deposition in a bifurcating lung model

Experiments carried out using a lung model with a single horizontal bifurcation under different steady inhalation conditions explored the orientation of depositing carbon fibers, and particle deposition frac- tions. The orientations of deposited fibers were obtained from micrographs. Specifically, the effects of the sedimentation parameter （γ）, fiber length, and flow rate on orientations were analyzed. Our results indicate that gravitational effect on deposition cannot be neglected for 0.0228 〈 γ 〈 0.247. The absolute orientation angle of depositing fibers decreased linearly with increasing y for values 0.0228 〈 γ 〈 0.15. Correspondence between Stokes numbers and y suggests these characteristics can be used to estimate fiber deposition in the lower airways. Computer simulations with sphere-equivalent diameter models for the fibers explored deposition efficiency vs. Stokes number. Using the volume-equivalent diameter model, our experimental data for the horizontal bifurcation were replicated. Results for particle deposition using a lung model with a vertical bifurcation indicate that body position also affects deposition.

The selection and design of an optimal solvent for extractive distillation require reliable vapour-liquid phase equilibrium data and knowledge of extraction mechanisms.Compared with time-consuming experiments,molecular simulation presents great potential in research on the properties of fluids.Therefore,in this work,Gibbs ensemble Monte Carlo was applied to successfully predict the vapour-liquid phase equilibrium data of binary and ternary systems containing benzene,thiophene and N,Ndimethylformamide(DMF) at P=101.3 kPa.The explicit hydrogen version of the transferable potentials for phase equilibria potential model was chosen for benzene and thiophene,whereas the OPLS potential model was selected for DMF.The predicted phase diagrams were compared with experimental data and the UNIQUAC thermodynamic model.A good agreement was obtained,which corroborated the validity of the potential models.In addition,the extraction mechanism was explored by radial distribution function(RDF) of the liquid-phase structure.The RDFs showed that thiophene and benzene shared a similar liquidphase structure because of the intermolecular interaction.The distinct difference between the RDFs of DMF/benzene and those of DMF/thiophene is that the oxygen atom of DMF is more associated with hydrogen atoms of thiophene than that of benzene,which may be responsible for the extraction effect of DMF.
